Computational studies have been developed to unravel the mechanism of action of the anti-malarial drugs and to give guidelines for the development of new derivatives with improved efficiency. In this study, dihydroorotate dehydrogenase (DHOD), a key enzyme in de novo pyrimidine biosynthesis and the major source of electrons for the mitochondrial electron transport chain of intraerythrocytic malaria parasites, was selected as a target for potential inhibitors. Beginning with the natural inhibitors found in crystallographed mols. representing *.pdb file, a general similarity search were done in PDB, NCI and PUBMED databases resemble probable anti-malarial compounds This made it possible to dock these ligand libraries with DHOD by FlexX 3.2, 2007 software and comparing the inter-mol. interactions and scores which are given to them by the software. The highest absolute value of interactions energies were considered be the best and proper ligands. These ligands were used for second 95% similarity search in PUBCHEM for proposed anti-malarial compound which have the same mechanism as A26. As an interesting point among 400 found mols. was presence of quinolines. This could propose a new mechanism for these important anti-malarial agents.

A forward chem. genetic approach was followed to discover new targets and lead compounds for Parkinson’s disease (PD) treatment. By anal. of the cell protection produced by some small mols., a di-Ph sulfide compound was revealed to be a new phosphodiesterase 7 (PDE7) inhibitor and identified as a new hit. This result allows us to confirm the utility of PDE7 inhibitors as a potential pharmacol. treatment of PD. On the basis of these data, a diverse family of di-Ph sulfides has been developed and pharmacol. evaluated in the present work. Moreover, to gain insight into the safety of PDE7 inhibitors for human chronic treatment, the authors evaluated the new compounds in a surrogate emesis model, showing nonemetic effects.

The SRN1 reaction has been used as a powerful tool for the synthesis of heterocycles, and only a few studies about photoinduced intramol. cyclization to generate a new C-O bond by a radical pathway have been reported. This work introduces two strategies for the synthesis of substituted dibenzofurans by electron transfer (eT) reactions. The first one is a three-step process that comprises bromination of o-arylphenols, Suzuki-Miyaura cross-coupling and photoinduced cyclization in order to obtain the above-mentioned products. The second one is a metal-free procedure and does not require any photocatalyst. Different solvents were tested, and the yields ranged from low to moderate. A comparison was established between both methodologies, showing that the second one is the most suitable for the synthesis of dibenzofurans.

A number of small-mol. inhibitors have been developed that target the catalytic domains of protein kinases that are not in an active conformation. An inactive form that has been observed in several kinases is the DFG-out conformation. This conformation is characterized by an almost 180° rotation of the conserved Asp-Phe-Gly (DFG) motif in the ATP-binding cleft relative to the active form. However, the sequence and structural determinants that allow a kinase to stably adopt the DFG-out conformation are not known. Here, we characterize a series of inhibitors based on a general pharmacophore for this inactive form. We demonstrate that modified versions of these inhibitors can be used to study the thermodn. and kinetics of ligand binding to DFG-out-adopting kinases and for enriching these kinases from complex protein mixtures

Carnitine, after extraction by appropriate solvents or liberation from biol. materials by acid hydrolysis, is separated electrophoretically. Separation is carried out on Whatman 3 MM paper at 400 v. by means of buffer prepared by adding pyridine to 0.3M AcOH to give a final pH of 4.1. Position of the carnitine spot is located with I vapor; the spot is cut out and eluted with HCl-EtOH, and carnitine is estimated by the method of Friedman (CA 52, 12978a) in which the ethyl ester is complexed in alk. medium with bromophenol blue. The procedure gives an error of the order of 10%. Application of the method to several betaines and the results are described.
