Preclin. experiments and clin. observations suggest the potential effectiveness of selective 5-HT1F receptor agonists in migraine. Identifying compounds with enhanced selectivity is crucial to assess its therapeutic value. Replacement of the indole nucleus in LY334370 (I) with a monocyclic Ph ketone moiety generated potent and more selective 5-HT1F receptor agonists. Focused SAR studies around this central Ph ring demonstrated that the electrostatic and steric interactions of the substituent with both the amide CONH group and the ketone C=O group play pivotal roles in affecting the adopted conformation and thus the 5-HT1F receptor selectivity. Computational studies confirmed the observed results and provide a useful tool in the understanding of the conformational requirements for 5-HT1F receptor agonist activity and selectivity. Through this effort, the 2-F-Ph and N-2-pyridyl series were also identified as potent and selective 5-HT1F receptor agonists.

The effect of inducers of systemic acquired resistance, dichloroisonicotinic acid (DCINA) and acibenzolar-S-Me (BION), on compatible interactions between Bremia lactucae and lettuce were examined using a detached cotyledon infection assay. Treatment with both activators caused a reduction in sporulation on susceptible cultivars Cobham Green challenged with isolate CL9W and Diana inoculated with isolate Tv, with DCINA being more effective than BION on an equimolar basis. Unexpectedly, treatment with both compounds suppressed the resistance conferred by the Dm7 gene in cv. Diana challenged by isolate CL9W (A7). The frequency of sporulation was greatly increased by DCINA in the incompatible interaction. The suppression of defense was associated with a delay in the onset of the Dm7-based hypersensitive reaction as indicated by the extended viability of penetrated epidermal cells, and reductions in both the accumulation of the phytoalexin lettucenin A and the deposition of autofluorescent phenolics such as syringaldehyde on plant and oomycete cell walls. The anal. of DCINA homologues indicated that 2-chloroisonicotinic acid was as effective as the dichloro-derivative in suppressing resistance in cv. Diana, whereas the absence of the carboxyl group rendered 2,6-dichloropyridine inactive. Infection of cotyledons by Botrytis cinerea was also found to be enhanced by DCINA treatment. Based on our results, we discuss the possibility that DCINA reduces Dm7 transcription through an epigenetic mechanism, as is supported by bioinformatic analyses of the resistance gene, and that it suppresses jasmonate-dependent resistance to B. cinerea.

Thaxtomin A (TA) is a natural cellulose biosynthesis inhibitor (CBI) synthesized by the potato common scab-causing pathogen Streptomyces scabies. Inhibition of cellulose synthesis by TA compromises cell wall organization and integrity, leading to the induction of an atypical program of cell death (PCD). These processes may facilitate S. scabies entry into plant tissues. To study the mechanisms that regulate the induction of cell deathin response to inhibition of cellulose synthesis, we usedArabidopsis thalianacell suspension cultures treated withtwo structurally different CBIs, TA and the herbicide isoxaben (IXB). The induction of cell death by TA and IXB was abrogated following pretreatment with the synthetic auxin2,4-dichlorophenoxyacetic acid (2,4-D) and the natural auxin indole-3-acetic acid (IAA). The addition of auxin efflux inhibitors also inhibited the CBI-mediated induction of PCD. This effect may be due to intracellular accumulation of auxin. Auxin has a wide range of effects in plant cells, including a role in the control of cell wall composition and rigidity to facilitate cell elongation. Using Atomic Force Microscopy (AFM)-based force spectroscopy, we found that inhibition of cellulose synthesis by TA and IXB in suspension-cultured cells decreased cell wall stiffness to a level slightly different than that caused by auxin. However, the cell wall stiffness in cells pretreated with auxin prior to CBI treatment was equivalent to that of cells treated with auxin only. Addition of auxin to Arabidopsis cell suspension cultures prevented the TA- and IXB-mediated induction of cell death. Cell survival was also stimulated by inhibition of polar auxin transport during CBI-treatment. Inhibition of cellulose synthesis perturbed cell wall mech. properties of Arabidopsis cells. Auxin treatment alone or with CBI also decreased cell wall stiffness, showing that the mech. properties of the cell wall perturbed by CBIs were not restored by auxin. However, since auxin’s effects on the cell wall stiffness apparently overrode those induced by CBIs, we suggest that auxin may limit the impact of CBIs by restoring its own transport and/or by stabilizing the plasma membrane – cell wall – cytoskeleton continuum.
