The synthesis of C-ring quinoxaline-substituted sinomenine 1,2,3-triazole derivatives at the 4-OH via click reactions was accomplished, and a total of 16 novel sinomenine double N-heterocyclic derivatives are obtained in 74%-95% yields. The C-ring is first transformed into a 1,2-diketone structure under the action of hydrochloric acid, and then reacted with o-phenylenediamine to obtain a C-ring quinoxaline-substituted structure. The 4-OH of sinomenine reacted with chloropropyne to give an alkynyl sinomenine, and then reacts with sodium azide and various benzyl chlorides to give the target compounds All the synthesized derivatives were characterized by Fourier-transform IR spectrometry, high resolution mass spectrometry, 1H NMR, and 13C NMR spectroscopy.

These transformations feature a simple protocol, broad functional group tolerance, high selectivity, and good to excellent yields. It is noteworthy that these reactions represent the first examples of the application of the selective Chan-Lam coupling.

The transcriptional regulator CprK controls the expression of the reductive dehalogenase CprA in organohalide-respiring bacteria. Desulfitobacterium hafniense CprA catalyzes the reductive dechlorination of the terminal electron acceptor o-chlorophenol acetic acid, generating the phenol acetic acid product. It has been shown that CprK has ability to distinguish between the chlorinated CprA substrate and the de-halogenated end product, with an estimated an estimated 104-fold difference in affinity. Using a green fluorescent protein GFPUV-based transcriptional reporter system, we establish that CprK can sense o-chlorophenol acetic acid at the nanomolar level, whereas phenol acetic acid leads to transcriptional activation only when approaching micromolar levels. A structure-activity relationship study, using a range of o-chlorophenol acetic-acid-related compounds and key CprK mutants, combined with pKa calculations on the effector binding site, suggests that the sensitive detection of chlorination is achieved through a combination of direct and indirect readout mechanisms. Both the phys. presence of the bulky chloride substituent as well as the accompanying electronic effects lowering the inherent phenol pKa are required for high affinity. Indeed, transcriptional activation by CprK appears strictly dependent on establishing a phenolate-K133 salt bridge interaction, rather than on the presence of a halogen atom per se. As K133 is strictly conserved within the CprK family, our data suggest that physiol. function and future applications in biosensing are probably restricted to phenolic compounds

The discovery and characterization of compound 48, a selective and in vivo active mGlu2 receptor pos. allosteric modulator (PAM), are described. Bentonite-supported nanoparticles of Cu, TiO2, and Ag were successfully synthesized and dispersed in a polyvinyl alc. (PVA)/glycerol (GLY) aqueous solution The decontamination effectiveness of the proposed solutions was evaluated by qual. and quant. anal. techniques on various micro-organisms, with sulfur mustard (HD) and di-Me methylphosphonate (DMMP) as contaminants. The results indicate that the peelable active nanocomposite films can be successfully used on contaminated surfaces to neutralize and entrap the hazardous materials and their degradation products. Mech. and thermal characterization of the polymeric films was also performed to validate the decontamination solution′s potential as peelable-film generating materials. The removal efficacy from the contaminated surfaces for the tested micro-organisms varied between 93% and 97%, while for the chem. agent HD, the highest decontamination factor obtained was 90.89%. DMMP was almost completely removed from the contaminated surfaces, and a decontamination factor of 99.97% was obtained.

Obesity-associated insulin resistance plays a central role in the pathogenesis of type 2 diabetes. A promising approach to decrease insulin resistance in obesity is to inhibit the protein tyrosine phosphatases that neg. regulate insulin receptor signaling. The low-mol.-weight protein tyrosine phosphatase (LMPTP) acts as a critical promoter of insulin resistance in obesity by inhibiting phosphorylation of the liver insulin receptor activation motif. Here, we report development of a novel purine-based chem. series of LMPTP inhibitors. These compounds inhibit LMPTP with an uncompetitive mechanism and are highly selective for LMPTP over other protein tyrosine phosphatases. We also report the generation of a highly orally bioavailable purine-based analog that reverses obesity-induced diabetes in mice.

The elec. mobility was demonstrated to be a simple function of the specific charge on the ion, a relation which holds also for closely related compounds such as creatine and creatinine which were separated sharply by electrorheophoresis. The specific charges differed greatly at different pH values. It was possible, when dissociation constants were known, to determine the pH for optimum separation of the different compounds The advantage of electrorheophoresis is that the rate of migration may be modified by changing the ionic force of the buffer used without altering the pH. The method is applicable to the separation of substances of biol. origin from attendant impurities.
