Transformations of anionic CuII chlorocomplexes have been studied under conditions of catalytic exchange reactions between carbon tetrachloride and n-alkanes. It was shown that chlorocuprates are just precursors and are easily reduced to the genuine catalysts, i.e., to the resp. CuI complexes. Both the composition and the geometric structure of the precursor (CuCl42-) and, probably, the active site (CuCl32-) have been investigated by several techniques (UV-vis spectroscopy, ESR, extended X-ray absorption fine structure (EXAFS), X-ray absorption near-edge structure (XANES), and static magnetic measurements). The dependence of the metathesis velocity on the [Cl]/[Cu] ratio was found to exhibit a maximum most likely corresponding to the highest content of trichlorocuprite CuCl32-.

Industrial effluents with dyes may contain appreciable concentrations of materials with high COD and suspended solids, posing adverse effects to both humans and aquatic life; therefore, these effluents require quant. monitoring. In the present study, an anal. method based on solid-phase extraction (SPE) and ultra-performance liquid chromatog.-tandem mass spectrometry (UPLC-MS/MS) has been optimized for the quant. anal. of methylene blue (MB) in environmental samples. For the extraction of MB, a variety of solvents, including formic acid, were investigated to obtain optimum recovery. MB extraction using the SPE method was best achieved using methanol with 1 M formic acid. Chromatog. separation of MB and methylene violet 3RAX (MV 3RAX, internal standard) was accomplished on an Acquity BEH C18 column using water (64.99%) with formic acid (0.01%) and acetonitrile (35%) in isocratic elution mode. Chromatog. separation for both MB and MV 3RAX was achieved in <2 min with good resolution and superior peak symmetry. MB and MV 3RAX were quantified with electrospray ionization coupled with MS/MS in the multiple reaction monitoring mode. Good quality parameters were achieved for instance linearity (r2 > 0.999), run-to-run and day-to-day precisions with relative standard deviations of <4%, sensitivity with a low limit of detection (LOD) of 0.1 ng mL-1 and limit of quantification (LOQ) of 0.4 ng mL-1 were obtained. Industrial wastewater samples (paper, textile, laundry and printing press) were analyzed, and an MB level was found in between 0.36 and 1.08 μg mL-1 with excellent recovery rates (95-99%) depending on samples.

The present study reported the successful synthesis and evaluation of a series of indole-picolinamide hybrids I, II [Ar = Ph, 3-CF3C6H4, 3,4-di-ClC6H3], III, IV and V as potent inhibitors of bovine carbonic anhydrase II, a promising therapeutic target for the treatment of neurol. disorders, osteoporosis, glaucoma, cancer and obesity. Various multistep synthetic approaches were utilized to access the desired structures having exclusive sites for chem. modifications and diverse spots for biol. interactions between the ligand and enzyme. Compound I was observed as a potent inhibitor of the bovine CA-II with an IC50 value of 0.0440 ± 0.009μM. The inhibition potency was about 22-fold higher than the standard drug acetazolamide (IC50 = 0.9618 ± 0.180μM). Several relevant structure-activity relationships were deduced that enlighten the crucial role of substituents as well as the key pharmacophores involved in the inhibition process. Mol. docking tools reinforced the in vitro assay results. The most active compound I was also investigated for anticancer potential using sulforhodamine B assay and results showed two-fold efficacy against HeLa cells when compared to standard drug cisplatin (IC50 = 8.045 ± 3.791μg/mL and 4.128 ± 1.473μg/mL, resp.). These findings were also confirmed by flow cytometry and comet tests. Moreover, fluorescence microscopy with DAPI staining revealed apoptosis as a mechanism of cancer cell death.

Thirteen novel isatin-ampyrone Schiff bases derivatives such as I [X = H, Cl; R = H, 3-F, 4-OMe, etc.] were synthesized by the reaction of isatins and benzyl halides followed by condensation with ampyrone. In vitro cytotoxic activity of these new Schiff bases against three human tumor cell lines (MCF-7, A549, and SCOV3) as well as normal breast cell line (MCF-10A) were evaluated by MTT assay. Structure-activity relationship of the tested compounds revealed that chlorine group at C-5 position of the isatin ring significantly increased the cytotoxic activity. This study generally led to introduce a highly active mol. I [X = Cl; R = 3-Cl] with IC50 values of 5.12, 25.5, and 12.9μM, on MCF-7, A549, and SCOV3, resp. Furthermore, mol. docking studies of the synthesized compounds were also done to investigate their binding modes to toward VEGFR-2 and JNK3-MAP kinase as the main targets for isatin-containing anticancer agents. Binding free energy values of the compounds showed pos. correlation with their cytotoxic activities. To confirm the docking results, mol. docking simulations of potent compound I [X = Cl; R = 3-Cl] against VEGFR-2 and JNK3 MAP kinase were also performed. According to the cytotoxic results and in silico ADMET predictions together, I [X = Cl; R = 3-Cl] could be considered as a potent candidate for the future anticancer studies.
