Novel 7-aryl benzo[1,4]oxazepin-2-ones were synthesized and evaluated as non-steroidal progesterone receptor (PR) modulators. The structure activity relationship of 7-aryl benzo[1,4]oxazepinones was examined using the T47D cell alk. phosphatase assay. A number of 7-aryl benzo[1,4]oxazepinones such as 10j and 10v demonstrated good in vitro potency (IC50 of 10-30 nM) and selectivity (over 100-fold) at PR over other steroidal receptors such as glucocorticoid and androgen receptors (GR and AR). Several 7-aryl benzo[1,4]oxazepinones were active in the rat uterine decidualization model. In this in vivo model, compounds 10j (I) and 10u (II) were active at 3 mg/kg when dosed orally.

The present work describes the synthesis of alkyl substituted N,4-di-Ph thiazole-2-amines (I) and (II) through a three step pathway. Further, the structures were confirmed by single crystal X-ray diffraction studies which revealed that compound I crystallizes in triclinic system with P-1 space group whereas compound II crystallizes as monoclinic with P21/c space group. The asym. unit consists of only one mol. in both the structures. The study of torsional angles of compound I and compound II indicated that S1 and C4 are cis to each other whereas N2 and C4 are trans. A detailed anal. of intermol. hydrogen bonding interactions has been performed. Also their C-H···π and π···π stacking interactions have been explored using Hirshfeld surface anal. and their 2D fingerprint plots. The most significant contributions in both the compounds are due to H···H interactions which also account for their stability.

Hydrocarbon-bonded stationary phases were prepared by reacting CH3(CH2)nSiMeCl2, where n = 7, 10, 14, and 17, with silica and were investigated and compared with phases obtained by the reaction of silica with alkyltrichlorosilanes. The reversed-phase chromatog. of halobenzenes with aq MeOH as mobile phase showed a linear relationship between the capacity ratios k‘ and the amount of C in the phases in all cases. The alkylmethyl phases were superior to the alkyl phases; capacity ratios and selectivities were increased and the efficiencies, with one exception, were higher. The improved properties are discussed in relation to residual silanol groups on the stationary phase surface.

A new class of nanoparticle-based dual-modality positron emission tomog./magnetic resonance imaging (PET/MRI) contrast agents has been developed. The probe consists of a superparamagnetic iron oxide (SPIO) or manganese oxide core coated with 3,4-dihydroxy-D,L-phenylalanine (dl-DOPA). The chelator 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) was conjugated to DOPA termini. The DOTA modified nanoparticles allow chelation of copper for PET imaging. These surface functionalized nanoparticle-based probes have been characterized by various anal. techniques. The cell-labeling efficacy, cytotoxicity and relaxivity of these nanoparticles have been evaluated and compared with the same properties of one of the most commonly utilized MRI contrast agents, Feridex. Evidently, this new nanoparticle has a great potential for use in cell tracking with MRI and PET in the absence of transfecting agent. It is noteworthy that there is a sharp increase in r2 relaxivity of these nanoparticles on coordination with Cu2+ ions. Thus these iron oxide nanoparticles can also be explored as the smart magnetic resonance (MR) sensor for the detection of micromolar changes in copper concentration for neurodegenerative diseases such as Alzheimer’s disease, Menkes and Wilson’s diseases, amyotrophic lateral sclerosis and prion diseases.

In the present work, a facile and efficient route for the synthesis of a series of N-substituted imidazole derivatives is described. Docking studies have revealed that N-substituted imidazole derivatives based on (E)-urocanic acid may be potential antihypertensive leads. Therefore, new AT1 receptor blockers bearing either the benzyl or the biphenylmethyl moiety at the N-1 or N-3 position, either the (E)-acrylate or the propanoate fragment and their related acids at the C-4 position as well as a halogen atom at the C-5 position of the imidazole ring [I; R = CH:CHCO2H, CH:CHCO2Me, CH2CH2 CO2H, CH2CH2CO2Me] and [II; X = H, R = CH:CHCO2R1 or CH2CH2CO2R1, R1 = H, Me, Q; X = Cl, Br, or iodo, R = CH2CH2R1, R1 = H or Q] were synthesized. The newly synthesized analogs were evaluated for binding to human AT1 receptor. The biol. results showed that this class of mols. possesses moderate or no activity, thus not always confirming high docking scores. Nonetheless, important conclusions can be derived for their mol. basis of their mode of action and help medicinal chemists to design and synthesize more potent ones. An aliphatic group as in losartan seems to be important for enhancing binding affinity and activity.
