We describe new synthetic routes developed toward a range of substituted analogs of bromo and extra-terminal (BET) bromodomain inhibitors I-BET762/JQ1 based on the triazolo-benzodiazepine scaffold. These new routes allow for the derivatization of the methoxyphenyl and chlorophenyl rings, in addition to the diazepine ternary center and the side chain methylene moiety. Substitution at the level of the side chain methylene afforded compounds targeting specifically and potently engineered BET bromodomains designed as part of a bump and hole approach. We further demonstrate that marked selectivity for the second over the first bromodomain can be achieved with an indole derivative that exploits differential interaction with an aspartate/histidine conservative substitution on the BC loop of BET bromodomains.

Herein, diastereo- and regioselective synthesis of trisubstituted alkenes via nickel-catalyzed benzylation and isomerization of terminal olefins with benzyl chlorides in presence of trimethylsilyl triflate and trimethylamine additives was described. Control experiments provided evidence for a mechanism involving branched-selective Heck-type benzylation that overrides substrate control, followed by trans-selective 1,3-hydrogen shift. The method represented a significant addition to the toolbox of reactions for the concise synthesis of unsaturated biol. active compounds

A redox-neutral ortho functionalization of aryl boroxines via Pd/NBE catalysis was reported. An electrophile, such as carboxylic acid anhydrides or O-benzoyl hydroxylamines, was coupled at the boroxine ortho position, and a proton as the second electrophile was introduced at the ipso position. This reaction did not require extra oxidants or reductants and avoided stoichiometric bases or acids, thereby tolerating a wide range of functional groups. In particular, orthogonal chemoselectivity between aryl iodide and boroxine moieties was demonstrated, which could be used to control reaction sequences. Finally, a deuterium-labeling study supported the ipso protonation pathway. This unique mechanistic feature could inspire the development of a new class of Pd/NBE-catalyzed transformations.

A novel catalyst by preparing silica supported EDTA (EDTA) platinum complex was developed. The preparation conditions, such as heating temperature and solvent types, were optimized. The catalyst was phosphine-free and sulfur-free which was characterized by IR spectroscopy (IR), energy dispersive X-ray spectrometer (EDS), transmission electron microscopy (TEM), high resolution transmission electron microscopy (HRTEM), XPS, UV spectroscopy (UV) and at. absorption spectroscopy (AAS). Its catalytic activity was evaluated by catalyzing solventless hydrosilylation reactions. In the hydrosilylation reaction between 1-hexene and methyldichlorosilane, the new catalyst showed a higher catalytic activity than that provided by Speier’s catalyst. Moreover, the new catalyst could be reused for 12 times without obvious loss of catalytic activity. To confirm its practical feasibility, catalyzing the hydrosilylation reaction between 1-octene and methyldichlorosilane was performed. Our results indicated that the new catalyst can efficiently catalyze the hydrosilylation reaction.

Platinum was immobilized on magnetic silica gel by means of boronic, nitric, carboxylic or sulfuric acid ligands to give four heterogeneous Pt nano-catalysts, designated as Fe3O4@SiO2-BA@Pt, Fe3O4@SiO2-NA@Pt, Fe3O4@SiO2-CA@Pt and Fe3O4@SiO2-SA@Pt, resp. Particles of these mono-dispersible Pt catalysts were 10-20 nm in size and could be separated for recycling by means of a magnet. Fe3O4@SiO2-BA@Pt (0.174 mmol/g Pt) showed the best catalytic activity and selectivity, which were better than Speier’s catalyst. Its turnover numbers were up to 1.7 X 106 and 1.1 X 106 for hydrosilylation of 1-hexene or styrene, resp. This material could also catalyze the hydrosilylation of a broad range of alkenes and alkynes as substrates and methyldichlorosilane or triethoxysilane as silanes. Similar yields of 1-hexyl-methyldichlorosilane at the first and eighth runs (96.5% and 95.2%, resp.), together with a final Pt content of 0.171 mmol/g indicated the outstanding stability of Fe3O4@SiO2-BA@Pt under the catalytic reaction conditions.

A smart design of transition metal nanocatalyst can efficiently improve its catalytic activity and use value. In this work, platinum (Pt) was successfully anchored to silica gel with phenylboronic acid group first time and formed nanoparticles (0.5-5 nm) which afforded a highly active supported catalyst for the hydrosilylation of olefins. The turnover number and regioselectivity in the reaction of methyldichlorosilane and 1-octene using 0.00000004 mol of the immobilized Pt catalyst approached 1,321,317 and 99% over 12 h. The corresponding hydrosilylation products in excellent yields and regioselectivities were obtained when we employed a broad range of terminal aliphatic olefins and methyldichlorosilane as substrates. The fully immobilized Pt nanoparticles could be recycled at least 10 runs without any obvious loss in catalytic activity.

Shao, Dongyun published the artcilePreparation of polycarboxylic acid-functionalized silica supported Pt catalysts and their applications in alkene hydrosilylation, HPLC of Formula: 14799-94-1, the publication is RSC Advances (2018), 8(36), 20379-20393, database is CAplus and MEDLINE.

A series of novel immobilized platinum catalysts was prepared by loading Pt onto silica particles modified with polycarboxylic acid groups such as diethylenetriaminepentaacetic acid (DTPA), nitrilotriacetic acid (NTA) and succinic acid (SA). The three modified heterogeneous Pt catalysts were characterized using IR spectroscopy (IR), transmission electron microscopy (TEM), XPS, energy dispersive X-ray spectroscopy (EDS) and at. absorption spectroscopy (AAS). The residual H2PtCl6 solutions were characterized using UV spectroscopy (UV). The polycarboxylic acid-functionalized silica supported Pt catalysts were used to catalyze alkene hydrosilylation and 1-hexene was chosen as a model alkene. The data indicated that the catalytic performance was strongly dependent on the properties of the polycarboxylic acid group bonded to the silica particles. Among them, DTPA-functionalized silica supported Pt (SiO2-DTPA-Pt) showed the best catalytic activity and reusability. Furthermore, some hydrosilylation reactions between other linear alkenes (1-heptene, 1-octene, 1-decene, 1-do-decene, 1-tetra-decene, 1-hexa-decene, 1-octa-decene, styrene or cis-hex-2-ene), or ring type alkenes (norbornene) with methyldichlorosilane could be catalyzed in the presence of these three Pt catalysts. Their high activities were more than 90%, and their selectivities were more than 99%, which were apparently better than homogeneous Pt catalysts. In addition, reactions with cyclohexene were also successfully catalyzed by the Pt catalysts. These results indicate that the polycarboxylic acid-functionalized silica gel supported Pt catalysts have potential value in industrial hydrosilylation reactions.
