This procedure is scalable and compatible with a wide range of functional groups and heteroarenes, providing an operationally simple protocol for the synthesis of o-hydroxybenzamides. The hydroxylation of nicotinamides gave 4-oxo-1,4-dihydropyridine-3-carboxamides selectively. Preliminary mechanistic studies implicate that a basic ligand-enabled, irreversible, rate-determining CMD step is most likely involved in this process.

A copper/silver-mediated oxidative ortho-ethynylation of unactivated aryl C-H bonds with terminal alkynes has been developed. The reaction uses the removable PIP [PIP = 2-(pyridin-2-yl)isopropyl] directing group and features broad substrate scope, high functional-group tolerance, and compatibility with a wide range of heterocycles, providing an efficient synthesis of aryl alkynes. E.g., Cu(OAc)2 and Ag2CO3 catalyzed the ethynylation of benzene derivative (I) with CHCTIPS to give 42% II. This procedure highlights the potential of copper catalysts to promote unique synthetically enabling C-H functionalization reactions that lie outside of the current scope of precious metal catalysis.

Reported is a novel palladium(II)-initiated Catellani-type reaction that uses widely accessible aryl boronic acids as the substrates instead of aryl halides, thereby greatly expanding the existing scope of this powerful transformation. This borono-Catellani reaction was promoted by cooperative catalysis between Pd(OAc)2 and the inexpensive 5-norbornene-2-carbonitrile. Practicality is the striking feature of the reaction: it is run open to air at ambient temperature and no phosphine ligand is needed. This mild, chemoselective, and scalable protocol is compatible with a large range of readily available functionalized aryl boronic acids and bromides, as well as terminating olefins (50 examples, 39-97% yields). Moreover, the orthogonal reactivity between the borono-Catellani and classical Catellani reaction was demonstrated. This work is expected to open new avenues for developing novel Catellani-type reactions.

In order to evaluate scientifically the differences in the flavor of macadamia nuts dried with different methods, electronic tongue and electronic nose sensory technologies combined with gas chromatog.-mass spectrometry (GC-MS) were used to analyze their flavor quality. The results showed that there were significant differences between fresh macadamia nuts and different dried macadamia nuts in umami, salty, sour and bitter tastes, and the cumulative contribution rate of the first two principal components of linear discriminant anal. (LDA) was 96.60%, which could distinguish completely macadamia nuts dried in different ways. The responses of the electronic nose to macadamia nuts processed differently differed significantly. The cumulative contribution rate of the first two principal components in principal component anal. (PCA) reached 98.79%, which could well distinguish the volatile flavor compounds of the macadamia nuts dried by different treatments. The sensors W1S, W3S and W3C played major roles in the principal component anal. As a supplement to the electronic nose, GC-MS identified a total of 227 volatile components, with alkanes and aldehydes being dominant in types and contents. The relative contents of alkanes in the four different treatments of macadamia nuts were 26.29%, 40.76%, 23.64% and 39.46%, resp., and the relative contents of aldehydes were 29.01%, 18.96%, 26.03% and 23.87%, resp. Therefore, the combination of electronic tongue, electronic nose and GC-MS can better distinguish macadamia nuts dried with different methods, thereby providing a theor. reference for postharvest primary processing of macadamia nuts in growing regions.

In order to find novel diacylhydrazine derivatives which possessed high activities against pests, 20 novel N-(tert-butyl)-N’-(fluorobenzoyl)pyridinecarboxylic acid hydrazide derivatives of formula I [Py = substituted pyridine; R = H, F] were synthesized by introducing fluoro-contained Ph and pyridine into the basic structure of diacylhydrazines. Their structures were confirmed by 1H NMR, 13C NMR, IR and HRMS. The bioassay results indicated that most of the compounds exhibited more than 80 % mortality against Mythimna separata at the concentration of 500 mg/L. Among these compounds, N-(tert-butyl)-N’-(2,6-difluorobenzoyl)-N-2-chloronicotinohydrazine and N-(tert-butyl)-N’-(2,6-difluorobenzoyl)-N-2,5-dichloronicotinohydrazine exhibited 100 % and 80 % mortality against Mythimna separata at 10 mg/L, resp. The LC50 of compound N-(tert-butyl)-N’-(2,6-difluorobenzoyl)-N-2-chloronicotinohydrazine against Mythimna separata was 8.77 mg/L, and it showed about 0.51 times of toxicity against Mythimna separata than tebufenozide and had 1.5 times of toxicity against Plutella xylostella than tebufenozide.

In conclusion, after in vivo and in vitro experiments, we successfully acquired an effective candidate (MZ58) showing antitumor effects with low cytotoxicity toward T cells as well as the ability to activate CD8+ T cells and reduce T cell exhaustion.

A series of novel thieno[2,3-d]pyrimidine analogs were designed and synthesized as KRAS G12D inhibitors via combinatorial virtual screening approach. Most compounds exhibited potent antiproliferative activity on KRAS G12D mutated cancer cell lines (Panc1, SW1990 and CT26) with IC50 values in the low micromolar range. Among them, I (R = 2-OCH3-5-CF3) showed the highest antiproliferative activity with an average IC50 of 2.1μM against three KRAS G12D-mutated cells (Panc1, SW1990 and CT26). CompoundI (R = 2-OCH3-5-CF3) decreased the active form of KRAS (KRAS-GTP) in KRAS G12D mutated cancer cell lines (CT26 and SW1990) but not in KRAS G13D mutated cancer cells (HCT116). Moreover, I (R = 2-OCH3-5-CF3) down-regulated the phosphorylated Raf and Erk in CT26 and SW1990 cancer cell lines but not in HeLa cells (KRAS WT). The binding affinity of I (R = 2-OCH3-5-CF3) was further confirmed by the isothermal titration calorimetry (ITC) assay in which I (R = 2-OCH3-5-CF3) exhibited a KD of 33 nM for binding to KRAS G12D protein. In addition, I (R = 2-OCH3-5-CF3) (40 mg/kg or 60 mg/kg) exhibited significant antitumor efficacy in a CT26 tumor model with a tumor growth inhibition (TGI) of 42% or 53% without causing apparent toxicity. Taken together the above results suggest that I (R = 2-OCH3-5-CF3) is a promising KRAS G12D inhibitor deserving further investigation.
