Ln(NO3)(3)center dot nH(2)O reacted with 2,4-dichlorobenzoic acid (2,4-HDCB) and NaOH under solvothermal conditions of water and ethanol to yield four lanthanide-organic coordination polymers (CPs) [Eu(2,4-DCB)(3)(CH3CH2OH)(2)] (1) and [Ln(2,4-DCB)(3)(H2O)] [Ln = Gd (2), Tb (3), Dy (4)]. The second ligand 4,4′-bipyridine (4,4′-bpy) was introduced into the above reaction systems to provide four new lanthanide-organic CPs [Ln(2)(2,4-DCB)(6)(4,4′-bpy)(H2O)(2)] [Ln = Eu (5), Gd (6), Tb (7), Dy (8)]. Single-crystal and powder X-ray diffraction analyses reveal that CPs 1 and 2 possess similar one-dimensional (1D) structures, and CPs 2-4 and 5-8 are isostructural, respectively. In 1-4, the 2,4-DCB anions bridge the Ln(III) ions to form a 1D chain. In 5-8, six 2,4-DCB anions link two Ln(III) ions into a binuclear Ln(2)(2,4-DCB)(6) unit, and the Ln(2)(2,4-DCB)(6) units are further connected by the 4,4′-bpy co-ligands to generate a 1D chain. The photoluminescent and magnetic properties of 1-8 were investigated.

A series of novel N-phenylbenzamide-4-methylamine acridine derivatives were designed and synthesized based initially on the structure of amsacrine (m-AMSA). Molecular docking suggested that the representative compound 9a had affinity for binding DNA topoisomerase (Topo) II, which was comparable with that of m-AMSA, and furthermore that 9a could have preferential interactions with Topo I. After synthesis of 9a and analogues 9b-9f, these were all tested in vitro and the synthesized compounds displayed potent antiproliferative activity against three different cancer cell lines (K562, CCRF-CEM and U937). Among them, compounds 9b, 9c and 9d exhibiting the highest activity with IC50 value ranging from 0.82 to 0.91 mu M against CCRF-CEM cells. In addition, 9b and 9d also showed high antiproliferative activity against U937 cells, with IC50 values of 0.33 and 0.23 mu M, respectively. The pharmacological mechanistic studies of these compounds were evaluated by Topo I/II inhibition, western blot assay and cell apoptosis detection. In summary, 9b effectively inhibited the activity of Topo I/II and induced DNA damage in CCRF-CEM cells and, moreover, significantly induced cell apoptosis in a concentration-dependent manner. These observations provide new information and guidance for the structural optimization of more novel acridine derivatives.

In this paper, a comprehensive method for the identification of the unknown halogenated DBPs (X-DBPs, X = CI, Br, and I) in the tap water of Wuhan, China via liquid chromatography-high resolution mass spectrometry (LC-HRMS) was developed. 123 X-DBPs were identified through the stepwise procedure, 94 of them were newly identified, and 3 of them were confirmed by standards. Most X-DBPs were aliphatic compounds and highly unsaturated and phenolic compounds, some X-DBPs contained multiple halogen atoms and rich in carboxyl groups, such as C2H2O2BrCl, C2H2O2Br2, and C2H2O2ClI. It was worth noting that the concentration of some X-DBPs had the same trend with time. Most CI-DBPs remained stable and I-DBPs were detected occasionally by monitoring the change of concentration of these X-DPBs with the time during three consecutive months. The results demonstrate that the proposed method could provide valuable molecular formula and structure information on unknown multiple halogenated DBPs, or be used for the identification of other multiple halogenated organic compounds in different media. Adsorption of a diverse set of chemicals onto goethite was evaluated by column chromatography. The pH of the effluents was 4.7-5.2. Van der Waals forces dominate the exothermic adsorption of 8 nonpolar compounds (e.g., PAHs and chlorobenzenes). H-bonding is responsible for the adsorption of 32 mono-carboxylic acids (i.e., benzoic acids, naphthoic acids and acidic pharmaceuticals) and their adsorption tends to be endothermic. Steric effects significantly decreased the bonding of monocarboxylic acids with ortho-substitutions. Exothermic adsorption of 10 monophenols is controlled by weak H-bonding. Bonding of these 50 solutes onto goethite is totally reversible. In contrast, inner-sphere complexation of phthalic acid and chlortetracycline with goethite occurred according to their low desorption ratio (1.1%-54.4%). Polyparameter linear free energy relationship (PP-LFER) models were established to provide acceptable fitting results of the goethite-solute distribution coefficients (RMSE = 0.32 and 0.30 at 25 degrees C and 5 degrees C, respectively). In this paper, synthesized Fe3O4 nanoparticles applied as catalyst in photodegradation of Direct Red 23 (DR23) dye using photo-Fenton process in aqueous solution. The Fe3O4 nanoparticles were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FT-IR). Design of experiments (DOE) based on Taguchi approach was used. Analysis the response of each experiment was based Signal to Noise (S/N) ratio was calculated. The effective parameters for the degradation of dye were determined and optimized using Taguchi (L-9 (3(4))) orthogonal array experimental design method with four factors having three levels for each factor. The Taguchi approach showed that pH 3 (level 1), catalyst amount = 25 mg/L (level 2), H2O2 concentration = 15 ppm (level 3), and temperature = 35 degrees C (level 3) was optimum conditions for this process. The factor most influencing the process was determined using analysis of variance (ANOVA) method. The most significant factor in this process was pH. The interaction between pH x catalyst amount was the most influencing interaction. The percent (p (%)) of each factor on the degradation of dye was found to be in the following the order: pH (50.306%), catalyst amount (6.887%), H2O2 concentration (39.272%), and temperature (3.456%). The percentage contribution of factors in this process was found to be in the following the order: pH (0.332), catalyst amount (0.101), H2O2 concentration (0.291), and temperature (0.082). So first order reaction with k = 0.0472 min(-1) was observed for the photocatalytic degradation reaction.

Background: Postprandial hyperglycemia can be reduced by inhibiting major carbohydrate hydrolyzing enzymes, such as alpha-glucosidase and alpha-amylase which is an effective approach in both preventing and treating diabetes. Objective: The aim of this study was to synthesize a series of 2,4-dichloro-5-[(N-and/alkyl)sulfamoyl]benzoic acid derivatives and evaluate alpha-glucosidase and alpha-amylase inhibitory activity along with molecular docking and in silico ADMET property analysis. Method: Chlorosulfonation of 2,4-dichloro benzoic acid followed by reaction with corresponding anilines/amines yielded 2,4-dichloro-5-[(N-aryl/alkyl)sulfamoyl]benzoic acid derivatives. For evaluating their antidiabetic potential alpha-glucosidase and alpha-amylase inhibitory assays were carried out. In silico molecular docking studies of these compounds were performed with respect to these enzymes and a computational study was also carried out to predict the drug-likeness and ADMET properties of the title compounds. Results: Compound 3c (2,4-clichloro-5-[(2-nitrophenyl)sulfamoyl]benzoic acid) was found to be highly active having 3 fold inhibitory potential against alpha-amylase and 5 times inhibitory activity against alpha-glucosidase in comparison to standard drug acarbose. Conclusion: Most of the synthesized compounds were highly potent or equipotent to standard drug acarbose for inhibitory potential against alpha-glucosidase and alpha-amylase enzyme and hence this may indicate their antidiabetic activity. The docking study revealed that these compounds interact with active site of enzyme through hydrogen bonding and different pi interactions.

In continuance with earlier reported work, an extension has been carried out by the same research group. Mulling over the ongoing condition of resistance to existing antimalarial agents, we had reported synthesis and antimalarial activity of certain pyrazole-1,3,4-oxadiazole hybrid compounds. Bearing previous results in mind, our research group ideated to design and synthesize some more derivatives with varied substitutions of acetophenone and hydrazide. Following this, derivatives 5a-r were synthesized and tested for antimalarial efficacy by schizont maturation inhibition assay. Further, depending on the literature support and results of our previous series, certain potent compounds (5f, 5n and 5r) were subjected to Falcipain-2 inhibitory assay. Results obtained for these particular compounds further strengthened our hypothesis. Here, in this series, compound 5f having unsubstituted acetophenone part and a furan moiety linked to oxadiazole ring emerged as the most potent compound and results were found to be comparable to that of the most potent compound (indole bearing) of previous series. Additionally, depending on the available literature, compounds (5a-r) were tested for their antileishmanial potential. Compounds 5a, 5c and 5r demonstrated dose-dependent killing of the promastigotes. Their IC50 values were found to be 33.3 +/- 1.68, 40.1 +/- 1.0 and 19.0 +/- 1.47 mu g/mL respectively. These compounds (5a, 5c and 5r) also had effects on amastigote infectivity with IC50 of 44.2 +/- 2.72, 66.8 +/- 2.05 and 73.1 +/- 1.69 mu g/mL respectively. Further target validation was done using molecular docking studies. Acute oral toxicity studies for most active compounds were also performed.

The anionic form-dependent binding interaction of halo-phenolic substances with human transthyretin (hTTR) has been observed previously. This indicates that ionizable compounds should be the primary focus in screening potential hTTR disruptors. Here, the potential binding potency of halo-benzoic acids, halo-benzenesulfonic acids/sulfates and halo-phenylboronic acids with hTTR was determined and analyzed by competitive fluorescence displacement assay integrated with computational methods. The laboratorial results indicated that the three test groups of model compounds exhibited a distinct binding affinity to hTTR. All the tested halo-phenylboronic acids, some of the tested halo-benzoic acids and halo-benzenesulfonic acids/sulfates were shown to be inactive with hTTR Other halo-benzoic acids and halo-benzenesulfonic acids/sulfates were moderate and/or weak hTTR binders. The binding affinity of halo-benzoic acids and halo-benzenesulfonic acids/sulfates with hTTR was similar. The low distribution ability of the model compounds from water to hTTR may be the reason why they exhibited the binding potency observed with hTTR. By introducing other highly hydrophobic compounds, we observed that the binding affinity between compounds and hTTR increased with increasing molecular hydrophobicity. Those results indicated that the highly hydrophobic halo-benzoic acids and halo-benzenesulfonic acids/sulfates may be high-priority hTTR disruptors. Finally, a binary classification model was constructed employing three predictive variables. The sensitivity (S-n), specificity (S-p), predictive accuracy (Q) values of the training set and validation set were >0.83, indicating that the model had good classification performance. Thus, the binary classification model developed here could be used to distinguish whether a given ionizable compound is a potential hTTR binder or not. (C) 2019 Elsevier Ltd. All rights reserved.

Objectives: To evaluate the performance of a high-throughput research assay for HIV drug resistance testing based on whole genome next-generation sequencing (NGS) that also quantifies HIV viral Load. Methods: Plasma samples (n = 145) were obtained from HIV-positive MSM (HPTN 078). Samples were analysed using clinical assays (the ViroSeq HIV-1 Genotyping System and the Abbott RealTime HIV-1 Viral Load assay) and a research assay based on whole-genome NGS (veSEQ-HIV). Results: HIV protease and reverse transcriptase sequences (n =142) and integrase sequences (n =138) were obtained using ViroSeq. Sequences from all three regions were obtained for 100 (70.4%) of the 142 samples using veSEQ-HIV; results were obtained more frequently for samples with higher viral Loads (93.5% for 93 samples with >5000 copies/mL; 50.0% for 26 samples with 1000-5000 copies/mL; 0% for 23 samples with <1000 copies/mL). For samples with results from both methods, drug resistance mutations (DRMs) were detected in 33 samples using ViroSeq and 42 samples using veSEQ-HIV (detection threshold: 5.0%). Overall, 146 major DRMs were detected; 107 were detected by both methods, 37 were detected by veSEQ-HIV only (frequency range: 5.0%-30.6%) and two were detected by ViroSeq only. HIV viral Loads estimated by veSEQ-HIV strongly correlated with results from the Abbott RealTime Viral Load assay (R-2 = 0.85; n = 142). Conclusions: The NGS-based veSEQ-HIV method provided results for most samples with higher viral Loads, was accurate for detecting major DRMs, and detected mutations at Lower Levels compared with a method based on population sequencing. The veSEQ-HIV method also provided HIV viral Load data.
