Among them, compound II with 2-(4-fluorobenzyloxy) group was the most potent direct inhibitor of Keap1-Nrf2 PPI with an IC50 of 64.5 nM in the fluorescent polarization (FP) assay and 14.2 nM in a time-resolved fluorescence resonance energy transfer (TR-FRET) assay. Moreover, cell-based biol. assay showed that compound II significantly increased the mRNA levels of Nrf2 downstream genes, GSTM3, HMOX2 and NQO1, through Nrf2 activation. The discovery of the new scaffolds possessing diverse O-linked fragments at the C2 position offers opportunities to further modify the chem. structures of Keap1-Nrf2 PPI inhibitors to improve their pharmacokinetic, efficacy and safety profiles.

Lee, Sumin;Rovis, Tomislav research published 《 Rh(III)-Catalyzed Three-Component Syn-Carboamination of Alkenes Using Arylboronic Acids and Dioxazolones》, the research content is summarized as follows. A Rh(III)-catalyzed three-component carboamination of alkenes, e.g., 1,4-dihydro-1,4-epoxynaphthalene from readily available aryl boronic acids R1B(OH)2 (R1 = C6H5, 1-naphthyl, 2H-1,3-benzodioxol-5-yl, etc.) as a carbon source and dioxazolones I (R2 = CH3, cyclopropyl, 2-cyclohexylethyl, etc.) as nitrogen electrophiles is described. This protocol provides facile access to valuable amine products including α-amino acid derivatives, e.g., II in good yield and regioselectivity without the need for a directing functionality. A series of experiments suggest a mechanism in which the Rh(III) catalyst undergoes transmetalation with the aryl boronic acid, followed by turnover limiting alkene migratory insertion into the Rh(III)-aryl bond. Subsequently, fast Rh-nitrene formation provides the syn-carboamination product selectively after reductive elimination and proto-demetalation. Importantly, the protocol provides three-component coupling products in preference to a variety of two-component undesired byproducts.

An anti-folded bowl-shaped bisdibenzocorannulene (BDBC) featuring a new chair-cyclohexane-like hexagon as a bridge of two dibenzocorannulene moieties was reported. The neutral compound showed multiple redox-active properties and could be converted to the corresponding redox states through chem. reduction or oxidation Chem. reduction of BDBC by stoichiometric addition of metallic potassium in the presence of [18]crown-6 ether, provided a radical anion BDBC. and a dianion BDBC2-, resp.; while chem. oxidation by silver hexafluoroantimonate(V), converted the neutral compound to an open-shell singlet diradical dication (BDBC••)2+. The structural consequences of both electron-reduction and oxidation were closely related to the release of ring-strain of the bowl-shaped π-scaffold and imposed steric hindrance of the hexagonal bridge. In addition, the unusual open-shell nature of the dication could mainly be attributed to the changing of localized antiaromaticity in the closed-shell structure to delocalized character in the biradical, and thus the emergence of weakly bonded π-electrons.

An improved, gram-scale synthesis of the blockbuster drug sildenafil, used for the treatment of male erectile dysfunction, has been developed. Unlike the previous literature, the current method demonstrates the use of arylacetic acid as an acyl source, a cheap oxidant K2S2O8, and water as the reaction medium in the key step of pyrazolo[4,3-d]pyrimidin-7-one ring formation. As well as being a green and benign approach, the current method reduces the cost by half compared to our previous strategy. In addition, the general relevance of the method has been demonstrated in the synthesis of a variety of quinazolinone and benzothiazole derivatives with excellent functional group tolerance.
