Polukeev, Alexey V.;Wallenberg, Reine;Uhlig, Jens;Hulteberg, Christian P.;Wendt, Ola F. research published 《 Iridium-Catalyzed Dehydrogenation in a Continuous Flow Reactor for Practical On-Board Hydrogen Generation From Liquid Organic Hydrogen Carriers》, the research content is summarized as follows. To enable the large-scale use of hydrogen fuel cells for mobility applications, convenient methods for on-board hydrogen storage and release are required. A promising approach is liquid organic hydrogen carriers (LOHCs), since these are safe, available on a large scale, and compatible with existing refueling infrastructure. Usually, LOHC dehydrogenation is carried out in batch-type reactors by transition metals and their complexes and suffers from slow H2 release kinetics and/or inability to reach high energy d. by weight, owing to low conversion or the need to dilute the reaction mixture In this study, a continuous flow reactor is used in combination with a heterogenized iridium pincer complex, which enables a tremendous increase in LOHC dehydrogenation rates. Thus, dehydrogenation of isopropanol is performed in a regime that, in terms of gravimetric energy d., hydrogen generation rate, and precious metal content, is potentially compatible with applications in a fuel-cell powered car.

Currently, the therapeutic options for treatment of liver failure are very limited. As mitogen-activated protein kinase kinase 4 (MKK4) has recently been identified by in vivo RNAi experiments to be a major regulator in hepatocyte regeneration, authors pursued the development of a small mol. targeting this protein kinase. Starting from the approved BRAFV600E inhibitor vemurafenib, that showed a high off-target affinity to MKK4 in an initial screening, authors followed a scaffold-hopping approach, changing the core heterocycle from 1H-pyrrolo[2,3-b]pyridine to 1H-pyrazolo[2,3-b]pyridine I. Affinity to MKK4 could be conserved while the selectivity against off-target protein kinases was slightly improved. Further modifications led to II and III showing high affinity to MKK4 in the low nanomolar range and excellent selectivity profile from mandatory multiparameter-optimization for the essential anti-targets (MKK7, JNK1) and off-targets (BRAF, MAP4K5, ZAK) in the MKK4 pathway. Herein authors report the first selective MKK4 inhibitors in this class.

Dechlorination of these complexes with sodium tetrakis[3,5-bis(trifluoromethyl)phenyl]borate (NaBArF4) yielded their cationic derivatives [{μ-(IDipp)PH}{Ir(cod)}2{μ-Cl}][BArF4] (4) and [{(IDipp)PH}Ir(cod)(PPh3)][BArF4] (5). All complexes were structurally characterized and showed the expected square-planar coordination geometry and significant polarization of the P-C bond of the NHC-phosphinidene adduct upon metal coordination. The complexes 4 and 5 were tested for their applicability in catalytic H/D exchange reactions, and both complexes showed high activity for a broad range of aromatic substrates, ranging from ketones, amides, esters, nitro compounds and heterocycles.

C.;Zheng, Jia;Krajangsri, Suppachai;Andersson, Pher G. research published 《 Stereoselective Iridium-N,P-Catalyzed Double Hydrogenation of Conjugated Enones to Saturated Alcohols》, the research content is summarized as follows. A study on the effect of additives on the Ir-N,P-catalyzed hydrogenation of enones was described. The combination of benzamide and the development of a reactive catalyst unlocked a novel reactivity mode of Crabtree-type complexes toward C=O bond hydrogenation. The role of benzamide was suggested to extend the lifetime of the dihydridic iridium intermediate, which was prone to undergo irreversible trimerization, deactivating the catalyst. This unique reactivity was then coupled with C=C bond hydrogenation for the facile installation of two contiguous stereogenic centers in high yield and stereoselectivity (up to 99% ee, 99/1 d.r.) resulting in a highly stereoselective reduction of enones.

Fungal infections remain a high-incidence worldwide health problem that is aggravated by limited therapeutic options and the emergence of drug-resistant strains. Cinnamic and benzoic acid amides have previously shown bioactivity against different species belonging to the Candida genus. Here, 20 cinnamic and benzoic acid amides were synthesized and tested for inhibition of C. krusei ATCC 14243 and C. parapsilosis ATCC 22019. Five compounds inhibited the Candida strains tested, with compound 16 (MIC = 7.8 μg/mL) producing stronger antifungal activity than fluconazole (MIC = 16 μg/mL) against C. krusei ATCC 14243. It was also tested against eight Candida strains, including five clin. strains resistant to fluconazole, and showed an inhibitory effect against all strains tested (MIC = 85.3-341.3 μg/mL). The MIC value against C. krusei ATCC 6258 was 85.3 mcg/mL, while against C. krusei ATCC 14243, it was 10.9 times smaller. This strain had greater sensitivity to the antifungal action of compound 16. The inhibition of C. krusei ATCC 14243 and C. parapsilosis ATCC 22019 was also achieved by compounds 2, 9, 12, 14 and 15. Computational experiments combining target fishing, mol. docking and mol. dynamics simulations were performed to study the potential mechanism of action of compound 16 against C. krusei. From these, a multi-target mechanism of action is proposed for this compound that involves proteins related to critical cellular processes such as the redox balance, kinases-mediated signaling, protein folding and cell wall synthesis. The modeling results might guide future experiments focusing on the wet-lab investigation of the mechanism of action of this series of compounds, as well as on the optimization of their inhibitory potency.

The development of efficient and straightforward methods for obtaining all optically active isomers of structurally rigid spirocycles from readily available starting materials is of great value in drug discovery and chiral ligand development. However, the stereodivergent synthesis of spirocycles bearing multiple stereocenters remains an unsolved challenge owing to steric hindrance and ring strain. Herein, we report an enantio- and diastereodivergent synthesis of rigid spirocycles through dual-metal-catalyzed [3+2] annulation of oxy π-allyl metallic dipoles with less commonly employed nucleophilic dipoles (imino esters). A series of spiro compounds bearing a pyrroline and an olefin were easily synthesized in an enantio- and diastereodivergent manner (up to 19:1 dr, >99% ee), which showed great promise as a new type of N-olefin ligand. Preliminary mechanistic studies were also carried out to understand the process of this bimetallic catalysis.

MicroRNAs (miRNAs) are key players in human hepatocellular carcinoma (HCC) tumorigenesis. Therefore, small mols. targeting components of miRNA biogenesis may provide new therapeutic means for HCC treatment. By a high-throughput screening and structural simplification, we identified a small mol., CIB-3b (I), which suppresses the growth and metastasis of HCC in vitro and in vivo by modulating expression profiles of miRNAome and proteome in HCC cells. Mechanistically, CIB-3b phys. binds to transactivation response (TAR) RNA-binding protein 2 (TRBP) and disrupts the TRBP-Dicer interaction, thereby altering the activity of Dicer and mature miRNA production Structure-activity relationship study via the synthesis of 45 CIB-3b derivatives showed that some compounds exhibited a similar inhibitory effect on miRNA biogenesis to CIB-3b. These results support TRBP as a potential therapeutic target in HCC and warrant further development of CIB-3b along with its analogs as a novel therapeutic strategy for the treatment of HCC.

Peng, Mengqi;Li, Hengzhao;Qin, Zixuan;Li, Junyu;Sun, Yanhao;Zhang, Xiaoxu;Jiang, Ling;Do, Hainam; An, Jie research published 《 Pentafluorophenyl Group as Activating Group: Synthesis of α-Deuterio Carboxylic Acid Derivatives via Et3N Catalyzed H/D Exchange》, the research content is summarized as follows. Discovered that highly regioselective H/D exchange was achieved on the α-position of the pentafluorophenyl (Pfp) ester using Et3N as the catalyst and D2O as the deuterium source. Computational and exptl. results showed that the Pfp group can significantly increase the acidity of the α-hydrogen of the ester. This mild reaction condition tolerated a variety of functional groups. 90%∼98% yields and 91%∼98% deuterium incorporations were obtained with all the tested Pfp esters. Taking advantage of the high reactivity of Pfp esters, a series of valuable α-deuterio carboxylic acid derivatives, including herbicide, drugs and drug intermediates, was synthesized using α-deuterio Pfp esters as precursors.
