Myeloid differentiation protein 2 (MD2) is a co-receptor of toll-like receptor 4 (TLR4) responsible for the recognition of lipopolysaccharide (LPS) and mediates a series of TLR4-dependent inflammatory responses in inflammatory lung diseases including acute lung injury (ALI). Targeting MD2 thus may provide a therapeutic strategy against these lung diseases. In this study, we identified a novel compound 4k with the potent anti-inflammatory activity among 39 methyl gallate derivatives (MGDs). MGD 4k exhibited a high binding affinity to MD2, which in turn prevented the formation of the LPS/MD2/TLR4 complex. In addition, MGD 4k significantly reversed the upregulation of LPS-induced inflammatory mediators such as tumor necrosis factor-alpha, interleukin-6, intracellular adhesion molecule-1, vascular cell adhesion molecule-1, and monocyte chemoattractant protein-1 in vitro and in vivo. Mechanistically, MGD 4k performed anti-inflammatory function by inactivating JNK, ERK and p38 signaling pathways. Taken together, our study identified MGD 4k as a novel potential therapeutic agent for ALI through inhibiting MD2, inflammatory responses, and major inflammation-associated signaling pathways.

Background:Licensed therapies for nonalcoholic fatty liver disease (NAFLD) do not yet exist, but clinical trials are testing treatment options. Inclusion criteria often require liver biopsy showing fibrosis (F2/3) or cirrhosis (F4) and nonalcoholic steatohepatitis (NASH). However, histological criteria pose a serious obstacle for recruitment. Aims:Characterize the relevance of liver biopsies in the selection of patients with NAFLD. Methods:Patients between 2013 and 2018 with the ICD-10 code K76.0 were analyzed. Fibrosis was defined by the NASH clinical research network (CRN) fibrosis staging system, NASH by a NAFLD activity score (NAS) >= 4. Predictive factors were determined by logistic regression. Results:Liver biopsy was performed in 87/638 (13.6%) patients (49% female, age 52.5 +/- 14.0, BMI 30.4 +/- 5.9 kg/m(2)). Fibrosis stage F0/F1/F2/F3/F4 was observed inN = 7/47/7/17/9, an NAS >= 4 inN = 27. Fibrosis stage F2/F3 and F4 along with NAS >= 4 was found in 1.7% and 0.5% of cases. Liver stiffness measurement, LSM (OR 2.3 per doubling of value; CI 1.3-4.4,p = .005) and FIB-4 (OR 2.3 per doubling of value; CI 1.2-4.4,p = .012) were significant predictors for fibrosis >= F2. Predictive factors for NASH were not identified. Conclusion:The biopsy rate in NAFLD patients is low and fibrosis >= F2 along with NAS >= 4 only present in a few cases. Transient elastography and FIB-4 are useful to select patients at risk for fibrosis for liver biopsy.

Graphene oxide (GO)-catalyzed selective synthesis of amides via C-Sp3-H activation of methylarenes and consequent C-N bond formation with anilines under aqueous medium has been described. Oxygen functionality allied with GO surface played a dual role both as acid catalyst and oxidizing agent to some extent. However, GO has a copious effect on the reaction, shown by a high TOF value with TBHP as co-oxidant. The decisive role of carboxylic acid functional groups on GO nanosheets in this metal-free strategy has been confirmed and was monitored by various analytic techniques viz. Fourier transform-infrared, UV-Vis, Raman and XPS. A plausible mechanism was proposed by control experiments and by the isolation of the intermediate. Over-oxidation of methylarenes was not detected, and high recyclability of the carbocatalyst with its heterogeneous behavior facilitated the isolation and purification of the desired products. We have further explored the utility of this process for the chemoselective synthesis of benzimidazoles.

Adsorption of a diverse set of chemicals onto goethite was evaluated by column chromatography. The pH of the effluents was 4.7-5.2. Van der Waals forces dominate the exothermic adsorption of 8 nonpolar compounds (e.g., PAHs and chlorobenzenes). H-bonding is responsible for the adsorption of 32 mono-carboxylic acids (i.e., benzoic acids, naphthoic acids and acidic pharmaceuticals) and their adsorption tends to be endothermic. Steric effects significantly decreased the bonding of monocarboxylic acids with ortho-substitutions. Exothermic adsorption of 10 monophenols is controlled by weak H-bonding. Bonding of these 50 solutes onto goethite is totally reversible. In contrast, inner-sphere complexation of phthalic acid and chlortetracycline with goethite occurred according to their low desorption ratio (1.1%-54.4%). Polyparameter linear free energy relationship (PP-LFER) models were established to provide acceptable fitting results of the goethite-solute distribution coefficients (RMSE = 0.32 and 0.30 at 25 degrees C and 5 degrees C, respectively). Hangzhou, one the most prosperous cities in China, suffers from severe atmospheric quality degradation in recent years. For the good of nine million local citizens and incoming Asian Games, Recurrent Neural Network (RNN) and Random Forest are used to analyze the air pollution in Hangzhou. Compared with the traditional atmospheric models, machine learning models are faster, more accurate and less costly in simulating all the pollutants without using the pollution inventory. The Feature Importance (Fl) generated by Random Forest reveals the complicated relationships among air pollutants and meteorology. Carbon monoxide (CO) plays an important role in shaping ground-level ozone, nitrogen dioxide (NO2) and particulate matters (PM) in the atmospheric environment. Dew-point deficit plays a more important role than relative humidity in shaping air pollutants. Urban heat island effect is not obvious for the air pollutants from non-point source. Furthermore, a WRF/RNN-based method to forecast air pollutants, including SO2, NO2, CO, PM2.5, PM10 and O-3, in the future 24 h is proposed and a RNN-based method to estimate regional transport rate of air pollutants and reversely identify air pollution emission sources is introduced. Most microplastics (MPs) have undergone extensive aging in the environment. Aged MPs exhibit different physical and chemical properties from unaged ones. Here, we studied the effects of accelerated aging on the characteristics and pyrolysis of commercial pigmented MPs, as well as pigments leaching and their interactions in simulated gastric and intestinal fluids of mammals. We report that the carbonyl index, surface area, and color change of MPs increased after aging treatment. Cracks and fragmentation of MPs facilitated the accessibility of light and oxygen to internal layer and therefore accelerated the aging process. TGA/GC-MS analysis showed that the high temperature resistance of MN decreased after aging. Thermal decomposition of pigments and polyethylene occurred in temperature ranges of 340-406 degrees C and 406-550 degrees C, respectively. Mono (di)-alkenes and saturated alkanes were the thermal decomposition products of polyethylene. Aging of MPs also caused an increased release of pigments and prolonged aging time led to more release in simulated fluids. Pigments would result in fluorescence quenching of the enzymes through binding interactions once they were released from MPs into simulated fluids. Charge neutralization and polymer bridging accounted for the formation of pigment-enzyme complexes and flocs. These novel findings will allow us to better assess how aging process affects the characteristics, leaching, and toxicity of MPs. (C) 2020 Published by Elsevier B.V.

Background The side reactions of dehalogenation or C-N coupling tend to occur when halogenated aromatic amines are prepared by catalytic hydrogenation reduction of halogenated aromatic nitro compounds. In this paper, we prepared the sub-microspherical Fe3O4@PDA-Pd NPs catalyst apply it efficiently in the hydrogenation reduction of halogenated aromatic nitro compounds to prepare the halogenated aromatic amines under atmospheric pressure. The catalyst shows a high selectivity of greater than 96% and can effectively inhibit the occurrence of the side reactions of dehalogenation and C-N coupling. Results The optimum condition of the hydroreduction reaction is when tetrahydrofuran is used as solvent and the reaction happens at 50 degrees C for 5 h. The selectivity of the chlorinated aromatic amine and the fluorinated aromatic amine products exceed 99% and the yield exceeds 90%. Only a small amount of dehalogenated products and C-N coupling by-products were produced in the brominated aromatic compound and the iodinated aromatic compound. Conclusion We developed a promising method for preparing the superparamagnetic and strongly magnetic Fe3O4@PDA core-shell sub-microsphere-supported nano-palladium catalyst for catalyzing the hydrogenation reduction of halogenated aromatic nitro compounds. The halogenated aromatic amines were efficiently and highly selectively prepared under atmospheric pressure, with the side reactions of dehalogenation and C-N coupling effectively inhabited simultaneously.

Organic air pollutants have considerable influence particularly on atmospheric chemistry, compared to other air pollutants. Traffic is one of the major sources of air pollutants occurred in Canakkale atmosphere by both remarkable road and marine traffic loads around the harbors. The aims of this study were finding the daily, week/weekend, and spatial variations of VOC at multiple sampling points of Canakkale and Kilitbahir. In this study, active air samples were collected from the major roads of Kilitbahir and Canakkale Harbors during rush hours and off-peak in 2018 summer. According to the measurement results, average levels of total volatile organic compounds (TVOC) were around 129 mu g/m(3) in Canakkale and 93 mu g/m(3) in Kilitbahir. Toluene was the most abundant compound. Average B: T: E: X ratio was calculated as 2.4:7.0:1:5.5 in Canakkale and 3.2:7.6:1:5.2 in Kilitbahir. According to the sampling points of both locations, levels of TVOC, benzene, toluene, and m,p-xylene in Canakkale and toluene levels in Kilitbahir showed statistically significant differences (p < 0.05). Also, benzene, which was already classified as a human carcinogen, varied diurnally in Kilitbahir (p < 0.05). Toluene levels varied between week and weekend days (p < 0.05). Moreover, measured VOC levels were compared with air quality monitoring station (AQMS) data and meteorological parameters. TVOC levels showed positive correlations with SO2 and PM10 (p < 0.05). Furthermore, a multivariate factor analysis applied to data set including VOC measurements, AQMS data, and meteorological parameters implied that traffic is the predominant factor influencing the air quality around the study sites. Heavy metal pollution in lakes is becoming increasingly of interest to researchers. Because heavy metals have high mobility and do not degrade, they migrate easily between different environmental mediums through processes such as suspended solids deposition, sediment resuspension, and diffusion, among others. These processes are particularly pronounced in shallow lakes since the hydrodynamism is higher in bodies of water with minimal depth. Lake Ulansuhai a typical shallow lake in the Hetao irrigation district in Inner Mongolia-also experiences intense sandstorm activity, which compounds the suspended solids exchange intensity between water and sediment, strengthening the migration of heavy metals in the lake system. This study examines the fate and transport of two heavy metals-Zn and Pb-within this lake, using a field experiment to determine the flux of sediment re-suspension and deposition and a laboratory experiment to modify the QWASI model for shallow bodies of water. The aguivalence and mass balance approaches were used to develop this modified QWASI model. The margins of error between the modeled and the measured average concentrations of Zn and Pb in water were 5.0%-30.6% and 5.8%-29.5%, respectively, and in sediment were 0.3%-4.9% and 0.9-5.5%, respectively. These results suggest that the modified QWASI model developed here could indeed be used to more accurately represent the fate and transport of Zn and Pb during the icefree period of a shallow lake. Electrochemical reduction-oxidation processes with the aid of cathode catalysts are promising technologies for the decomposition of organic compounds. High-efficiency and low-cost catalysts for electrochemical reductive dechlorination and two-electron oxygen reduction reaction (ORR) are vital to the overall degradation of chlorinated organic compounds. This study reports electrochemical dechlorination using a single-atom Co-loaded sulfide graphene (Co-SG) catalyst via atomic hydrogen generated from the electrochemical reduction of H2O and electrolysis of hydrogen. The Co-SG electrocatalyst exhibited a remarkable performance for H2O2 synthesis with a half-wave potential of 0.70 V (vs RHE) and selectivity over 90%. The high electrochemical performance was achieved for bifunctional electrocatalysis with regard to the smaller overpotentials, faster kinetics, and higher cycling stability compared to the noble metal-based electrocatalysts. In this study, 2,4-dichlorobenzoic acid was well degraded and the TOC concentration was effectively reduced. This work introduces the preparation of a new active site for high-performance single-atom catalysts and also promotes its application in the electrochemical degradation of chlorinated organic pollutants.
