Carnitine, after extraction by appropriate solvents or liberation from biol. materials by acid hydrolysis, is separated electrophoretically. Separation is carried out on Whatman 3 MM paper at 400 v. by means of buffer prepared by adding pyridine to 0.3M AcOH to give a final pH of 4.1. Position of the carnitine spot is located with I vapor; the spot is cut out and eluted with HCl-EtOH, and carnitine is estimated by the method of Friedman (CA 52, 12978a) in which the ethyl ester is complexed in alk. medium with bromophenol blue. The procedure gives an error of the order of 10%. Application of the method to several betaines and the results are described.
