Retentions of selected solute groups were measured in 4 mobile phases of various elution strength, prepared from MeOH-H2O mixtures at various ratios. Dependence of k‘ on the length of the “bristle”, on the size and structure of the solute mol., and the elution strength of the mobile phase were plotted. Results are compared for pairs of phases having different structures but either the same total number of C atoms in the bonded mol. or the same length of the “bristle”. The synthesis of several halosilanes used for preparation of the bonded phases is described.

We previously reported the phenylchloronitrobenzamides (PCNBs), a novel class of compounds active against the species of trypanosomes that cause Human African Trypanosomiasis (HAT). Herein, we explored the potential to adjust the reactivity of the electrophilic chloronitrobenzamide core. These studies identified compound 7d that potently inhibited the growth of trypanosomes (EC50 = 120 nM for Trypanosoma b. brucei, 18 nM for Trypanosoma b. rhodesiense, and 38 nM for Trypanosoma b. gambiense) without significant cytotoxicity against mammalian cell lines (EC50 > 25 μM for HepG2, HEK293, Raji, and BJ cell lines) and also had good stability in microsomal models (t1/2 > 4 h in both human and mouse). Overall these properties indicate the compound 7d and its analogs are worth further exploration as potential leads for HAT.

The phytohormone auxin is transported through the plant body either via vascular pathways or from cell to cell by specialized polar transport machinery. This machinery consists of a balanced system of passive diffusion combined with the activities of auxin influx and efflux carriers. Synthetic auxins that differ in the mechanisms of their transport across the plasma membrane together with polar auxin transport inhibitors have been used in many studies on particular auxin carriers and their role in plant development. However, the exact mechanism of action of auxin efflux and influx inhibitors has not been fully elucidated. In this report, the mechanism of action of the auxin influx inhibitors (1-naphthoxyacetic acid (1-NOA), 2-naphthoxyacetic acid (2-NOA), and 3-chloro-4-hydroxyphenylacetic acid (CHPAA)) is examined by direct measurements of auxin accumulation, cellular phenotypic anal., as well as by localization studies of Arabidopsis thaliana auxin carriers heterologously expressed in Nicotiana tabacum, cv. Bright Yellow cell suspensions. The mode of action of 1-NOA, 2-NOA, and CHPAA has been shown to be linked with the dynamics of the plasma membrane. The most potent inhibitor, 1-NOA, blocked the activities of both auxin influx and efflux carriers, whereas 2-NOA and CHPAA at the same concentration preferentially inhibited auxin influx. The results suggest that these, previously unknown, activities of putative auxin influx inhibitors regulate overall auxin transport across the plasma membrane depending on the dynamics of particular membrane vesicles.

DNA damage repair enzymes are promising targets in the development of new therapeutic agents for a wide range of cancers and potentially other diseases. The enzyme poly(ADP-ribose) glycohydrolase (PARG) plays a pivotal role in the regulation of DNA repair mechanisms; however, the lack of potent drug-like inhibitors for use in cellular and in vivo models has limited the investigation of its potential as a novel therapeutic target. Using the crystal structure of human PARG in complex with the weakly active and cytotoxic anthraquinone 8a, novel quinazolinedione sulfonamides PARG inhibitors have been identified by means of structure-based virtual screening and library design. 1-Oxetan-3-ylmethyl derivatives 33d and 35d were selected for preliminary investigations in vivo. X-ray crystal structures help rationalize the observed structure-activity relationships of these novel inhibitors.

However, AOP did increase the amount of easily biol. degradable compounds, but these were diluted with non-AOP treated infltration water and degraded during transport to the dunes, making the effects on MARR negligible. Several OMP that were not removed during AOP, were removed during MARR. The overall OMP concentrations in resulting drinking water therefore decreased after introduction of AOP, without showing a measurable neg. effect on other water quality parameters.

Redox-switchable hemilabile ligands (RHLs) were synthesized that incorporates ferrocene as the redox group and phosphine ether or phosphine thioether moieties as binding groups. These ligands, which complex to Rh(I) and Pd(II), yield electrochem. control over ligand binding affinity for transition metals in complexes of the following type: [M(η4-(η5-C5H4XCH2CH2PR2)2Fe)]y+ (I: M = Rh, X = O, R = Ph (phenyl), y = 1; II: M = Rh, X = O, R = Cy (cyclohexyl), y = 1; III: M = Rh, X = S, R = Ph, y = 1; IV: M = Pd, X = O, R = Ph, y = 2; V: M = Pd, X = O, R = Cy, y = 2). In the case of V, ligand based oxidation decreases the ligand to metal binding constant by nearly ten orders of magnitude. An examination of the crystal structures of I, III, IV, and V and the electrochem. behavior of RHL-complexes and isoelectronic model complexes reveals that electrostatic effects play a significant role in the charge dependent behaviors of these complexes. Addnl., there is a correlation between the phosphine substituents and RHL-complex stability. As a general rule cyclohexyl groups stabilize the complexes in their oxidized states over Ph groups. RHLs provide a viable means of electrochem. controlling ligand binding affinity and thus the steric and electronic environment of bound transition metals.

Since the computer industry enables one to generate smaller and smaller structures, silicon surface chem. is becoming increasingly important for (bio-)anal. and biol. applications. For controlling the binding of charged biomacromols. such as DNA and proteins on modified silicon surfaces, the surface pKa is an important factor. Here the authors present a fluorescent nanoparticle adhesion assay as a novel method to determine the surface pKa of silicon surfaces modified with weak acids or bases. This method is based upon electrostatic interactions between the modified silicon surface and fluorescent nanoparticles with an opposite charge. Silicon slides were modified with 3-aminopropyltriethoxysilane (APTES) and were further derivatized with succinic anhydride. Layer thickness of these surfaces was determined by ellipsometry. After incubating the surfaces with an amine-reactive fluorescent dye, fluorescence microscopy revealed that the silicon surfaces were successfully modified with amine- and carboxyl-groups. Two surface pKa values were found for APTES surfaces by the fluorescent nanoparticle adhesion assay. The 1st surface pKa (6.55 ± 0.73) was comparable with the surface pKa obtained by contact angle titration (7.3 ± 0.8), and the 2nd surface pKa (9.94 ± 0.19) was only found by using the fluorescent nanoparticle adhesion assay. The surface pKa of the carboxyl-modified surface by the fluorescent nanoparticle adhesion assay (4.37 ± 0.59) did not significantly differ from that found by contact angle titration (5.7 ± 1.4). In conclusion, the authors have developed a novel method to determine the surface pKa of modified silicon surfaces: the fluorescent nanoparticle adhesion assay. This method may provide a useful tool for designing pH-dependent electrostatic protein and particle binding/release and to design surfaces with a pH-dependent surface charge for (bio-)anal. lab-on-a-chip devices or drug delivery purposes.

A series of N-(2-arylmethylthio-4-chloro-5-methylbenzenesulfonyl)amides I [R = Ph, 3-F3CC6H4, 4–F3CC6H4, 1-naphthyl, 2-naphthyl] and II was synthesized. The antimicrobial activity of compounds I and II was evaluated on Gram-pos., Gram-neg. bacteria and fungal species. Experiments took into account investigation of antibacterial activity against planktonic cells as well as biofilms. Compounds I and II showed high bacteriostatic activity against staphylococci, with the most active mols. I [R = 3-F3CC6H4] and II [R = 3-F3CC6H4] presenting low MIC values of 4-8μg/mL against reference methicillin-resistant S. aureus (MRSA) and methicillin-sensitive S. aureus (MSSA) strains as well as clin. isolates. Compounds I [R = 3-F3CC6H4] and II [R = 3-F3CC6H4] also showed an ability to inhibit biofilms formed by MRSA and MSSA. The potential of I [R = 3-F3CC6H4] and II [R = 3-F3CC6H4] as antibiofilm agents was supported by cytotoxicity assays that indicated no cytotoxic effect either on normal cells of human keratinocytes or on human cancer cells, including cervical, colon, and breast cancer lines.
