Various enantiomeric and geometric oxime O-(α- and β-methylcholinyl) ethers were prepared as potential anticholinergic agents. The synthesis, separation, resolution, and structural characterization of these compounds are reported. The 1st step of the synthetic pathway involved an oxime formation, with subsequent O-alkylation of the resp. oxime with 2-chloro-N,N-dimethylpropylamine hydrochloride. The separation of the α- and β-structural isomers utilized vacuum fractional distillation and/or column chromatog., and the resolution of the enantiomers was accomplished via the formation of tartrate diastereoisomers. A preliminary pharmacol. evaluation for anticholinergic activity was conducted using a rat ileum assay. Structure-activity relationships, including some stereochem. properties and antimuscarinic activity, are discussed.

During inflammation, neutrophil- and monocyte-derived myeloperoxidase catalyzes the formation of hypochlorous acid, which can chlorinate tyrosine residues in proteins to form chlorotyrosine. However, little is known of the metabolism and disposition of chlorotyrosine in vivo. Following infusion of deuterium-labeled [D4]chlorotyrosine into Sprague-Dawley rats, the major urinary metabolites were identified by mass spectrometry. 3-Chloro-4-hydroxyphenylacetic acid was identified as the major chlorinated metabolite of chlorotyrosine and accounted for 3.6 ± 0.3% of infused [D4]chlorotyrosine. The striking observation was that ∼40% (39 ± 1%) of infused [D4]chlorotyrosine was dechlorinated and excreted in the urine as deuterated 4-hydroxyphenylacetic acid, a major metabolite of tyrosine. 1.1 ± 0.1% of infused [D4]chlorotyrosine was excreted as [D4]tyrosine. To determine whether protein-bound chlorotyrosine could undergo dechlorination, chlorinated albumin was incubated with liver homogenate from mutant rats, which did not synthesize albumin. There was ∼20% decrease in the chlorotyrosine content over 1 h. This study is the first to describe the dechlorination of chlorotyrosine as the major metabolic pathway to eliminate this modified amino acid in vivo.

A homologous series of poly(n-alkylmethylsiloxanes), [SiMe(R)O]n, were prepared by cationic ring-opening polymerization (ROP) of cyclotrisiloxanes containing n-alkyl substituents, such as C2H5, n-C3H7, n-C4H9, n-C5H11, and n-C6H13, on the Si. The resultant polymers were characterized by NMR, GPC, and DSC measurements. All of these asym. substituted polymers were obtained in high mol. weight form and were found to have an atactic configuration. Their glass transitions were found to increase smoothly from Et to n-hexyl (i.e., -139, -120, -115, -111.9, and -108 °C, for Et to n-hexyl, resp.). These Tgs are much lower than those of the corresponding polyolefins, [C(H)(R)CH2]n. Moreover, in the case of these polyolefins, the Tgs decrease continuously from R = Me to n-hexyl. The calculated Tgs for the polysiloxanes that were obtained by using published empirical equations show the same general trend as the exptl. values.

We have investigated the enzyme inhibition characteristics of a natural product (NP)-based phenolic library against a panel of human carbonic anhydrases (hCAs, EC 4.2.1.1) which included hCAs I and II (cytosolic) and hCA VA/VB (mitochondrial isoforms). Most of these compounds were weak, micromolar inhibitors of the two cytosolic hCAs (K Is >10 μM) but showed good hCA VA/VB inhibitory activity with inhibition constants in the range of 70-125 nM. The selectivity ratios for inhibiting the mitochondrial over the cytosolic isoforms for these phenol derivatives were in the range of 120-3800, making them the most isoform-selective compounds for inhibiting hCA VA/VB known to date. The CA VA/VB enzymes are involved in biosynthetic processes such as gluconeogenesis, lipogenesis and ureagenesis, and no pharmacol. inhibitors with good selectivity are currently available. Thus the NP inhibitors identified during these studies are excellent leads for obtaining even more effective compounds that selectively target mitochondrial hCAs, and also have the potential to be used as tools for understanding the physiol. processes that are regulated by the two mitochondrial CA isoforms.

Diamide I, prepared by treatment of 3-methylglutaric acid with 4(R)-(methoxycarbonyl)-1,3-thiazolidine-2-thione [4(R)-MCTT] in the presence of DCC in pyridine, was subjected to aminolysis with 1 equiv of piperidine in CH2Cl2 at -30° to give a mixture of diastereomers II [R2N = piperidino, R12N = MCTT moiety (III)] and II (R2N = MCTT moiety, R12N = piperidino) in a 88:12 ratio. III was separated by silica gel column chromatog. and treated with various nucleophiles to give optically pure bifunctional synthons (II; R2N and R12N = nucleophilic group) in high yields. Highly selective chiral induction into cis-4-cyclohexen-1,2-ylenebis(acetic acid) was also performed. Aminolysis of IV (R2N and R12N = MCTT moiety) (V) with 1 equiv of piperidine gave 94% diastereomer IV (R2N = piperidino, R12N = MCTT moiety). Similar chiral induction into cis-cyclohexan-1,2-xylenebis(acetic acid) and aminolysis of its 4(R)-MCTT diamide (VI) with piperidine were also carried out; opposite selectivity was obtained. The conformations of V and VI in a solvent, the relationship between the susceptibility of their conformations and environmental temperature, and the diastereoselectivity of the reaction are discussed on the basis of the 400-MHz 1H NMR spectra.

A flow apparatus for microwave irradiation is constructed; the apparatus is used for the reactions of (ethoxymethylene)malononitrile with hydrazines RNHNH2 (R = H, Me, Ph, 4-ClC6H4, 4-O2NC6H4, 2-F3CC6H4, 3-Cl-4-MeC6H3, 2,5-Cl2C6H3, 4-FC6H4, 4-MeOC6H4, 4-HO2CC6H4, 3-Cl-4-FC6H3, 2-Cl-5-F3CC6H3, 2-pyridinyl) under microwave irradiation to give aminopyrazolecarbonitriles I (R = H, Me, Ph, 4-ClC6H4, 4-O2NC6H4, 2-F3CC6H4, 3-Cl-4-MeC6H3, 2,5-Cl2C6H3, 4-FC6H4, 4-MeOC6H4, 4-HO2CC6H4, 3-Cl-4-FC6H3, 2-Cl-5-F3CC6H3, 2-pyridinyl) in 62-96% yields and in 90->95% purities (all but one in >95% purity). Microwave irradiation of hydrazines RNHNH2 (R = H, Me, Ph, 4-ClC6H4, 4-O2NC6H4, 2-F3CC6H4, 3-Cl-4-MeC6H3, 2,5-Cl2C6H3, 4-FC6H4, 4-MeOC6H4, 4-HO2CC6H4, 3-Cl-4-FC6H3, 2-Cl-5-F3CC6H3, 2-pyridinyl) with (ethoxymethylene)malononitrile using the described flow apparatus followed by flow of the mixture through a column of resin-bound benzylamine and a column of activated carbon provides I (R = H, Me, Ph, 4-ClC6H4, 4-O2NC6H4, 2-F3CC6H4, 3-Cl-4-MeC6H3, 2,5-Cl2C6H3, 4-FC6H4, 4-MeOC6H4, 4-HO2CC6H4, 3-Cl-4-FC6H3, 2-Cl-5-F3CC6H3, 2-pyridinyl) in 62-91% yields and >95% purities; I (R = 4-HO2CC6H4) could not be purified using the resin-bound benzylamine column and is isolated in 96% yield and 90% purity. This method is used to prepare >250 g of I (R = 4-HO2CC6H4). I (R = Me, Ph, 4-ClC6H4, 2-F3CC6H4, 3-Cl-4-MeC6H3, 2,5-Cl2C6H3, 4-FC6H4, 4-MeOC6H4, 2-Cl-5-F3CC6H3, 2-pyridinyl) undergo cyclocondensation and dimerization under microwave irradiation with potassium tert-butoxide in toluene at 160° to give the pyrazolylpyrazolopyrimidines II (R = Me, Ph, 4-ClC6H4, 2-F3CC6H4, 3-Cl-4-MeC6H3, 2,5-Cl2C6H3, 4-FC6H4, 4-MeOC6H4, 2-Cl-5-F3CC6H3, 2-pyridinyl) in 28-82% yields; the products are purified by stirring with Amberlyst 15 in DMF followed by stirring of the resin with triethylamine in DMF. I (R = H) reacts with nitriles R1CN (R1 = Ph, 3-BrC6H4, 2-pyridinyl) under microwave irradiation with potassium tert-butoxide in toluene at 160° to give pyrazolopyrimidines III (R1 = Ph, 3-BrC6H4, 2-pyridinyl) in 88-94% yields.
