Aiming at generating a series of monoterpene indole alkaloids with enhanced multidrug resistance (MDR) reversing activity in cancer, two major epimeric alkaloids isolated from Tabernaemontana elegans, tabernaemontanine and dregamine, were derivatized by alkylation of the indole nitrogen. Twenty-six new derivatives were prepared by reaction with different aliphatic and aromatic halides, whose structures were elucidated mainly by NMR, including 2D NMR experiments Their MDR reversal ability was evaluated through a functional assay, using as models resistant human colon adenocarcinoma and human ABCB1-gene transfected L5178Y mouse lymphoma cells, overexpressing P-glycoprotein (P-gp), by flow cytometry. A considerable increase of activity was found for most of the derivatives, being the strongest P-gp inhibitors those sharing N-phenethyl moieties, displaying outstanding inhibitory activity, associated with weak cytotoxicity. Chemosensitivity assays were also performed in a model of combination chemotherapy in the same cell lines, by studying the in vitro interactions between the compounds and the antineoplastic drug doxorubicin. Most of the compounds have shown strong synergistic interactions with doxorubicin, highlighting their potential as MDR reversers. QSAR models were also explored for insights on drug-receptor interaction, and it was found that lipophilicity and bulkiness features were associated with inhibitory activity, although linear correlations were not observed

As an extension of our anal. of the effect of halogenation on thiourea TRPV1 agonists, we have now modified selected 4-hydroxy(or 4-amino)-3-methoxyphenyl acetamide TRPV1 agonists by 5- or 6-halogenation on the aromatic A-region and evaluated them for potency for TRPV1 binding and regulation and for their pattern of agonism/antagonism (efficacy). Halogenation shifted the functional activity at TRPV1 toward antagonism with a greater extent of antagonism as the size of the halogen increased (I > Br > Cl), as previously observed for the thiourea series. The extent of antagonism was greater for halogenation at the 5-position than at the 6-position, in contrast to SAR for the thiourea series. In this series, compounds 55 (I) and 75 showed the most potent antagonism, with K i (ant) = 2.77 and 2.19 nM, resp., on rTRPV1 expressed in Chinese hamster ovary cells. The compounds were thus ca. 40-60-fold more potent than 6′-iodononivamide.

A reactive disperse azo dye having an α-bromoacrylamido group was synthesized and its dyeing and fastness properties were compared with those of a a disperse dye. The amount of absorbed reactive disperse dye was less on polyester fabric than the disperse dye, and was much higher on nylon fabric. When polyester and nylon fiber were simultaneously dyed, the nylon absorbed more reactive disperse dye than polyester. The use of a carrier or a variation of dyebath pH was not able to provide the same color depth on both fibers. A nylon/polyester union fabric could be dyed with the reactive disperse dye and the color fastness was good to excellent.

Cholestyramine is the most widely used bile acid sequestrant in the treatment of hypercholesterolemia. However, cholestyramine has unpleasant side effects as a consequence of its hydrophobic backbone. Therefore, high-capacity bile acid sequestering biopolymers with cationic chitosan derivatives were developed, because electrostatic interactions are important for binding with bile acid anions. Dialkylaminoalkylation and reductive amination of chitosan were done to add dialkylaminoalkyl and an addnl. free amino group at a hydroxyl site in the chitosan backbone resp. and the aminoderivatized chitosan derivatives were quaternized with Me iodide to produce a cationic polyelectrolyte. The in vitro bile acid binding capacity of the chitosan derivatives in aqueous NaCl was measured by reversed-phase HPLC. The binding capacities of sodium glycocholate (a major bile acid) to chitosan, DEAE-chitosan, quaternized DEAE-chitosan, and cholestyramine were 1.42, 3.12, 4.06, and 2.78 mmol/g resin, resp. With quaternized DEAE-chitosan, the bile acid binding capacity increased ∼50% over that of cholestyramine. The bile acid binding capacity of dialkylaminoalkyl chitosan derivatives increased with the number of carbons in the alkyl groups, indicating that hydrophobic interaction is a secondary factor for the sequestration of bile acids.

Trialkyl and triaryl organoboranes undergo multiple, repetitive homologations upon reaction with dimethylsulfoxonium methylide (1). This multiple homologation reaction, or polyhomologation, produces polymethylene in a living reaction. Applying the polyhomologation reaction to cyclic and polycyclic organoboranes permits the construction of unique oligomeric and polymeric architectures that are not readily accessible by standard olefin polymerization The polyhomologation of 1-boraadamantane·THF (2) by ylide 1 generates novel macrotricyclic trialkylboranes (3). The oxidation of these macrocyclic organoboranes generates a three-armed star polymethylene polymer (4) incorporating a cis,cis-1,3,5-trisubstituted cyclohexane core. Interestingly, only one-third of the initiators lead to product formation, resulting in an observed d.p. 3 times higher than expected. Close examination of the initial stages of polymerization show that 1-boraadamantane·THF reacts with 1 equiv of 1 to afford a monohomologated product. Subsequent homologations were found to contain branch points leading to isomeric tricyclic products after the third, fourth, and fifth methylene insertions. At these stages of homologation, all of the propagating species result in tricyclic trialkylborane cages with collapsed, inverted pyramidal boron centers that are substantially less reactive toward ylide. Approx. two-thirds of the species discontinue polymerization at these stages. However, one-third of these species continue to propagate and eventually result in the formation of giant macrotricyclic polymers of narrow polydispersity. Mol. modeling and kinetic simulation have aided in the anal. of the probable pathways through which the reaction proceeds.

Also, their resist effectiveness was improved by increasing the number of sulfonate groups in the dye-resist agents and the reactive groups in the reactive dyes applied over them.
