It is established that the interaction with these cationic surfactants changes the spectral characteristics of the flavonoids in solutions and essentially increases their adsorption on the surface of highly dispersed silica as compared with their aqueous solutions It is shown that the efficiency of flavonoid adsorption from decamethoxine and miramistin solutions is governed by solution pH, flavonoid hydrophobicity, and the nature of a cationic surfactant.

We describe new synthetic routes developed toward a range of substituted analogs of bromo and extra-terminal (BET) bromodomain inhibitors I-BET762/JQ1 based on the triazolo-benzodiazepine scaffold. These new routes allow for the derivatization of the methoxyphenyl and chlorophenyl rings, in addition to the diazepine ternary center and the side chain methylene moiety. Substitution at the level of the side chain methylene afforded compounds targeting specifically and potently engineered BET bromodomains designed as part of a bump and hole approach. We further demonstrate that marked selectivity for the second over the first bromodomain can be achieved with an indole derivative that exploits differential interaction with an aspartate/histidine conservative substitution on the BC loop of BET bromodomains.

Na2SeSO3, which can be generated in situ by the reaction of Na2SO3 with Se power, was found to be an odorless reagent for the selenenylation of alkyl halides RX (R = Bu, cyclohexyl, 3-fluorobenzyl, etc.; X = Cl, Br) to produce dialkyl diselenides RSeSeR. By using aqueous EtOH as the solvent and avoiding the generation of a malodorous selenol intermediate, the selenylation reaction with Na2SeSO3 is much more environmentally friendly than conventional methods. Owing to the cheap and abundant starting materials and selenium reagents, the novel synthetic method reduces the production costs of dialkyl diselenides as organoselenium catalysts, thereby advancing practical applications of organoselenium-catalysis technologies.

To develop novel CDK2 inhibitors as anticancer agents, a series of novel pyrimidine-based benzothiazole derivatives I (R = 4-(S(O)2NH2), 4-(piperidin-1-yl), 3-F, etc.) and II (R1 = H, F, Cl, Me, MeO; R2 = H, Me, F; R3 = Me, NH2) was designed and synthesized. Initial biol. evaluation demonstrated that some of target compounds displayed potent antitumor activity in vitro against five cancer cell lines. Especially, the analog II (R1 = F; R2 = H; R3 = NH2) exhibited approx. potency with AZD5438 toward four cells including HeLa, HCT116, PC-3, and MDA-MB-231 with IC50 values of 0.45, 0.70, 0.92, 1.80 μM, resp. More interestingly, the most highly active compound II (R1 = F; R2 = H; R3 = NH2) in this study also possessed promising CDK2/cyclin A2 inhibitory activities with IC50 values of 15.4 nM, which was almost 3-fold potent than pos. control AZD5438, and mol. docking studies revealed that the analog bound efficiently with the CDK2 binding site. Further studies indicated that compound II (R1 = F; R2 = H; R3 = NH2) could induce cell cycle arrest and apoptosis in a concentration-dependent manner. These observations suggest that pyrimidine-benzothiazole hybrids represent a new class of CDK2 inhibitors and well worth further investigation aiming to generate potential anticancer agents.

Structure-based macrocyclization of a 6-carboxylic acid indole chemotype has yielded potent and selective finger-loop inhibitors of the hepatitis C virus (HCV) NS5B polymerase. Lead optimization in conjunction with in vivo evaluation in rats identified several compounds showing (i) nanomolar potency in HCV replicon cells, (ii) limited toxicity and off-target activities, and (iii) encouraging preclin. pharmacokinetic profiles characterized by high liver distribution. This effort culminated in the identification of TMC647055 (I), a nonzwitterionic 17-membered-ring macrocycle characterized by high affinity, long polymerase residence time, and broad genotypic coverage. In vitro results of the combination of I with the HCV protease inhibitor TMC435 (simeprevir) supported an evaluation of this combination in patients with regard to virus suppression and resistance emergence. In a phase 1b trial with HCV genotype 1-infected patients, I was considered to be safe and well-tolerated and demonstrated potent antiviral activity, which was further enhanced in a combination study with TMC435.

A series of novel 2,4-disubstituted quinazolines I [R = hexyl, cyclopentyl, Ph, etc.] were synthesized and evaluated for their anti-tumor activity against five human cancer cells (MDA-MB-231, MCF-7, PC-3, HGC-27 and MGC-803) using MTT assay. Among them, compound I [R = 4-F3CC6H4] showed the most potent cytotoxicity against breast cancer cells. Compound I [R = 4-F3CC6H4] also significantly inhibited the colony formation and migration of MDA-MB-231 and MCF-7 cells. Meanwhile, compound I [R = 4-F3CC6H4] induced cell cycle arrest at G1 phase and cell apoptosis, as well as increased accumulation of intracellular ROS. Furthermore, compound I [R = 4-F3CC6H4] exerted antitumor effects in-vitro via decreasing the expression of anti-apoptotic protein Bcl-2 and increasing the pro-apoptotic protein Bax and p53. Mechanistically, compound I [R = 4-F3CC6H4] markedly decreased p-EGFR and p-PI3K expression, which revealed that compound I [R = 4-F3CC6H4] targeted breast cancer cells via interfering with EGFR-PI3K signaling pathway. Mol. docking suggested that compound I [R = 4-F3CC6H4] could indeed bind into the active pocket of EGFR. All the findings suggest that compound I [R = 4-F3CC6H4] might be a valuable lead compound for antitumor agents targeting breast cancer cells.

Reaction condition based regioselectivity has been achieved where fused benzimidazo[1,2-b][1,2,4]thiadiazines are exclusively formed under thermal conditions, whereas benzimidazo[2,1-c][1,2,4]thiadiazines were created only under microwave irradiation The salient features of this protocol include a regioselective sulfonylation of 2-aminobenzimidazole with o-halo sulfonyl chlorides followed by N-C bond formation. The acid forms of these fused regioisomers have been used to introduce novel guanidine-containing isocoumarin frameworks.
