The bactericidal activity (BA) of antimicrobial agents is generally derived from the results of killing assays. A reliable quant. characterization and particularly a comparison of these substances, however, are impossible with this information. We here propose a new method that takes into account the course of the complete killing curve for assaying BA and that allows a clear-cut quant. comparison of antimicrobial agents with only one number The new Integral Method, based on the reciprocal area below the killing curve, reliably calculates an average BA [log10 CFU/min] and, by implementation of the agent’s concentration C, the average specific bactericidal activity SBA = BA / C [log10 CFU/min/mM]. Based on exptl. killing data, the pertaining BA and SBA values of exemplary active halogen compounds were established, allowing quant. assertions. N-chlorotaurine (NCT), chloramine T (CAT), monochloramine (NH2Cl), and iodine (I2) showed extremely diverging SBA values of 0.0020 ± 0.0005, 1.11 ± 0.15, 3.49 ± 0.22, and 291 ± 137 log10 CFU/min/mM, resp., against Staphylococcus aureus. This immediately demonstrates an approx. 550-fold stronger activity of CAT, 1730-fold of NH2Cl, and 150,000-fold of I2 compared to NCT. The inferred quant. assertions and conclusions prove the new method suitable for characterizing bactericidal activity. Its application comprises the effect of defined agents on various bacteria, the consequence of temperature shifts, the influence of varying drug structure, dose-effect relationships, ranking of isosteric agents, comparison of competing com. antimicrobial formulations, and the effect of additives.

Photoactive derivatives of cellulose were prepared by a mild esterification of the biopolymer with 2-[(4-methyl-2-oxo-2H-chromen-7-yl)oxy]acetic acid via the activation of the carboxylic acid with N,N’-carbonyldiimidazole. Subsequently, modification with the cationic carboxylic acid (3-carboxypropyl)trimethylammonium chloride was carried out. Thus, water soluble polyelectrolytes decorated with high amounts of photochem. active chromene moieties were obtained. The structures of the novel polysaccharide esters and the polyelectrolytes were evaluated by means of NMR and IR spectroscopy. Moreover, the light triggered photodimerization of the chromene moieties of the photoactive polyelectrolytes was studied by means of UV-Vis spectroscopy in the dissolved state. The photochem. observed may be used to control the properties of the new polysaccharide derivatives and are thus of interest in the design of smart materials.

Xylan was isolated from birch wood chips by using pressurized hot water extraction (PHWE). The extracted xylan was chem. modified yielding three different xylan derivatives (XDs): xylan sulfate (XS), carboxymethyl xylan (CMX) and xylan-4-[N,N,N-trimethylammonium]butyrate chloride (XTMAB). The structure and mol. weight of XDs was determined by using NMR spectroscopy and size exclusion chromatog. (SEC). The potential utilization of xylan polyelectrolytes for modifying fiber surfaces was assessed by sorption experiments using bleached pine Kraft pulp as substrate. Polyelectrolyte titration method was chosen for estimating the amount of sorbed XDs onto the fibers. The cationic xylan derivative XTMAB had a strong interaction with fibers while the anionic derivatives did not show any sorption. XPS and time of flight secondary ion mass spectrometry (ToF-SIMS) were selected as advanced surface analyses for studying the amount of surface anionic groups and the surface distribution of the XTMAB. XPS and polyelectrolyte titration results suggested that the XTMAB is sorbed onto the fiber surfaces. ToF-SIMS imaging showed that XTMAB was evenly distributed on fiber surfaces.

In this contribution we propose, for the first time, the use of Methylene Violet 3 RAX dye for the detection and determination of nitrite in cured meats and vegetables. Quantification is based on the decrease in absorbance of the dye, which is proportional to the nitrite concentration The proposed method presents linear response (r > 0.99) in the range from 1.8 to 9 μmol L-1 of nitrite, with a limit of detection of 0.14 μmol L-1. Satisfactory accuracy with the official Association of Official Agricultural Chemists (AOAC) method was achieved, and the method was successfully applied to a wide range of cured meats and vegetables. The proposed method offers similar or superior performance to other optical methods used for the determination of nitrite in different matrixes, with advantage in terms of reduction of chem. waste for each anal., by minimizing amounts of reagents and products.

A series of 14 novel 2,6-dimethoxybenzamides related to the parent drug (S)-(-)-3-bromo-2,6-dimethoxy-[(1-ethyl-2-pyrrolidinyl)methyl]benzamide, were synthesized by chlorination and amination of the corresponding dimethoxybenzaldehydes, and were evaluated for the ability to block the apomorphine syndrome in rats. The 70 eV electron impact mass spectra of a wide range of 2-substituted benzazoles are reported and discussed. Particular attention is paid to the mechanistic significance and anal. utility of [M-H]+ and [M-X]+ signals in the spectra of benzazoles in which the 2-substituent contains a terminal aryl group with one or more substituents, X. Loss of H or X occurs preferentially from an ortho-position from ionized 2-benzylbenzimidazoles, 2-phenethylbenzimidazoles, 2-styrylbenzimidazoles, 2-styrylbenzoxazoles and 2-styrylbenzothiazoles. In the three styrylbenzazole series, the [M-H]+ and/or [M-X]+ signals dominate the spectra. This unusually facile loss of H or X may be attributed to a proximity effect, in which cyclization of the ionized mol. is followed by elimination of an ortho-substituent to give an exceptionally stable polycyclic ion. Formation of a new five- or six-membered ring by the proximity effect occurs rapidly; cyclization to a seven-membered ring takes place rather less readily; but formation of a ring with only four atoms or more than seven atoms is not observed to a significant extent. The proximity effect competes effectively with loss of a Me radical by simple cleavage of an Et, iso-Pr and even a t-Bu group in the pendant aromatic ring of ionized 2-(4-alkylstyryl)benzazoles.
