Two red fluorescent triads containing photochromic dithienylethene moiety linked to two 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY) via acetylene bond, 1a or single bond, 1b were synthesized and characterized. Although no evident absorption and NMR spectra changes were observed upon irradiation with visible light, the absorption and fluorescence spectra of 1a and 1b are remarkably solvent dependent, in contrast to those of the reference BODIPY-thienyl compounds 2a and 2b, which are essentially solvent independent. Fluorescence intensities of 1a and 1b decreased as the solvent polarity increased, and the fluorescence decay showed two exponentials in polar solvents other than n-hexane. Respiratory chain succinate-ubiquinone oxidoreductase (SQR or complex II) and ubihydroquinone-cytochrome (cyt) c oxidoreductase (cyt bc1 or complex III) have been demonstrated as the promising targets of numerous antibiotics and fungicides. As a continuation of our research work on the development of new fungicides, a series of 1,2,4-triazole-1,3-disulfonamide derivatives with dual functions targeting both SQR and cyt bc1 were designed and synthesized by coupling diverse di-Ph ether moieties with triazolesulfonamide units. These newly synthesized compounds were characterized by elemental analyses, 1H NMR and ESI-MS spectrometry. The in vitro assay indicated that most of the synthesized compounds displayed good inhibition against porcine succinate-cytochrome reductase (SCR) with IC50 values ranging from 3.2 to 81.8 μM, revealing much higher activity than that of the com. control amisulbrom whose IC50 value is 93.0 μM. Further evaluation against the resp. SQR and cyt bc1 indicated that most compounds exhibited SQR-inhibiting activity as well as cyt bc1-inhibiting activity, but the inhibition potency against SQR is much higher than that against cyt bc1, showing that the SCR inhibition might be contributed greatly by the SQR inhibition. The further antibacterial evaluation against Xanthomonas oryzae pv. oryzae revealed that four compounds showed excellent potency at the concentration of 20 μg mL-1. In particular, compounds (I) and (II) exhibited much better antibacterial activity than the com. control bismerthiazol in terms of their EC50. Impressively, II has an EC90 of 33.62 μg mL-1, more than 10-fold higher than that of bismerthiazol.

Gambogic acid (GA), a natural product with unique structure, was reported to have broad antiproliferation activities against cancer cell lines. As a reactive Michael acceptor, the 10-position of GA is susceptible to nucleophiles, thus limiting its clin. application as an anticancer agent. Moreover, the 6-OH forms an intramol. hydrogen bond with 8-C=O, which can make the 9, 10 double bond more reactive to nucleophiles. In this essay, two strategies (A and B) were applied to solve the above-mentioned problems. Strategy A was to increase the steric hindrance of C-10 to reduce the activity of GA towards nucleophiles. Strategy B was to replace the hydroxyl of C-6 with other substituents based on the assumption that the intra-mol. hydrogen bond could increase the electrophilicity of C-10. Results showed the electrophilicity of C-10 disappeared as well as the antiproliferation activity against cancer cell lines by introducing a Me group at C-10. Strategy B showed that the electrophilicity of C-10 was reduced dramatically while maintained the activity by replacement of the hydroxyl of C-6 with neutral or basic groups.

A series of photochromic liquid crystal dithienylcyclopentene derivatives I (n = 4, 5, 6, 7) was synthesized from the initial material 2-chloro-5-methylthiophene. They were characterized by 1H NMR, high resolution mass spectrometer, differential scanning calorimetry (DSC) and polarized optical microscopy (POM) with a hot stage. All of them exhibited excellent photochem. reversible isomerization between the open-ring form and the closed-ring form in both organic solvents and LC media, resulting in the color change between colorless and purple under different light irradiations. The typical textures from POM showed that their isomers both had nematic phase and smectic phase with different colors. These compounds with bent shape were more sensitive to light than those of diarylethenes reported.

Targeting bromodomain-containing protein 4 (BRD4) has been proved to be an effective strategy for cancer therapy. To date, numerous BRD4 inhibitors and degraders have been identified, some of which have advanced into clin. trials. In this work, a focused library of new [1,2,4]triazolo[1,5-a]pyrimidine derivatives were discovered to be able to inhibit BRD4. WS-722 inactivated BRD4 (BD1/BD2), BRD2 (BD1/BD2) and BRD3 (BD1/BD2) broadly with the IC50 values less than 5μmol/L. Besides, WS-722 inhibited growth of THP-1 cells with an IC50 value of 3.86μmol/L. Like (+)-JQ1, WS-722 inhibited BRD4 in a reversible manner and enhanced protein stability. Docking studies showed that WS-722 occupied the central acetyl-lysine (Kac) binding cavity and formed a hydrogen bond with Asn140. In THP-1 cells, WS-722 showed target engagement to BRD4. Cellular effects of WS-722 on THP-1 cells were also examined, showing that WS-722 could block c-MYC expression, induce G0/G1 phase arrest and p21 up-regulation, and promote differentiation of THP-1 cells. BRD4 inhibition by WS-722 resulted in cell apoptosis and up-regulated expression of cleaved caspased-3/7 and PARP in THP-1 cell lines. The [1,2,4]triazolo[1,5-a]pyrimidine is a new template for the development of new BRD4 inhibitors.

The present paper reports on the results of the joint interaction of two organic cationic compounds of different structure and nature, namely bisquaternary ammonium decamethoxinum (Dec) with flexible dication and methylene blue (MB) with heterocyclic planar cation, with graphene oxide (GO) in aqueous dispersion. The qual. differences in the 3D structures of GO modified in binary (GO + Dec), (GO + MB) and ternary (GO + Dec + MB) systems were observed and described on the basis of the UV-vis spectral anal. of the systems. On mixing the components of the binary (GO + Dec) system, a rather rapid gelation of GO dispersion was observed The appearance of binary (GO + MB) system showed dependence on GO to MB ratio: at low GO content, MB-induced coagulation of GO to tiny bluish flakes took place followed by their sedimentation with time while at high GO content, the GO complexes with MB preserved solubility in water. The result of the joint addition of both Dec and MB to GO dispersion differed qual. from the above results of action of individual components. On creation of the (GO + Dec + MB) ternary system rapid flocculation of all components of the system was observed resulting in the formation of dark residue surfacing over the purified decolorized liquid UV-vis spectra of the liquid phase showed suppression of the lines of all components or their complexes practically to the baseline. The compression of (GO + Dec + MB) composite to flocs is explained by the matching of the size of MB cation to that of Dec dication linker, which provides tight filling of the gaps between GO sheets. The observed flocculation effect in the (GO + Dec + MB) system may be of practical interest in the development of nanocomposite and sorption materials. Information on the features of GO interactions with organic cations differing in structure may be useful for advances in drug delivery problem.
