Combined pretreatment of sugarcane bagasse using alkali and ionic liquid to increase hemicellulose content and xylanase production

Background Lignin in sugarcane bagasse (SB) hinders its utilization by microorganism, therefore, pretreatment methods are employed to make fermentable components accessible to the microbes. Multivariate analysis of different chemical pretreatment methods can aid to select the most appropriate strategy to valorize a particular biomass. Results Amongst methods tested, the pretreatment by using sodium hydroxide in combination with methyltrioctylammonium chloride, an ionic liquid, (NaOH+IL) was the most significant for xylanase production by Bacillus aestuarii UE25. Investigation of optimal levels of five significant variables by adopting Box-Behnken design (BBD) predicted 20 IU mL(- 1) of xylanase and experimentally, a titer of 17.77 IU mL(- 1) was obtained which indicated the validity of the model. The production kinetics showed that volumetric productivity of xylanase was much higher after 24 h (833.33 IU L- 1 h(- 1)) than after 48 h (567.08 IU L- 1 h(- 1)). The extracted xylan from SB induced more xylanase in the fermentation medium than pretreated SB or commercially purified xylan. Nuclear Magnetic Resonance, Fourier transform infrared spectroscopy and scanning electron microscopy of SB indicated removal of lignin and changes in the structure of SB after NaOH+IL pretreatment and fermentation. Conclusion Combined pretreatment of SB with alkali and methyltrioctylammonium chloride appeared better than other chemical methods for bacterial xylanase production and for the extraction of xylan form SB.

GJ-4 ameliorates memory impairment in focal cerebral ischemia/reperfusion of rats via inhibiting JAK2/STAT1-mediated neuroinflammation

Ethnopharmacological relevance: Gardenia jasminoides J. Ellis (Fructus Gardenia) is a traditional Chinese medicine with diverse pharmacological functions, such as anti-inflammation, anti-depression, as well as improvement of cognition and ischemia brain injury. GJ-4 is a natural extract from Gardenia jasminoides J. Ellis (Fructus Gardenia) and has been proved to improve memory impairment in Alzheimer’s disease (AD) mouse model in our previous studies. Aim of the study: This study aimed to evaluate the therapeutic effects of GJ-4 on vascular dementia (VD) and explore the potential mechanisms. Material and methods: In our experiment, a focal cerebral ischemia and reperfusion rat model was successfully developed by the middle cerebral artery occlusion and reperfusion (MCAO/R). GJ-4 (10 mg/kg, 25 mg/kg, 50 mg/kg) and nimodipine (10 mg/kg) were orally administered to rats once a day for consecutive 12 days. Learning and memory behavioral performance was assayed by step-down test and Morris water maze test. The neurological scoring test was performed to evaluate the neurological function of rats. 2,3,5-Triphenyltetrazolium chloride (TTC) staining and Nissl staining were respectively employed to determine the infarct condition and neuronal injury of the brain. Ibal immunohistochemistry was used to show the activation of microglia. Moreover, the synaptic damage and inflammatory level were detected by Western blot. Results: GJ-4 could significantly improve memory impairment, cerebral infraction, as well as neurological deficits of VD rats induced by MCAO/R. Further research indicated VD-induced neuronal injury was alleviated by GJ-4. In addition, GJ-4 could protect synapse of VD rats by upregulating synaptophysin (SYP) expression, post synaptic density 95 protein (PSD95) expression, and downregulating N-Methyl-D-Aspartate receptor 1 (NMDAR1) expression. Subsequent investigation of the underlying mechanisms identified that GJ-4 could suppress neuroinflammatory responses, supported by inhibited activation of microglia and reduced expression of inflammatory proteins, which ultimately exerted neuroprotective effects on VD. Further mechanistic study indicated that janus kinase 2 (JAK2)/signal transducer and activator of transcription 1 (STAT1) pathway was inhibited by GJ-4 treatment. Conclusion: These results suggested that GJ-4 might serve as a potential drug to improve VD. In addition, our study indicated that inhibition of neuroinflammation might be a promising target to treat VD.

Effects of chloride ions on the durability and mechanical properties of sea sand concrete incorporating supplementary cementitious materials under an accelerated carbonation condition

The aim of this study is to investigate the effects of chloride ions on the durability and mechanical properties of non-desalted sea sand (NSS) concrete containing fly ash (FA) or ground granulated blast furnace slag (BFS) under accelerated carbonation. Six mixtures were prepared using a constant water-to-cementitious materials ratio of 0.50. The cementitious materials consisted of primarily ordinary Portland cement with a portion replaced by a supplementary material, either FA (15% by mass) or BFS (45% by mass). After being cured with a sealed condition of 20 degrees C for 28 days, half of the concrete specimens remained sealed while the other half were exposed to an accelerated carbonation chamber for 182 days. The accelerated carbonation chamber consisted of a 5% CO2 concentration with 60% relative humidity. The durability and mechanical properties of the concrete were investigated, including carbonation resistance, sorptivity, compressive strength, and the modulus of elasticity. The chloride binding capacity was also evaluated. Porosity, crack evaluation, and scanning electron microscopy tests were implemented to better understand the macro- and microscopic structures of the different concrete compositions. The results showed that the presence of chloride ions in NSS could improve the carbonation resistance of concrete. Carbonation shrinkage generated cracks which led to a significant increase in sorptivity for the FA and BFS concretes under accelerated carbonation. However, this increase was restricted by the chloride ions in NSS. In general, the presence of chloride ions enhanced the mechanical properties of the concrete, regardless of curing ages, FA or BFS replacement, or exposure conditions. Even considering the effects of carbonation, NSS is found to be potentially viable material for use in concrete production. (C) 2020 Elsevier Ltd. All rights reserved.

Extracts of medicinal plants with natural deep eutectic solvents: enhanced antimicrobial activity and low genotoxicity

Natural deep eutectic solvents (NADES) are a new alternative to toxic organic solvents. Their constituents are primary metabolites, non-toxic, biocompatible and sustainable. In this study four selected NADES were applied for the extraction of two medicinal plants: Sideritis scardica, and Plantago major as an alternative to water-alcohol mixtures, and the antimicrobial and genotoxic potential of the extracts were studied. The extraction efficiency was evaluated by measuring the extracted total phenolics, and total flavonoids. Best extraction results for total phenolics for the studied plants were obtained with choline chloride-glucose 5:2 plus 30% water; but surprisingly these extracts were inactive against all tested microorganisms. Extracts with citric acid-1,2-propanediol 1:4 and choline chloride-glycerol 1:2 showed good activity against S. pyogenes, E. coli, S. aureus, and C. albicans. Low genotoxicity and cytotoxicity were observed for all four NADES and the extracts with antimicrobial activity. Our results confirm the potential of NADESs for extraction of bioactive constituents of medicinal plants and further suggest that NADES can improve the effects of bioactive extracts. Further studies are needed to clarify the influence of the studied NADES on the bioactivity of dissolved substances, and the possibility to use such extracts in the pharmaceutical and food industry.

A novel experimental method to determine the critical chloride content in cement-based composites

In reinforced-concrete structural members, the protective passive film on the rebar surface is broken when a critical content of chloride ions (C-crit) is accumulated at the rebar-concrete interface by diffusing through the concrete cover. The value of C-crit is often used for determining maintenance schedules and service life modeling of reinforced-concrete infrastructure. A novel method for accurate determination of C-crit is proposed in this article, which addresses some of the limitations of current practice. For the development of this method, seventeen identical beam specimens, made with reinforced-StrainHardening Cementitious Composite (SHCC), were prepared according to the ASTM G109 procedure. The corrosion activity was monitored using various electrochemical techniques to detect the initiation of corrosion. Immediately after corrosion initiation, each specimen was sectioned to determine the chloride contents at multiple points along the rebar-SHCC interface and to observe the depths of the tiny corrosion pits on the surface of the rebars. We found a linear correlation between the chloride content and the average corrosion pit-depth and determined the C-crit of SHCC as the y-intercept of this linear correlation. (C) 2020 Elsevier Ltd. Regulatory influences of methyl jasmonate and calcium chloride on chilling injury of banana fruit during cold storage and ripening

Fruit quality is preserved through cold storage, but climacteric fruits are prone to chilling injury (CI) which limits their shelf life and marketability. Two postharvest treatments, 1 mM methyl jasmonate (MeJA) and 4% (wt/vol) calcium chloride (Ca2+), were separately used to investigate their influences on chilling injury (CI) incidence and fruit quality in unpacked banana cultivar Grand Nain during cold storage and subsequent ripening. Banana fruits were dipped for 2 min in aqueous emulsions containing 1% Tween-80-used here as a surfactant with untreated fruits being used as control. Fruits were stored at 10 +/- 2 or optimal 14 +/- 2 degrees C temperature and relative humidity 85%-90% for a 20-day cold storage period and then removed from cold storage at 5, 10, 15, and 20 days followed by ripening at 22 +/- 2 degrees C. Treatments with MeJA or Ca2+ significantly reduced CI in banana fruit during cold storage and subsequent ripening temperature. Untreated controls exhibited increased CI, weight loss, and decreased hue angle, as well as firmness. In contrast, the aforementioned changes were considerably delayed after treatments with MeJA or Ca2+. Application of MeJA or Ca2+ also increased total phenolic compound contents and maintenance of total antioxidant activity throughout cold storage and during ripening periods as compared to that of the control. These findings indicate that coating bananas with 1 mM MeJA or 4% (wt/vol) Ca2+ can improve the postharvest quality and shelf life of fruits, and it can ameliorate chilling injury during cold storage and at ripening temperature.

Bio-improved hydraulic properties of sand treated by soybean urease induced carbonate precipitation and its application Part 2: Sand-geotextile capillary barrier effect

Nonwoven-geotextile is used to construct soil-geotextile capillary barrier for minimizing water infiltration. However, the sand-geotextile capillary barrier breakthrough can occur due to the low water retention ability and the high saturated permeability coefficient of sand. To address this, enzyme induced carbonate precipitation (EICP) technology was applied to the overlying sand layer to improve its hydraulic properties and enhance the sand-geotextile capillary barrier. The precipitated calcium carbonate has the potential to enhance the capillary barrier effect by coating soil particles and filling the soil macro-pores. To investigate the effect of EICP treatment on the sand-geotextile capillary barrier, three column tests were conducted, including one untreated sand-geotextile column and two sand-geotextile columns treated with 0.5 and 1 mol/L urea-calcium chloride solutions and soybean urease liquid, respectively. Transient seepage analysis was carried out based on the column tests to calibrate the hydraulic parameters. A numerical parametric analysis was conducted to assess the effect of EICP-enhancement on the hydraulic behavior and slope stability of a sand-geotextile capillary barrier retaining wall during exposure to rainfall with a 100-year return period. The results of the column infiltration tests show that the occurrence of breakthrough in the sand-geotextile capillary barrier is significantly delayed with increasing concentration of the urea-calcium chloride solutions. The enhanced capillary barrier effect is attributed to the increased water retention ability and reduced saturated permeability coefficient. The results of the parametric analysis show that the matric suction within the backfill is maintained and the factor of safety remains almost unchanged due to the EICP-enhanced sand-geotextile capillary barrier effect.
