The challenging synthesis of multicrystalline polymers has resulted in only a limited number of tricryst. terpolymers having been reported to date. We report, for the first time, the synthesis of polyethylene-b-poly(ethylene oxide)-b-poly(ε-caprolactone)-b-poly(L-lactide) (PE-b-PEO-b-PCL-b-PLLA), a tetracryst. tetrablock quarterpolymer, by combining polyhomologation, ring-opening polymerization, and an organic/metal “catalyst switch” strategy. 1H NMR spectroscopy and gel-permeation chromatog. confirmed the formation of the tetrablock quarterpolymer, while differential scanning calorimetry, X-ray diffraction, and wide-line separation solid-state NMR spectroscopy revealed the existence of four different crystalline domains.

This study presents for the first time, a photo stimuli responsive membrane system based on NAAMs functionalized with an azobenzene-containing photoswitchable peptide (PSP). PSP mols. were selectively immobilized along the internal surface of the nanoporous anodic alumina membranes (NAAM) pores, in order to manipulate the effective pore diameter of the NAAMs, depending on their isomeric form. This pore diameter modulation then regulates the transport of model dye mol. (Rose Bengal (RosB)) across the PSP modified NAA membranes. Exposure to specific wavelengths of light (364 nm in this case) switches the azobenzene component of PSP from a trims to a cis isomer, such that the associated, change in PSP geometry controls the pore diameter of NAAMs and hence the selective transport of RosB.

Early detection of skin diseases is imperative for their effective treatment. However, fluorescence mol. probes that allow this are rare. The first activatable near-IR (NIR) fluorescent mol. probe is reported for sensitive imaging of keloid cells, skin cells from abnormal scar fibrous lesions. As keloid cells have high expression levels of fibroblast activation protein-alpha (FAPα), the probe (FNP1) is designed to have a caged NIR dye and a FAPα-cleavable peptide substrate linked by a self-immolative segment. FNP1 can quickly and specifically turn on its fluorescence at 710 nm by 45-fold in the presence of FAPα, allowing it to effectively recognize keloid cells from normal skin cells. Integration of FNP1 with a simple microneedle-assisted topical application enables sensitive detection of keloid cells in metabolically-active human skin tissue with a theor. limit of detection down to 20 000 cells.

Herein, we report that highly chemoselective and enantioselective reduction of 2-vinyl-substituted pyridines has been achieved for the first time. The reaction, which uses chiral spiro-bicyclic bisboranes as catalysts and HBpin and an acidic amide as reducing reagents, proceeds through a cascade process involving 1,4-hydroboration followed by transfer hydrogenation of a dihydropyridine intermediate. The retained double bond in the reduction products permits their conversion to natural products and other useful heterocyclic compounds by simple transformations.

Subcellular delivery of nanomedicines has emerged as a promising approach to enhance the therapeutic efficacy of anticancer drugs. Nuclear accumulation of anticancer drugs are essential for its therapeutic efficacy because their targets are generally located within the nucleus. However, strategies for the nuclear accumulation of nanomedicines with anticancer drugs rarely reported. In this study, we reported a promising nanomedicine, comprising a drug-peptide amphiphile, with enhanced cellular uptake and nuclear accumulation capability for cancer therapy. The drug-peptide amphiphile consisted of the peptide ligand PMI (TSFAEYWNLLSP), which was capable of activating the p53 gene by binding with the MDM2 and MDMX located in the cell nucleus. Peptide conformations could be finely tuned by using different strategies including heating-cooling and enzyme-instructed self-assembly (EISA) to trigger mol. self-assembly at different temperatures Due to the different peptide conformations, the drug-peptide amphiphile self-assembled into nanomedicines with various properties, including stabilities, cellular uptake, and nuclear accumulation. The optimized nanomedicine formed by EISA strategy at a low temperature of 4 C showed enhanced cellular uptake and nuclear accumulation capability, and thus exhibited superior anticancer ability both in vitro and in vivo. Overall, our study provides a useful strategy for finely tuning the properties and activities of peptide-based supramol. nanomaterials, which may lead to optimized nanomedicines with enhanced performance.
