The residue was purified by column chromatography on silica gel (solvent; n-hexane/ethyl acetate = 4/1 -? 1/1) to give 7-amino-4-(4-chloro- 3-methylphenyl)-2,2-dimethyl-2H-l,4-benzoxazin-3(4H)-one (98 mg) as a pale orange powder.MS(APCI) m/z: 317/319 [M+H]+

HPLC retention time: 1.12 min (analysis condition A) 1H-NMR (400 MHz, CDCl3) delta: 7.92 (1H, s), 7.40 (1H, s), 1.43 (18H, s).

Into a 100-mL round-bottom flask, was placed a solution of N-[(2,4-dimethoxyphenyl)methyl]-3- (methoxymethyl)-l ,2,4-thiadiazol-5-amine (300 mg, 1 .02 mmol, 1 .00 equiv) in N,N-dimethylformamide (30 mL). To the solution were added 2,4,5-trifluorobenzene-1 -sulfonyl chloride (626.4 mg, 2.72 mmol, 1 .00 equiv) and sodium hydride (162.7 mg, 6.78 mmol, 1 .50 equiv). The resulting solution was stirred for 1 6 h at room temperature. The resulting solution was extracted with ethyl acetate (3×30 mL) and the organic layers combined and concentrated under vacuum . The residue was applied onto a silica gel column with ethyl acetate/petroleum ether (1 :10). The collected fractions were combined and concentrated under vacuum . This resulted in 350 mg (70%) of N-[(2,4-dimethoxyphenyl)methyl]-2,4,5-trifluoro-N-[3- (methoxymethyl)-l ,2,4-thiadiazol-5-yl]benzene-1 -sulfonamide as a white solid.

To a mixture of 3-bromo-6-chloro-1-yl-imidazo[1,2-b]pyridazine (1.00 g, 4.3 mmol) in 1,2-dimethoxy ethane (1 mL) was added diisopropyl ethyl amine (0.5 mL) and 1-BOC-piperazine (2.41 g, 13 mmol). The mixture was degassed (4 cycles of vacuum and nitrogen) and heated at 120 C. for 27 hours. The reaction mixture was cooled to room temperature, treated with saturated aqueous sodium bicarbonate (100 mL) and extracted with ethyl acetate (2¡Á100 mL). The organic layers were combined and dried with magnesium sulfate, filtered and evaporated to a residue, which was purified by chromatography on silica gel; elution with a gradient of hexane/ethyl acetate gave 4-(3-bromo-imidazo[1,2-b]pyridazine-6-yl)-piperazine-1-carboxylic acid tert-butyl ester as an off-white solid (1.01 g, 83%): mp 176-9 C.; LC/MS [M+H]+ 382, 384.

The residual brown oil was chromatographed on silica gel with a 95:5 v/v hexane-EtOAc eluent to afford 10.0 g (81%) of 1-(2,6-dichloro-4-trifluoromethylphenyl)-2,5-dimethylpyrrole as a light brown solid of melting point 63 C.
