The direct C2-functionalization of pyridines I (R = H, Me, Cl, pyrrolidin-1-yl, etc.) through a transition-metal-free protocol by using arynes 2-Si(CH3)3-3-R1-4-R2-5-R3-6-R4C6OS(O)2CF3 (R1 = H, Me, OMe; R2 = H, Me, F, Cl; R1R2 = -(CH2)4-, -CH=CH-CH=CH-; R3 = H, Me, F; R2R3 = -OCH2O-, -(CH2)2-, -CH=CH-CH=CH-; R4 = H, Me) multicomponent coupling is demonstrated. The reaction allowed a broad-scope synthesis of C2-substituted pyridine derivatives bearing the -CF3 group II in good yields with α,α,α-trifluoroacetophenones ArC(O)CF3 (Ar = Ph, pyren-1-yl, 1-benzothiophen-3-yl, etc.) as the third component. Activated keto esters could also be employed as the third component in this formal 1,2-di(hetero)arylation of ketones. Performing the reaction under dilute conditions inhibited the competing pyridine-aryne polymerization pathway. Nucleophilic attack by the initially generated pyridylidene intermediate on the carbonyl followed by an SNAr process resembling the Smiles rearrangement affords the desired products.

3,3-Disubstituted 3-silaglutarates were synthesized by reaction of the lithium enolate of Et acetate with dichlorosilanes at -94°. When a Ph group is linked to the silicon atom the reaction in THF gave mainly the silaglutarate. In other cases alkylsilyl ketene acetals were formed in proportions similar to or greater than that of the silaglutarate. The presence of HMPA increased the ratio of C- to O-silylated products and allowed the preparation of 3,3-dialkyl 3-silaglutarates in good yield.

The configuration culminates in a large-scale campaign where the flow platform is utilized to process over 100 g of pure product over 7 h, in a yield of 80% after workup.

Maternal obesity programs liver derangements similar to those of NAFLD. Our main goal was to evaluate whether these liver anomalies were related to aberrant PPARα function. Obesity was induced in female Albino-Wistar rats by a fatty diet (FD rats). Several parameters related to NAFLD were evaluated in both plasma and livers from fetuses of 21 days of gestation and 140-day-old offspring. FD fetuses and offspring developed increased levels of AST and ALT, signs of inflammation and oxidative and nitrative stress-related damage. FD offspring showed dysregulation of Plin2, CD36, Cyp4A, Aco, Cpt-1, Hadha and Acaa2 mRNA levels, genes involved in lipid metabolism and no catabolic effect of the PPARα agonist clofibrate. These results suggest that the FD offspring is prone to develop fatty liver, a susceptibility that can be linked to PPARα dysfunction, and that this could in turn be related to the liver impairments programmed by maternal obesity.
