As our research focuses on anticancer drugs, a series of novel derivatives of flexicaulin A (FA), an ent-kaurene diterpene, condensed with an aromatic ring were synthesized, and their antiproliferative activities against four human cancer cell lines (TE-1, EC109, MCF-7, and MGC-803) were evaluated. The activities of most of the new compounds were better than those of FA. Compound 2y exhibited the best activity with an IC50 value reaching 0.13 mu M against oesophageal cancer cells (EC109 cells). The IC50 values for 2y in normal cells (GES-1 cells and HUVECs) were 0.52 mu M and 0.49 mu M, respectively. Subsequent mechanistic investigations found that compound 2y can inhibit the proliferation of cancer cells and cell cloning. In addition, 2y could reduce the mitochondrial membrane potential, increase the apoptosis rate, and increase the ROS level in EC109 cells. Moreover, 2y can upregulate the expression of ROS/JNK pathway-related proteins (p-ASK1, p-MKK4, p-JNK, and p-Cjun (ser63)) and pro-apoptotic proteins (Bax, Bad, and Bim). In vivo experiments showed that 2y can inhibit tumour growth in nude mice. The mechanism involves an increase in protein expression in the ROS pathway, leading to changes in apoptosis-related proteins. In addition, compound 2y shows low toxicity. Electrocatalytic hydrodechlorination (ECH) has been proposed as a potential technology for the effective degradation of organochlorine contaminants. In this study, a nanoscale Pd/TiC catalyst with low resistance was dip-coated onto Ti, aiming to fabricate a stable Pd/TiC/Ti electrode with enhanced ECH reactivity for efficient dechlorination. Electrochemical techniques indicated that the Pd/TiC/Ti electrode had a larger electrochemically active surface area and more rapid interfacial charge transfer than the conventional Pd/C/Ti electrode. In a batch experiment, the dechlorination rate for ECH of 2,4-dichlorobenzoic acid (2,4-DCBA, a model pollutant) by Pd/TiC/Ti was 1.3-14.5 times higher than those by Pd/C/Ti, Pd/MWCNTs/Ti, and Pd/MoS2/Ti, respectively. Scavenger experiments, ESR spin-trapping spectroscopy, and cyclic voltammetry clarified the intensified atomic H* generation by Pd/TiC/Ti. DFT calculations revealed that Pd/TiC/Ti had stronger binding ability with atomic H* (E-ads: -2.89 eV vs. -2.44 eV) and 2,4-DCBA (E-ads: -3.62 eV vs. -2.15 eV), respectively, when compared with Pd/C/Ti. As a result, enhanced generation of atomic H*, together with strengthened adsorption of atomic H* and 2,4-DCBA, would contribute to a faster ECH reaction. The Pd/TiC/Ti electrode retained its reactivity after 10 successive batch experiments (80 h), and showed high tolerance to the constituents in actual water matrices including lake water and river water. This study proposed a promising ECH electrode for the treatment of chlorinated organics in water matrices.

Dual effects of cathodic dechlorination and oxidative decomposition of 2,4-dichlorobenzoic acid (2,4-DCBA) can be achieved using electrochemical reduction-oxidation technology. The removal efficiency was improved by optimizing the experimental conditions affecting electrode activity and contaminant degradation. The 2,4-DCBA removal efficiency reached 90.8% at a current density at 50 mA.cm(-2), initial pH of 5, and sodium sulfate concentration of 0.05 mol.L-1, with a 2,4-DCBA concentration of 20 mg.L-1. Furthermore, a good total organic carbon removal efficiency of 81.6% was achieved in the electrocatalytic reduction and oxidation process, with the present system retaining high stability after many experiment cycles. The reaction mechanism under different conditions is discussed, which provides new approaches to the degradation of chlorinated organic compounds.

Green, simple, and highly efficient various strategies for the straightforward reduction of carboxylic acids to corresponding alcohols have been developed by employing four different types of cost-effective hydrogen donors, namely sodium borohydride (NaBH4), ammonium formate (HCO2NH4), glycerol, and isopropanol (i-PrOH) in the presence of Fe3O4@APTMS@ZrCp2Clx ((x = 0, 1, 2)) as a robust and durable nanocatalyst under neat conditions. Also, the mentioned magnetically separable nanocatalyst was recovered and reused at least five times without significant loss in catalytic activity. In comparison with the known protocols for the direct reduction of carboxylic acids to corresponding alcohols, our current methods offer several benefits such as being straightforward (without isolation of aldehyde intermediates), mild reaction conditions, eco-friendly, short reaction times, good to excellent yields of the products, use of inexpensive hydrogen donors, recoverability, and reusability of the catalyst and so on.

Unlike other DNA topoisomerase II (topo II) inhibitors, our recently identified acridone derivative E17 exerted strong cytotoxic activity by inhibiting topo II without causing topo II degradation and DNA damage, which promoted us to explore more analogues of E17 by expanding its chemical diversification and enrich the structure-activity relationship (SAR) outcomes of acridone-oriented chemotypes. Asymmetric Schiff base manganese(III) complexes involving salen ligands, N,N’-bis(salicylidene)2,3-diaminopyridine, N,N’-bis(3-methoxysalicylidene)2,3-diaminopyridine, N,N’-bis(3,5-di-tert-butylsalicylidene)2,3-diaminopyridine and N,N’-bis(3,5-di-chloro-salicylidene)2,3-diaminopyridine were prepared and their catalytic activity was investigated in the oxidation of some primary and secondary alcohols. During optimization of oxidation reactions, Mn-4, bearing electron withdrawing N,N’-Bis(3,5-di-chloro-salicylidene)2,3-diaminopyridine ligand, showed higher activity than other catalysts tested. The catalytic reactions were carried out in the presence of various oxidants such as oxygen, hydrogen peroxide or tert-butyl hydroperoxide (TBHP) and additives such as acetic acid and imidazole. The oxidant/additive combination of TBHP and imidazole was shown to be effective for the oxidation process and the degree of their impact on oxidation reaction was found highly dependent on a balanced ratio between them. Mn-4 was selected as the most effective catalyst under optimized reaction conditions and revealed efficient for the oxidation of secondary alcohols.

N-benzhydrylpiperazine and 1,3,4-oxadiazoles are pharmacologically active scaffolds which exhibits significant inhibitory growth effects against various cancer cells, however, antiproliferation effects and the underlying mechanism for inducing apoptosis for aforementioned scaffolds addressing HeLa cancer cells remains uncertain. In this study, N-benzhydrylpiperazine clubbed with 1,3,4-oxadiazoles (4a-4h) were synthesized, subsequently characterized using high resolution spectroscopic techniques and eventually evaluated for their antiproliferation potential by inducing apoptosis in HeLa cancer cells. The MTT assay screening results revealed that among all, compound 4d (N-benzhydryl-4-((5-(4-aminophenyl)-1,3,4-oxadiazol-2-yl)methyl)piperazine) in particular, exhibited IC (50) value of 28.13 +/- 0.21g/mL and significantly inhibited the proliferation of HeLa cancer cells in concentration-dependent manner. The in vitro anticancer assays for treated HeLa cells resulted in alterations in the cell morphology, reduction in colony formation, and inhibition of cell migration in concentration-dependent treatment. Furthermore, G2/M phase arrest, variations in the nuclear morphology, degradation of chromosomal DNA confirmed the ongoing apoptosis in treated HeLa cells. Increase in the expression of cytochrome C and caspase-3 confirmed the involvement of intrinsic mitochondrial pathway regulating the cell death. Also, elevation in reactive oxygen species level and loss of mitochondrial membrane potential signified that compound 4d induced apoptosis in HeLa cells by generating the oxidative stress. Therefore, compound 4d may act as a potent chemotherapeutic agent against human cervical cancer.

Despite remarkable advances to promote regio- and stereoselective decarboxylative arylation of inactivated olefins with benzoic acid derivatives, methodologies involving hetero-substituted alkenes are still lacking. Herein, Pd-II-catalyzed decarboxylative Heck coupling of alpha-alkoxyacrylates with (hetero)aryl carboxylic acids for the stereocontrolled production of (Z)-beta-heteroarylated vinyl ethers is reported. This methodology offers a rational and step-economical route to the synthesis of attractive beta-arylated alpha-alkoxy alpha,beta-unsaturated carboxylates family which emerged as a relevant class of building blocks with different applications. Mechanistically, whereas electron rich benzoic acids undergo a Pd-II-catalyzed decarboxylation, electron-deficient substrates proceed through silver(I)-mediated decarboxylation, explaining thus the formation of stereoisomers (E) and (Z) of beta-arylated vinyl ethers in presence of these latter.
