It was demonstrated as to how carbohydrates can be used as conformational control units of organocatalysts to tune their catalytic properties was introduced. Taking full advantage of the modular nature of the carbohydrate motif, both reactivity and stereoselectivity were significantly improved. By simple structural changes, such as the elimination of the methoxy group in the C4 position of the tetrahydropyran ring, we obtained two complementary catalysts that allow accessed to both enantiomers of the γ-nitroaldehydes with excellent yields, diastereoselectivity and enantiomeric excesses between 97 and 99%, using a catalytic load even below 1 mol%.

Herein, we demonstrate a facile fabrication strategy to prepare novel template-shape-replicated nitrogen-rich mesoporous carbon nanospheres (NMCN) with high surface area, pore volume and ordered pore geometry via a combination of in situ co-condensation, surface-confined atom transfer radical polymerization (SC-ATRP) and post-polymerization modification. NMCN exhibited efficient CO2 capture performance and contaminant removal capability from wastewater. This work may open a new direction for the design of novel porous nanomaterials for CO2 capture and other emerging applications.

Background: Farnesyl pyrophosphate synthase (FPPS) is a key regulatory enzyme in the biosynthesis of cholesterol and in the post-translational modification of signaling proteins. It has been reported that non-bisphosphonate FPPS inhibitors targeting its allosteric binding pocket are potentially important for the development of promising anti-cancer drugs. Methods: The following methods were used: organic syntheses of non-bisphosphonate quinoline derivatives, enzyme inhibition studies, fluorescence titration assays, synergistic effect studies of quinoline derivatives with zoledronate, ITC studies for the binding of FPPS with quinoline derivatives, NMR-based HAP binding assays, mol. modeling studies, fluorescence imaging assay and MTT assays. Results: We report our syntheses of a series of quinoline derivatives as new FPPS inhibitors possibly targeting the allosteric site of the enzyme. Compound 6b showed potent inhibition to FPPS without significant hydroxyapatite binding affinity. The compound showed synergistic inhibitory effect with active-site inhibitor zoledronate. ITC experiment confirmed the good binding effect of compound 6b to FPPS, and further indicated the binding ratio of 1:1. Mol. modeling studies showed that 6b could possibly bind to the allosteric binding pocket of the enzyme. The fluorescence microscopy indicated that these compounds could get into cancer cells. Conclusions: Our results showed that quinoline derivative 6b could become a new lead compound for further optimization for cancer treatment. General significance: The traditional FPPS active-site inhibitors bisphosphonates show poor membrane permeability to tumor cells, due to their strong polarity. The development of new non-bisphosphonate FPPS inhibitors with good cell membrane permeability is potentially important.

Substituted N-Ph cinnamamide derivatives I (R = OMe, OEt, CN, Cl, N(Me)2) were designed and synthesized to confirm activation of nuclear factor (erythroid-derived 2)-like 2 (Nrf2) pathway by the electronic effect on beta-position of Michael acceptor according to introducing the R1 and R2 group. Compounds were screened using the Nrf2/antioxidant response element (ARE)-driven luciferase reporter assay. Compound II (R = N(Me)2) showed desirable luciferase activity in HepG2 cells without cell toxicity. MRNA and protein expression of Nrf2/ARE target genes such as NAD(P)H quinone oxidoreductase 1, hemeoxygenase-1, and glutamate-cysteine ligase catalytic subunit (GCLC) were upregulated by compound II (R = N(Me)2) in a concentration-dependent manner. Treatment with compound II (R = N(Me)2) resulted in increased endogenous antioxidant glutathione, showing strong correlation with enhanced GCLC expression for synthesis of glutathione. In addition, tert-Bu hydroperoxide (t-BHP)-generated reactive oxygen species were significantly removed by compound II (R = N(Me)2) , and the results of a cell survival assay in a t-BHP-induced oxidative cell injury model showed a cytoprotective effect of compound II (R = N(Me)2) in a concentration dependent manner. In conclusion, the novel compound II (R = N(Me)2) can be utilized as an Nrf2/ARE activator in antioxidative therapy.

To investigate the alignment behavior of liquid crystal (LC) mols., we synthesized a series of vanillin derivative-substituted polystyrene films (PVAN#, # = 20, 40, 60, 80, and 100), where # is the substitution ratio (%) of the vanillyl Bu ether (VAN) moiety as one of the bio-renewable vanillin derivatives In general, a vertical LC alignment was observed in the LC cell fabricated using the polymer film having a higher molar content of VAN. Moreover, the surface energies of these polymer films were strongly related to the vertical LC alignment behavior. For example, a uniform vertical alignment was observed when the polar surface energy of the polymer was lower than approx. 2.05 mJ m-2, induced by the nonpolar and long carbon groups in the plant-based vanillin moiety. The LC cell fabricated using PVAN100 had good electrooptical characteristics such as voltage holding ratio and residual direct-current voltage and stable alignment under extreme external conditions.

This manuscript describes the structure-property-morphol. relationships of doubly charged 1,4-diazabicyclo[2.2.2]octane (DABCO) salt-containing ABA triblock ionomers. The triblock copolymers consist a soft poly(Bu acrylate) (PnBA) central block and two external styrenic hard blocks bearing amphiphilic pendant C18-alkyl groups and doubly charged salt units. Surprisingly, the DABCO salt-containing ABA block copolymers preserved the thermomech. integrity until degradation, which indicated the formation of a reinforcing phys. network compared to the corresponding doubly charged random copolymers and singly charged block copolymer analogs. Small-angle X-ray scattering data revealed that the DABCO-based ABA block copolymers self-assembled into highly ordered hierarchical microstructures, in which the soft and hard domain of the block copolymers phase-separated into highly ordered lamellar morphologies. Moreover, a secondary structure that originated from the ordering of the amphiphilic pendant groups formed within the lamellar hard domain. The interesting thermal, thermomech., and morphol. properties of doubly charged ionic block copolymers open promising avenues for the synthesis of novel thermoplastic elastomers.
