The present study demonstrated the utility of a reverse ligand similarity based approach to identify potential targets for a new series of pyrazole carboxamides I [R = H, R1 = H, methoxy, etc.; R2 = H, Me, fluoro, etc.] that showed a potent anticancer activities against MCF-7 cells compared to other structurally related mols. and controls. Mol. docking studies revealed that the compound I [R = R1 = H, R2 = fluoro] interacted very strongly with human estrogen receptor with docking score of -8.82kcal/mol. In case of hERα-LBD, it interacted very well via hydrogen bond with Thr347 with docking score of -8.19 kcal/mol and very good binding with Asp351 via both hydrogen bonding and salt bridge. These results indicated that compound I [R = R1 = H, R2 = fluoro] acted as better inhibitor for estrogen receptors comparable with the standard inhibitor tamoxifen. Further, ADMET studies revealed that pyrazole carboxamides I [R = H, R1 = H, methoxy, etc.; R2 = H, Me, fluoro, etc.] had good ADME properties as compared with the data from 95% of drugs readily available in the market. The pyrazole carboxamides I [R = H, R1 = H, methoxy, etc.; R2 = H, Me, fluoro, etc.] possessed three dimensional supramol. self-assembly, in which N-H···O bonds connected the mols. and build up two dimensional arrays, which were extended to 3D network through C-H···Cg and C-Cl···Cg interactions. The structure also exhibited π···π stacking interactions, which contributed to the crystal packing. The Hirshfeld surface anal. was carried out to quantify the intermol. interactions involved in the crystalline environment. The mol. geometry of the compound I [R = H, R1 = methoxy, R2 = H] by DFT method with B3LYP hybrid functional and 6-311+G(d,p) basis set. The optimized structure was in good agreement with the exptl. findings. The electrostatic potential map was plotted to understand the energy distribution and chem. reactive regions of the mol.

Kalhor, Mehdi;Zarnegar, Zohre research published ã€?1-Methylimidazolium ionic liquid supported on Ni@zeolite-Y: fabrication and performance as a novel multi-functional nanocatalyst for one-pot synthesis of 2-aminothiazoles and 2-aryl benzimidazolesã€? the research content is summarized as follows. In the present study, 1-methyl-3-(3-trimethoxysilylpropyl)-1H-imidazol-3-ium chloride-supported Ni@zeolite-Y-based nanoporous materials (Ni@zeolite-Im-IL) were synthesized and their structures were confirmed using different characterization techniques such as FT-IR, FE-SEM, EDX, XRD, BET and TGA-DTG analyses. In order to synthesize this multi-functional nano-system, zeolite-NaY was modified first, with exchanged Ni2+ ions and 3-chloropropyltriethoxysilane (CPTES) as a coupling reagent and then functionalized to imidazolium chloride ionic liquid by N-methylimidazole. New multi-functional nano-material of Ni@zeolite-Im-IL demonstrated high activity in the catalytic synthesis of 2-aminothiazoles 3a-l by one-pot reaction of methylcarbonyls, thiourea and iodine at 80°C in DMSO with good to excellent yields (85-98%). Also, the catalytic synthesis of 2-aryl benzimidazoles, 6a-m was performed by the condensational reaction of o-arylendiamine and aromatic aldehydes in EtOH at room temperature with excellent yields (90-98%). Advantages of this efficient synthetic strategy include higher purity and shorter reaction time, excellent yield, easy isolation of products, the good stability, activity and feasible reusability of the metallic ionic liquid nanocatalyst. These benefits have made this method more compatible with the principles of green chem.

A series consisting of 117 2-(halogenated phenyl) acetamide and propanamide analogs were investigated as TRPV1 antagonists. The structure-activity anal. targeting their three pharmacophoric regions indicated that halogenated Ph A-region analogs exhibited a broad functional profile ranging from agonism to antagonism. Among the compounds, antagonists I and II exhibited potent antagonism toward capsaicin for hTRPV1 with Ki[CAP] = 2.6 and 6.9 nM, resp. Further, antagonist II displayed promising analgesic activity in vivo in both phases of the formalin mouse pain model. A mol. modeling study of II indicated that the two fluoro groups in the A-region made hydrophobic interactions with the receptor.

Karimi Askarani, Hajar;Karimi Zarchi, Mohammad Ali;Mirjalili, BiBi Fatemeh;Bamoniri, Abdolhamid research published One-pot synthesis of polyhydroquinoline derivatives using nano-Fe3O4@dextrin/BF3 as a magnetic biodegradable catalyst the research content is summarized as follows. In this study, the magnetized nano-dextrin-supported boron trifluoride (nano-Fe3O4@dextrin/BF3) as a green biopolymer-based catalyst is synthesized in two steps. In the first step, nano-dextrin is magnetized (nano-Fe3O4@dextrin), and in the second step, the BF3 is supported to the obtained nano-Fe3O4@dextrin. The structure of nano-Fe3O4@dextrin/BF3 is characterized by different techniques, including Fourier transform IR (FT-IR) spectroscopy, X-ray diffraction (XRD) pattern, energy dispersive X-ray (EDX) anal., SEM (SEM) imaging, mapping, vibrating sample magnetometer (VSM), and thermogravimetric anal. (TGA/DTG). The catalytic efficiency was also investigated in the synthesis of biol. active polyhydroquinoline derivatives via Hantzsch condensation reaction of aromatic aldehydes, dimedone, Et acetoacetate, and ammonium acetate under solvent-free conditions. The structure of products was confirmed by their m.ps. (MP), FT-IR, and 1H and 13C NMR (NMR) spectroscopy. This procedure enjoys advantages such as short reaction time, clean and fast work-up, and easy separation of the catalyst by an external magnet.

Katopodi, Annita;Tsotsou, Evangelia;Iliou, Triantafylia;Deligiannidou, Georgia-Eirini;Pontiki, Eleni;Kontogiorgis, Christos;Tsopelas, Fotios;Detsi, Anastasia research published ã€?Synthesis, Bioactivity, Pharmacokinetic and Biomimetic Properties of Multi-Substituted Coumarin Derivativesã€? the research content is summarized as follows. A series of novel multi-substituted coumarin derivatives I [R1 = H, F, Cl; R2 = H, Cl, Br, etc.; R3 = H, OH, Cl, etc.; R4 = H, Br; R5 = H, Br, Cl; R6 = H, OH, acetoxy] were synthesized, spectroscopically characterized, and evaluated for their antioxidant activity, soybean lipoxygenase (LOX) inhibitory ability, their influence on cell viability in immortalized human keratinocytes (HaCaT) and cytotoxicity in adenocarcinomic human alveolar basal epithelial cells (A549) and human melanoma (A375) cells, in-vitro. Coumarin analogs I, bearing a hydroxyl group at position 5 of the coumarin scaffold and halogen substituents at the 3-Ph ring, were the most promising ABTSâ€? scavengers. 6,8-Dibromo-3-(4-hydroxyphenyl)-4-methyl-chromen-2-one and 6-bromo-3-(4,5-diacetyloxyphenyl)-4-methyl-chromen-2-one exhibited significant lipid peroxidation inhibitory activity (IC50 36.9 and 37.1μM). In the DCF-DA assay, the 4′-fluoro-substituted compound I [R1=R2=R4=R5 = H, R3 = F; R6 = bacetoxy] (100%), and the 6-bromo substituted compounds I [R1=R2=R4=R6 = H; R3 =acetoxy; R5 =Br] (80.9%) and I [R1=R2=R4=R6 = H; R3 = OH; R5 = Br] (100%) presented the highest activity. The 3′-fluoro-substituted coumarins I [R1=R3=R4=R5 = H; R2 = F, R6 = acetoxy] and I [R1=R3=R4=R5 = H; R2 = F; R6 = OH], along with 3-(4-acetyloxyphenyl)-6,8-dibromo-4-methyl-chromen-2-one, were the most potent lipoxygenase (LOX) inhibitors (IC50 11.4, 4.1, and 8.7μM, resp.) while displaying remarkable hydroxyl radical scavenging ability, 85.2%, 100%, and 92.9%, resp. In silico docking studies of compounds I [R1=R3=R4=R5 = H; R2 = F; R6 = OH] and [R1=R2=R6 = H; R3 = acetoxy; R4=R5 = Br], revealed that they present allosteric interactions with the enzyme. The majority of the analogs (100μM) did not affect the cell viability of HaCaT cells, though several compounds presented over 60% cytotoxicity in A549 or A375 cells. Finally, the human oral absorption (%HOA) and plasma protein binding (%PPB) properties of the synthesized coumarins I were also estimated using biomimetic chromatog., and all compounds presented high %HOA (>99%) and %PPB (60-97%) values.
