The crude product was purified by preparative TLC (0-100% ethyl acetate in heptane) to give a 4:1 mixture of 6- and 5-nitrated product (232 mg). 80 mg of this mixture was dissolved in pyridine (1 mL). Nortropine (198 mg, 1.553 mmol) was added and the mixture was heated to 110 C. in a sealed flask for 20 hours and then concentrated. The residue was dissolved in 2 M HCl (20 mL) and extracted with dichloromethane (2×20 mL). The combined organic phases were dried over Na2SO4, filtered and evaporated, and the resulting oil was purified by preparative TLC (eluting with dichloromethane) to afford the title compound (35 mg, 14% from 2,3-dichlorotoluene) as a yellow solid. LC/MS m/z 297 [M+H]+. 1H-NMR (CDCl3) delta 7.76 (d, J=10.5, 1H), 6.80 (d, J=10.5, 1H), 4.24-4.16 (m, 1H), 4.14-4.05 (m, 2H), 2.59 (s, 3H), 2.40-2.25 (m, 4H), 1.97-1.81 (m, 4H), 1.55 (s, 1H).

The residue was then purified by TLC technique (3:1 (v/v) petroleum ether/ethyl acetate) to furnish sulfonylbenzotriazoles 3.

Then the mixture is cooled, acidified with hydrochloric acid to pH 3-4, filtered from inorganic impurities, alkalized with 25% ammonium hydroxide to pH 8-9, and evaporated on a rotary evaporator. The technical product obtained is distilled under vacuum and the high-boiling fraction is separated at a temperature of 150 C and pressure of 6 mm of mercury. The fraction is placed into a sealed vessel and kept at room temperature until the substance transition to the crystalline state. After crystallization the crystals are isolated, washed with acetone, and dried in the air. The product yield is 49 %.

LC/MS [M+H]+ = 399, retention time = 3.16 minutes.

The layers were separated and the organic washed with brine, dried and the solvent evaporated to give 1- Chloro-3-iodo-2-trifluoromethyl-benzene (38.3 g, 79%) as an orange solid, which was used without further purification. Water (0.5 mL), intermediate 17 (0.17 g; 0.51 mmol) followed by tetrabutylammonium bromide (41 mg; 0.13 mmol) were added at room temperature to a mixture of potassium hydroxide (0.50 g; 7.63 mmol) in 2-methyltetrahydrofuran (5 mL). The reaction mixture was stirred for 10 minutes at room temperature, then stirred at 50C for 1 hour and (2- chloroethyl)-methylamine hydrochloride (CAS 4535-90-4) (0.119 g; 0.92 mmol) was added. The reaction mixture was stirred for 24 hours at 50C. The reaction mixture was cooled down to room temperature, poured into water and extracted with EtOAc. The organic layer was washed with brine, dried over MgS04, filtered and evaporated to dryness. The residue was purified by chromatography over silica gel (Spherical SiOH, 10muiotaeta, 60g; mobile phase: gradient from 0.5% NH4OH, 97% DCM, 3% MeOH to 0.5% NH OH, 95% DCM, 5% MeOH). The pure fractions were collected and evaporated to give 30 mg (15%) which was taken up Et20 and evaporated to give 29 mg (14%) of compound 44 , MP.: 80C (gum, Kofler).
