A simple, efficient, and metal-free methodology for the preparation of 2-mercaptobenzothiazole and derivatives in excellent yields via microwave-assisted pathway is reported. Our condition provides a convenient protocol for the synthesis of a diverse collection of 2-mercaptobenzothiazoles and 6-(4-substituted-1H-1,2,3-triazol-1-yl)-2-mercaptobenzothiazoles with a very simple purification process. This report provides an alternative protocol for fast access to the wide range of compounds for sequence synthesis and biological studies.

Providing new methods for the selective functionalization of small molecules is highly desirable, because installing molecular diversity in a desired position, for example, allows one to modulate bioactive molecules. This work reports a method for the selective functionalization of anilines using hexafluoroisopropanol (HFIP) as a solvent to promote an acid-catalyzed hydroarylation of olefins. Mechanistic experiments revealed that HFIP both protonates the alkene and selectively enables anilines toward the electrophilic aromatic substitution. This powerful strategy has been applied to the functionalization of the anti-inflammatory mefenamic acid with chemocontrol and regiocontrol.

An efficient and operationally simple protocol has been demonstrated for the synthesis of 1,3,5,7-tetrasubstituted pyrimido[4,5-d]pyrimidines via TBHP-mediated direct oxidative coupling of N-uracil amidines and methylarenes under metal-free conditions. Due to the inherent stability of methylarenes compared to aldehydes, the presented synthetic protocol is adaptable to a broad substrate scope, is operationally simple, has no need for stringent protection in the whole preparation process, and has the potential to prepare valuable products that are currently inaccessible or challenging to prepare using conventional methods. It is a significantly important complement to the conventional synthetic methods. The reaction possesses an efficient tandem oxidation-imination-cyclization process.

Alternatives to the use of pressurized hydrogen (H-2) in chemical syntheses are of growing interest and importance in developing safer protocols. Ammonia borane (AB), with its high hydrogen content and ability to release H-2 under mild conditions, is one potential solution. However, optimization of AB dehydrogenation and further the use of AB as a H-2 source to facilitate tandem one-pot reactions with both high activity and stability is an ongoing research challenge. In this work, monodisperse AuPd nanoparticle (NP) catalysts were synthesized and systematically studied for AB dehydrogenation, and Au-rich NPs were found to be the most active composition for AB methanolysis with a turnover frequency (TOF) of 160 min(-1) surpassing most Pd-based NP catalysts to date. More importantly, Au-rich NPs were also active in the one-pot tandem reduction of a series of nitro-groups and the hydrodehalogenation of mono- and polyhalogenated persistent chemical pollutants under mild reaction conditions using only a 2:1 molar ratio of AB to substrate. The AuPd catalyst was stable after ten reaction runs for AB methanolysis and five reaction runs for tandem one-pot reactions without a loss of activity or change in NP structure and morphology.

A novel oxidative esterification of 1-arylprop-2-en-1-ols with toluene derivatives catalyzed by tetrabutylammonium iodide (TBAI) is reported. The optimization of the reaction conditions illustrates that each of experiment parameters including the catalyst, solvent, and oxidant is significant for present oxidative functionalization. This metal-free protocol has a broad substrate scope including the halogen groups for further functionalization and enriches the reactivity profile of allyl alcohol and toluene derivatives. In addition, this protocol represents a new transformation of allyl alcohol involving C-C bond cleavage and C-O bond forming.

An operationally simple nickel-catalyzed hydroarylation reaction for alkynes is described. This three component coupling reaction utilizes commercially available alkynes and aryl bromides, along with water and Zn. An air-stable and easily synthesized Ni(II) precatalyst is the only entity used in the reaction that is not commercially available. This reductive cross-coupling reaction displays a fairly unusual anti selectivity when aryl bromides with ortho substituents are used. In addition to optimization data and a preliminary substrate scope, complementary experiments including deuterium labeling studies are used to provide a tentative catalytic mechanism. We believe this report should inspire and inform other Ni-catalyzed carbofunctionalization reactions.

A green and efficient method for the synthesis of amides has been developed through oxidative amidation between methylarenes with amines in a two-step continuous flow system. This method integrates methylarene oxidation and amide formation into a single operation which is usually accomplished separately. Oxidation with tert-butyl hydroperoxide (TBHP) as green oxidant, the synthesis of amides under mild reaction conditions in continuous flow system and the utilization of methylarenes as starting material make this methodology novel and environment friendly. The challenging nickel-catalyzed mono-alpha-arylation of acetone with aryl chlorides, pivalates, and carbamates has been achieved for the first time. A nickel/Josiphos-based catalytic system is shown to feature unique catalytic behavior, allowing the highly selective formation of the desired mono-alpha-arylated acetone. The developed methodology was applied to a variety of (hetero)aryl chlorides including biologically relevant derivatives. The methodology has been extended to the unprecedented coupling of acetone with phenol derivatives. Mechanistic studies allowed the isolation and characterization of key Ni(0)and Ni(II)catalytic intermediates. The Josiphos ligand is shown to play a key role in the stabilization of Ni(II)intermediates to allow a Ni-0/Ni(II)catalytic pathway. Mechanistic understanding was then leveraged to improve the protocol using an air-stable Ni(II)pre-catalyst.
