Koller, Sebastian;Klein, Philippe;Reinhardt, Katja;Ochmann, Lukas;Seitz, Antonia;Jandl, Christian;Pothig, Alexander;Hintermann, Lukas research published ã€?New Access Routes to Privileged and Chiral Ligands for Transition-Metal Catalyzed Hydrogen Autotransfer (Borrowing Hydrogen), Dehydrogenative Condensation, and Alkene Isomerization Reactionsã€? the research content is summarized as follows. A group of transition-metal catalyzed hydrogen moving reactions, encompassing hydrogen autotransfer (HAT; also called borrowing hydrogen, BH), dehydrogenative condensation (DHC) and alkene isomerization, displays high atom economy and relies on widely available starting materials. Such reactions have considerable potential for clean reaction design and application in sustainable synthesis. With the aim to develop and study synthetic applications of the title reactions, authors have set up synthetic access routes to a toolbox of structurally varied ligands for and pincer complexes of some transition metals (cobalt, ruthenium, iridium) that are well established for the title reactions. Ligand target structures, for which often improved syntheses have been found, encompass 6,6′-dihydroxy-2,2′-bipyridine, 2(3-hydroxyphenyl)pyridines (as backbones for PCN pincers), 2(6-methylpyridine-2-yl)pyridines (as backbones for PNN pincers) and 2(3-tolyl)pyridines (as backbones for PCN pincers). To support research towards asym. versions of the title reactions, they have prepared asym. modified versions of well-established catalysts, including chiral, enantiopure versions of Milstein’s PNN-ruthenium pincer, Kempe’s triazinyl-diaminophosphanyl PNP-iridium- or -cobalt pincers, Huang’s PCN-iridium pincers, and Grotjahn’s alkene zipper complex. The strategy applied to ‘chiral switching’ relied on replacing sym. dialkylphosphine donor-groups by dimenthylphosphine or aryl(menthyl)phosphine donor units. The resulting ligands or complexes have been structurally characterized, and the catalytic potential of the catalysts has been established in exploratory model reactions (transfer hydrogenation; diol to lactone dehydrogenative condensation; alkene isomerization). Several model reactions have been designed which will allow to study asym. catalytic hydrogen moving reactions.

Antibiotic resistance is a major threat to global health; this problem can be addressed by the development of new antibacterial agents to keep pace with the evolutionary adaptation of pathogens. Computational approaches are essential tools to this end since their application enables fast and early strategical decisions in the drug development process. We present a rational design approach, in which acylide antibiotics were screened based on computational predictions of solubility, membrane permeability, and binding affinity toward the ribosome. To assess our design strategy, we tested all candidates for in vitro inhibitory activity and then evaluated them in vivo with several antibiotic-resistant strains to determine minimal inhibitory concentrations The predicted best candidate is synthetically more accessible, exhibits higher solubility and binding affinity to the ribosome, and is up to 56 times more active against resistant pathogens than telithromycin. Notably, the best compounds designed by us show activity, especially when combined with the membrane-weakening drug colistin, against Acinetobacter baumanii, Pseudomonas aeruginosa, and Escherichia coli, which are the three most critical targets from the priority list of pathogens of the World Health Organization.

An efficient, transition metal-free visible-light-driven continuous-flow C-3-alkylation of quinoxalin-2(1H)-ones had been demonstrated by employing Katritzky salts as alkylating agents in the presence of Eosin-y as a photoredox catalyst and DIPEA as a base at room temperature The present protocol was accomplished by utilizing abundant and inexpensive alkyl amine (both primary and secondary alkyl) and as well as this a few amino acid feedstocks were converted into their corresponding redox-active pyridinium salts and subsequently into alkyl radicals. A wide variety of C-3-alkylated quinoxalin-2(1H)-ones I [R = Bn, 4-ClC6H4CH2, cyclopentyl, etc.; R1 = Me, CH2C≡CH, Bn, etc.; R2 = H, 6-F, 6,7-di-Me] were synthesized in moderate to high yields. Further this environmentally benign protocol was carried out in a PFA (Perfluoroalkoxy alkane) capillary based micro reactor under blue LED irradiation, enabling excellent yields (72% to 91%) and shorter reaction times (0.81 min) as compared to a batch system (16 h).

We report a bottom-up approach to immobilize catalysts into MOFs, including copper halides and gold chloride in a predictable manner. Interestingly, the structures of MOFs bearing NHC metal complexes maintained a similar 4-fold interpenetrated cube. They exhibited exceptionally high porosity despite the interpenetrated structure and showed good stability in various solvents. Moreover, these MOFs possess high size activity depending on the size of the substrates in various reactions, compared to homogeneous catalysis. Also, the high catalytic activity of MOFs can be preserved 4 times without significant loss of crystallinity. Incorporation of the various metal complexes into MOFs allows for the preparation of functional MOFs for practical applications.

Described here is the rhodium(I)-catalyzed tandem addition-cyclization of 1,5-enynes with aryl- and alkenylboronic acids. In the presence of triethylamine and catalytic [Rh(OH)(COD)]2 in methanol, both the terminal and internal alkyne substrates possessing an electron-deficient alkene undergo inter- and intramol. C-C bond formations to yield alkylidene-cyclobutane products. The reaction proceeds through a series of processes involving generation of an aryl or alkenyl rhodium from the organoboronic acid, 1,2-syn-carbometalation of the alkyne and 4-exo-type-cyclization of the resulting alkenyl rhodium intermediate with the tethered, carbonyl-conjugated alkene. Thus, the reaction enables a net R,H-addition across the 1,5-enyne π-system – hydroarylation and hydroalkenylation – under mild rhodium catalysis while achieving 4-membered ring formation. Preliminary studies show that the reaction can also be rendered enantioselective by employing a chiral diene ligand for the rhodium catalyst.

A synthetic strategy for the formation of C(sp3)-N bonds, particularly through a Cu-catalyzed oxidative cross-coupling, is rare. Herein, the authors report a novel synthetic approach for the preparation of tertiary sulfonamides via Cu-catalyzed intra- and intermol. oxidative C(sp3)-N cross-coupling reactions. This method allows the use of the readily available C(sp3)-based pinacol boronate as a substrate and the tolerance of a wide range of functional groups under mild reaction conditions. The success of this strategy relies on the unprecedented additive effects of silanol and NaIO4.

Kim, Byungjun;Kim, Yongjae;Lee, Sarah Yunmi research published ã€?Stereodivergent Carbon-Carbon Bond Formation between Iminium and Enolate Intermediates by Synergistic Organocatalysisã€? the research content is summarized as follows. Herein a stereodivergent method for the Michael addition of aryl acetic acid esters to α,β-unsaturated aldehydes catalyzed by a combination of a chiral pyrrolidine and a chiral Lewis base has been reported. This reaction proceeds through a synergistic catalytic cycle which consists of one cycle leading to a chiral iminium electrophile and a second cycle generating a nucleophilic chiral enolate for the construction of a carbon-carbon bond. By varying the combinations of catalyst enantiomers, all four stereoisomers of the products with two vicinal stereocenters are accessible with high enantio- and diastereoselectivity. The products of the Michael addition, 1,5-aldehyde esters, can be readily transformed into a variety of other valuable enantioenriched structures, including those bearing three contiguous stereocenters in an acyclic system, thus providing an efficient route to an array of structural and stereochem. diversity.
