The retinoid X receptors (RXR) are ligand-activated transcription factors involved in multiple regulatory networks as universal heterodimer partners for nuclear receptors. Despite their high therapeutic potential in many pathologies, targeting of RXR has only been exploited in cancer treatment as the currently available RXR agonists suffer from exceptional lipophilicity, poor pharmacokinetics (PK), and adverse effects. Aiming to overcome the limitations and to provide improved RXR ligands, we developed a new potent RXR ligand chemotype based on the nonsteroidal anti-inflammatory drug oxaprozin. Systematic structure-activity relationship anal. enabled structural optimization toward low nanomolar potency similar to the well-established rexinoids. The structure of this hybrid nanomaterial (CaO@SiO2-NH2-Sal-Zn) was characterized using the anal. techniques such as Fourier transform IR spectroscopy, field emission-SEM, X-ray powder diffraction, energy-dispersive X-ray spectroscopy, transmission electron microscopy, thermogravimetric anal., and inductively coupled plasma at. mass spectrometry. The catalytic activity of this organic-inorganic hybrid nanostructure was investigated through the green synthesis of 4H-pyran derivatives via one-pot three-component condensation reaction which high yields of desired products obtained under green media. The reusability experiments confirmed the stability and high performance of this hybrid nanomaterial under the reaction conditions. The retrievability of this catalyst was proved by recycling six times in reactions without significant loss in its activity. Green conditions, use of available materials, easy work-up procedure, and high yield of products, low reaction times, and simple purification are some advantages of this protocol over earlier ones.

Signal transducer and activator of transcription 3 (STAT3) is a tumorigenic transcription factor that is persistently activated in various human cancers including hepatocellular carcinoma (HCC). Therefore, STAT3 is considered as a prominent target to counteract the uncontrolled proliferation of cancer cells. In the present report, pyrimidine-2,4-diones (N-methyluracil derivatives) (I [R =3-isopropoxyphenyl]-I [R =2-chlorophenyl]) were synthesized in an ionic liquid (BMIm PF6) medium employing a ligand-free Suzuki-Miyaura cross-coupling process. Among the I compound I [R = naphthalen-1-yl] showed good cytotoxicity towards both the tested cell lines and did not display a toxic effect against normal hepatocytes (LO2). MNK8 significantly increased the Sub-G1 cell count in both cell lines and the cytotoxic effect of I [R = naphthalen-1-yl] was found to be mediated through the suppression of constitutive phosphorylation of STAT3Y705. It also decreased the DNA interaction ability of nuclear STAT3 in HCC cells. Two naphthalene-diimide (NDI) bis-imidazolium salts have been used as N-heterocyclic carbene (NHC) precursors for the preparation of NDI-functionalized complexes of rhodium and iridium of general formula [MCl(NDI-NHC)(COD)] (M=Rh, Ir; NDI-NHC=NDI-functionalized NHC ligand). Comparison of the IR spectra of the complexes [IrCl(NDI-NHC)(CO)2] and their related one- and two-electron reduced forms, reveal that each one-electron reduction produces a decrease of the average ν(CO) of 9-10 cm-1, indicating a significant enhancement of the electron-richness of the metal. The [MCl(NDI-NHC)(COD)] complexes were tested in the catalytic cycloisomerization of alkynoic acids. The one-electron reduced forms showed greatly enhanced activities. For the cyclization of 5-hexynoic acid, the two-electron reduction of the ligand produced further enhancement of the catalytic activity, therefore showing that the catalyst can switch between three redox species with three distinct catalytic activities.
