Hematobia irritans (horn fly) infestation in cattle is responsible for over a billion dollars a year in global economic loss due to decreased milk production and lower feed conversion. There is significant need for new insecticidal agents since current treatments such as organophosphates and pyrethroids suffer from field resistance. Isoxazoline oxime ethers represent a new class of γ-aminobutyric acid (GABA) receptor channel blockers which show good activity (LD90 = 1.0 μg/mL) against horn flies in an in vitro feed assay and have demonstrated efficacy (>90% reduction at 1.0 mg/kg) as a topical treatment in a field study.

Trihexyl (tetradecyl) phosphonium tetrafluoroborate was found to be an active catalyst for the introduction of fluoride by nucleophilic aromatic substitution. With a decomposition temperature above 300 °C, this ionic liquid is suitable for reactions at high temperatures The addition of 1 mol% of the ionic liquid relative to KF increased the initial rate for the fluorination of 1,2-dichloro-4-nitrobenzene six-fold compared to the uncatalyzed reaction. The highest reaction rate was observed at 20 mol% ionic liquid relative to KF. Ease of handling without the need for stringent drying conditions and reuseability make this ionic liquid a useful phase transfer catalyst.

4-Anilinoquinazoline derivatives I [R = 2-OBn, 3-OBn, 4-OBn, 2-O(4-NO2Bn), etc.; n = 0, 1, 2] were designed and synthesized using a binding model with multi-target receptor tyrosine kinases and assessed their antitumor activity against five human tumor cell lines (HepG2, A549, MCF-7, DU145, SH-SY5Y). The majority of the compounds inhibited the proliferation of all the cancer cell types, with some compounds displaying selective inhibition. Compounds I [R = 3-OBn, n = 0], I [R = 4-O(4-NO2Bn), n = 0] and I [R = 2-OBn, n = 1] displayed IC50 values of 7.588, 8.619 and 6.936μM, resp., for A549 cells, which were much lower than that of Gefitinib (14.803μM). Compound I [R = 3-O(4-CF3Bn), n = 1] displayed an IC50 value of 2.756μM for DU145 cells. The representative compound I [R = 3-O(4-CF3Bn), n = 1] had unexceptionable broad-spectrum inhibition for all cancer cell types, and demonstrate inhibition of vascular endothelial growth factor receptor 2 (VEGFR-2), platelet-derived growth factor receptor beta (PDGFR-β), and epidermal growth factor receptor (EGFR) with IC50 values of 46.4, 673.6 and 384.8 nM, resp., which were similar to those of Sorafenib for VEGFR-2 and PDGFR-β (140.6 and 582.7 nM, resp.). Mol. docking results supported the mol. level assay results. Data for production of reactive oxygen species and assessment of matrix metalloproteinase corroborated the strong anti-proliferative effect of compound I [R = 3-O(4-CF3Bn), n = 1]. The compound also displayed robust antitumor efficacy and relativity lower toxicity in a xenograft model. These attributes were similar to those of Sorafenib.

The method of Appleton, et al. (CA 48, 3437a), for the determination of choline, through the absorbance of its periodide at 365 mμ, was modified for the determination of other quaternary N compounds (e.g. betaine, trigonelline, carnitine, stachydrine, and γ-butyrobetaine) as each was separated from an extract of corn grain by ion-exchange chromatography (cf. following abstract). Certain N compounds (such as nicotinic acid, histidine, adenine, and thiamine), which give precipitates with I, may occur in some eluates but they do not interfere in this method. As many as 50 tubes of anal. samples containing 10-100 γ of a compound can be handled. Anthranilic acid derivatives are important pharmacophores in drug discovery. Several of them are currently being used, such as mefenamic acid and meclofenamates, possess analgesic, anti-inflammatory and antipyretic activities. Some anthranilic acid-based scaffolds have also been reported for the management of metabolic disorders. The aim of the current study was to investigate the antiglycation potential of 2-anilino benzoic acid derivatives against (N-phenylanthranilic acid) fructose- human serum albumin (HSA) glycation. The study also analyzed the effects of newly identified antiglycation inhibitors on AGEs-mediated intracellular reactive oxygen species production, and associated impaired proliferation of the hepatocytes. The present study focuses on the antiglycation activity of 2- anilinobenzoic acid derivatives 1-18 in in-vitro human serum albumin (HSA)- fructose model. These derivatives were also identified as non-toxic to 3T3 mouse fibroblast cell-line using metabolic assay. The effect of the most promising derivative 1, 2- (2, 4- dinitroanilino)benzoic acid, was studied in a dose dependent manner, co-incubated with fructose-derived AGEs (0- 200 μg/mL), on rat hepatocytes proliferation and associated intracellular generation of ROS via MTT assay and DCFH-DA technique, resp. We found that derivative 1 ameliorates the elevated intracellular oxidative stress and associated diminished proliferation of the hepatocytes in response to AGEs. In conclusion, we identify novel 2- anilino benzoic acid derivatives as antiglycation agents through in-vitro and cellular-based models.

Bacterial quorum sensing refers to the ability of bacteria to control gene expression through the detection of a threshold concentration of certain chems. called autoinducer(s), which are secreted by self and/or other bacteria. Quorum sensing is implicated in the regulation of pathol. relevant events such as biofilm formation, virulence, conjugation, sporulation, and swarming mobility. Inhibitors of bacterial quorum sensing are valuable research tools and potential antimicrobial agents. Here, the authors describe the discovery of several boronic acid inhibitors of bacterial quorum sensing in Vibrio harveyi with IC50 values in the low to sub-micromolar range in whole cell assays.

By modifying ligand steric and electronic profiles it is possible to C-H borylate ortho or meta to substituents in aromatic and heteroaromatic compounds, where steric differences between accessible C-H sites are small. Dramatic effects on selectivities between reactions using B2pin2 or 4,4,5,5-tetramethyl-1,3,2-dioxaborolane (HBpin) are described for the 1st time. Judicious ligand and borane combinations give highly regioselective C-H borylations on substrates where typical borylation protocols afford poor selectivities.

Over the last decade, much attention has been paid to the prevention and treatment of chronic diseases of the oral mucosa. This is primarily due to the increase in the number of patients who seek dental care for diseases of the oral muco-sa (DOM). Currently, due to the lack of special epidemiol. studies, information on the pathol. of the oral mucosa in the literature is almost non-existent. The etiol. and pathogenesis have not been definitively elucidated. It is established that a significant role in the pathogenesis of chronic inflammatory processes belongs to the state of the microbiocenosis of the oral mucosa. One of the most striking examples of domestic drugs of the antioxidant group is thiotriazoline, developed by the staff of the NGO ‘Pharmatron’, as well as specialists of the Department of Pharmaceutical Chem. of Zaporozhye State Medical University under the leadership of professor Mazur I. This drug has an antioxidant and membrane-stabilizing effect. The combined use of several drugs can be complicated by the formation of their supramol. complexes. Therefore, before the introduction of several substances in it is necessary to model the possibility of intermol. interactions between them. For this purpose, methods of quantum chem. are used. We examined the structures and energy characteristics of the complexes, formed thiotriazoline and decamethoxin, also carried out anal. and established at what temperature the formation of a substance from two active components was possible. As a result, the probability of the formation of intermol. hydrogen bonds is reduced, which is very well seen in the trend of interactions between acid and morpholine. Quantum chem. study of a two-component system consisting of thiotriazoline and decamethoxine showed that the most energetically advantageous three-component complexes have a sufficiently low interaction energy of thiotriazoline and decamethoxine. In addition, the data that are provided in the anal. of the tree diagram suggest that in the technol. process in the manufacture of dosage forms, it is advisable to use a temperature not higher than 115°C.
