A formal C-H carboxylation of unactivated arenes e.g., I using CO2 in green solvents is described. The present strategy combines a sterically controlled Ir-catalyzed C-H borylation followed by a Cu-catalyzed carboxylation of the in situ generated organoboronates. The reaction is highly regioselective for the C-H carboxylation of unactivated arenes e.g., I (1,3-disubstituted and 1,2,3-trisubstituted benzenes, 1,2- or 1,4-sym. substituted benzenes, fluorinated benzenes and different heterocycles). The developed methodol. was applied to the late-stage C-H carboxylation of com. drugs and ligands.

Screening tests of insecticidal activity for ∼50 organophosphorus compounds used the sucking insect, green peach aphid, and the chewing insect, tobacco cutworm. The established systemic insecticides were usually highly toxic to the aphid while being less toxic to the cutworm. One of the thiophosphate esters, ((EtO)2P(S)SCH2CO2(CH2)2Cl) was toxic to the cutworm. Five phosphate esters were toxic to both insects. O,O-Di-Me dichlorohydroxyethanephosphonate and its Ac ester, O,O-di-Me trichlorohydroxyethanephosphonate and its Et Me homolog were more toxic to the cutworm than the aphid. The test plant was cabbage and the test chems. were formulated with Me2CO-C6H6-emulsifier 30:30:30% weight/weight Insecticidal activity was indicated by mortality at concentrations of 1000 ppm. or lower.

The first examples of selective and regiodivergent chlorination of unsym. benzyl-substituted epoxides, I [R = 4-methoxyphenyl, Ph, 2H-1,3-benzodioxol-5-yl, 1-[(4-methylbenzene)sulfonyl]-2,3-dihydro-1H-indol-5-yl, etc.; R1 = Pr, 4-methoxyphenyl, 4-cyanobutyl, 4-(benzyloxy)butyl, etc.] have been reported. These reactions are enabled by the dual role of SnCl4 and TiCl4 as Lewis acids and chloride nucleophiles. Reagent control dictates addition of chloride at either the substituted internal position (SnCl4) or unsubstituted terminal position (TiCl4) of the phenonium ion. These reactions are highly selective, stereospecific, and operationally simple, and proceed in good to excellent yields. Diverse product utility is demonstrated.

In the aftermath of WWII large amount seized German chem. munitions were dumped in the Baltic Sea by Allied forces. In this work, we have compared the chem. content of the solidified blocks of dumped WWII mustard gas collected from the Baltic Sea with solid precipitate from stored mustard gas, known as heel. We have identified the same cyclic sulfonium ions in both samples. In assessing the environmental and toxicol. impact of dumped sulfur mustard munitions on the worlds oceans the potential risk posed by cyclic sulfur mustard salts have so far not been incorporated. The toxicity of 1-(2-chloroethyl)-1,4-dithiane and its hydrolysis product 1-(2-hydroxyethyl)- 1,4-dithiane was evaluated using three different cell lines. Their effect on released pro-inflammatory cytokines was also measured. The toxicity tests showed low toxicity and low pro-inflammatory response and we therefore conclude that the environmental threat posed by these compounds is low.

A comparative structural study on sodium and potassium N-chloroarenesulfonamidates was carried out using x-ray diffraction and IR spectroscopy as exptl. techniques. As shown by crystallog. studies, the sodium ions tend to incorporate more water oxygen atoms in their coordination spheres than the potassium ions, while the potassium congeners prefer the interaction with sulfonamidate moieties. The marked dissimilarity between the compositions of the coordination spheres as well as the different tendency of the sodium and potassium salts to absorb moisture from the air were attributed to the different sizes of the sodium and potassium ions. The opposite shifts of the ν as(SO2) and ν(SN) bands upon dehydration were ascribed to an increased polarization of the sulfonyl group by the metal centers. Based on IR spectroscopic observations, a weakening of the N-Cl bonds is suspected in the water-free compounds, which may contribute to the known thermal instability of the dehydrated salts of N-chloroarenesulfonamides. Various sections of IR spectra as well as x-ray powder diffraction patterns proved to be suitable to identify the water-free and hydrated samples.

The use of enantiopure, sulfonated SPhos (sSPhos), an existing ligand that has until now been used only in racemic form and that derived its chirality from an atropisomeric axis that was introduced through sulfonation. The attractive noncovalent interactions involving the ligand sulfonate group was responsible for the high levels of asym. induction that we obtain in the 2,2′-biphenol products of Suzuki-Miyaura coupling, and a highly practical resolution of sSPhos via diastereomeric salt recrystallization was developed.

In order to discover novel probes that may help in the investigation and control of infectious diseases through a new mechanism of action, we have evaluated a library of phenol-based natural products (NPs) for enzyme inhibition against four recently characterized pathogen β-family carbonic anhydrases (CAs). These include CAs from Mycobacterium tuberculosis, Candida albicans, and Cryptococcus neoformans as well as α-family human CA I and CA II for comparison. Many of the NPs selectively inhibited the mycobacterial and fungal β-CAs, with the two best performing compounds displaying submicromolar inhibition with a preference for fungal over human CA inhibition of more than 2 orders of magnitude. These compounds provide the first example of non-sulfonamide inhibitors that display β over α CA enzyme selectivity. Structural characterization of the library compounds in complex with human CA II revealed a novel binding mode whereby a Me ester interacts via a water mol. with the active site zinc.
