Comparison of pKa values for the ammonia acids Me3N+(CH2)nCO2H ClO4- (n = 1, 2, 3), I (R = 2-, 3-, 4-CO2H), and II (same R) with the pKa values for the homomorphous acids Me3C(CH2)nCO2H (n = 1, 2, 3), III (R = 2-, 3-, 4-CO2H) and IV (same R) permits an evaluation of the steric effect of the Me3N+ group. The pKa values, corrected for the steric effect, correlate with the reciprocal distance between the pos. N atom and the dissociable proton. Thus, the acidity of these acids is determined by the field effect, not the inductive effect, of the Me3N+ groups.

An n-octylmethyl, diphenyl-polysiloxane called SOP-50-Octyl was prepared by a condensation reaction of bis(dimethylamino) n-octylmethylsilane with diphenylsilanediol. The resulting copolymer was a gum with high mol. weight and was obtained with a yield of 80%. 1H and 29Si NMR spectroscopy revealed that the copolymer was a 52% octylmethyl, 48% diphenyl-polysiloxane with random microstructure. Small cyclic impurities could be almost quant. removed via a purification step. SOP-50-Octyl was used as stationary phase for the preparation of wall coated open tubular fused silica capillary columns for gas-liquid chromatog. The capillary columns exhibited high separation efficiency and high inertness. The stationary phase offered a unique selectivity due to its unique composition The Rohrschneider-McReynolds constants indicated a low overall polarity in spite of the high Ph content, as the polarity was distinctly decreased by the octyl substituent. Also, the octyl substituent was responsible for a high column bleed, reducing the maximum allowable operating temperature to 280°. The elution temperatures of apolar compounds were increased due to increased interaction of the octyl substituent with the analytes. Some applications with volatile and semi-volatile organic compounds illustrate that SOP-50-Octyl is an excellent choice for confirmational analyses.

The kinetic investigation of homogeneously Ir(III) chloride catalyzed oxidation of Ethylene glycol by Chloramine- T [CAT] in perchloric acid medium has been carried out in the temperature range of 30 to 45°C. The reaction was carried out in the presence of mercuric acetate as a scavenger for chloride ion. The reaction exhibits first order kinetics with respect to the oxidant [CAT] and catalyst [Ir(III)] while zero order with respect to substrate, i.e., Ethylene glycol (EG) was observed The reaction shows negligible effect of [Hg(OAc)2], [H+] and ionic strength of the medium. Chloride ion pos. influenced the rate of reaction. The various activation parameters have been calculated from the rate measurements at different temperatures (30 to 45°C). The product of the reaction has been identified as the corresponding monocarboxylic acid. The reaction between Chloramine-T and Ethylene glycol in acid medium shows 2:1 stoichiometry. On the basis of kinetic studies, reaction stoichiometry and product anal., a suitable mechanism has been proposed and rate law has been derived.

The mechanism of the reaction of P(OEt)3 (I) with CCl4 (II) was studied. I reacts at 80° with II to form mostly (EtO)2P(O)CCl3 and EtCl. Trichloromethyl radical is trapped by BHT. The reaction is initiated by UV light (254 nm) and a charge-transfer complex is formed between I and II, and the reaction is inhibited by TCNQ. Tris(cyclopropylmethyl) phosphite and tri(1-hexene-6-yl) phosphite are used as potential radical clocks in these reactions. The first leads to CH2:CHCHClMe and the second to CH2:CHCH2CH2CHClMe. The first suggests a radical mechanism (but also may be rationalized by an ionic mechanism), but not the second. Electrochem. oxidation of I with added CCl4 does not account for a radical chain process as the main pathway. Marcus anal. of the reaction viewed as an electron transfer gives a calculated rate constant in the range of 10-20 M-1s-1. Thus, the thermal reaction is in fact an SNCl+ substitution. The radical intermediates are derived from the electron-transfer reaction between CCl3 and CCl4. The reaction of I with II is an example where the observed paramagnetic species during a donor (D)/acceptor (A) interaction suggests an electron-transfer between D and A where in fact they originate from an interaction between A and an electron donor formed after or during the first step of the reaction.

Pharmacol. inhibition of cyclin-dependent kinases has emerged as a possible treatment option for various cancer types. Authors recently identified substituted imidazo[1,2-c]pyrimidin-5(6H)-ones as inhibitors of cyclin-dependent kinase 2 (CDK2). Here, the synthesis of derivatives modified at positions 2, 3, 6 or 8 prepared using Suzuki-Miyaura cross-coupling, halogenation, Dimroth-type rearrangement and alkylation as the main synthetic methods was reported. The compounds displayed micro- to submicromolar inhibition of CDK2/cyclin E activity. Binding of the most potent compound I to CDK2 was determined using isothermal titration calorimetry. The co-crystal structure of I in complex with fully active CDK2 was solved, revealing the binding mode of I in the ATP pocket and a hydrogen bonding interaction with hinge region residue Leu83. Evaluation against leukemia cell lines revealed low cytotoxicity, which is in line with the high selectivity towards CDK2. This study demonstrates that substituted imidazo[1,2-c]pyrimidines can be exploited for future kinase inhibitor development.

Oxidation of IV with Na-OCl at 85° (1.5 hrs.) gives 97% I; if the excess NaOCl is not removed before acidification, there results some 3-chloro-2,6-dimethoxybenzoic acid (V), m. 132°; 2 g. V and AlCl3 in C6H6, refluxed 4 hrs., give 1.65 g. 3-chloro-2,6-dihydroxybenzoic acid, m. 193°, blue color with FeCl3. 2,6-(HO)2C6H3Me (50 g.) in NaOH, treated with Me2SO4 and heated 2 hrs. on the water bath, gives 57 g. 2,6-(MeO)2C6H3Me; oxidation with KMnO4 (1 hr. at 80°) gives 73% I. The preparation of II in 51% yield from 1.58 g. resorcinol bis(tetrahydro-2-pyranyl) ether and BuLi by the method of Parham and Anderson (C.A. 43, 2203h) is reported.
