The fungicide prochloraz was subjected to degradation by the pathogen causing rice Bakanae disease, Fusarium fujikuroi, in order to gain an insight into the mechanisms of sensitivity and resistance to the fungicide. Growth-inhibiting assays of pathogens were conducted on potato dextrose agar (PDA) plates by a paper-disk agar-diffusion method. Significant growth inhibition of the sensitive strain CF106 was observed at the recommended treatment level of prochloraz, whereas negligible growth inhibition of the resistant strain CF245 was observed at the same treatment level. The strain CF245 was shown to be able to grow on PDA with 500 mg/L of the fungicide, which is significantly higher than its recommended treatment level. Growth-inhibiting assays of pathogens were also conducted in potato dextrose broth (PDB) medium supplemented with prochloraz at different concentrations, measuring their biomass weights over the incubation period. Significant growth inhibition was observed in the strain CF106 at a level of 0.5 mg/L, but negligible growth inhibition was observed in the strain CF245 at the same treatment level with the strain CF106. The strain CF245 could grow in PDB supplemented with 1.0 mg/L of prochloraz. The degradation of prochloraz by the 2 strains was evaluated by gas liquid chromatog. and mass spectrometry analyses. The strain CF245 completely degraded 1.0 mg/L prochloraz in 5 days after incubation, whereas no degradation of prochloraz was observed by the strain CF106 at the same treatment level. Liquid chromatog. Q-TOF MS detected N-(2-(2,4,6-trichlorophenoxy)ethyl)propan-1-amine as a major degradation product of prochloraz by the strain CF245. These results indicated that the degradation of prochloraz may account for the reduced sensitivity of the strain CF245 to prochloraz.

To explore the effect of substituents on Ph motif on sulfonyl function of novel anticancer 4-phenyl-1-benzenesulfonylimidazolidinones, electron donating or withdrawing substituents were introduced at 3 or 4-position and the analogs were tested against human lung (A549) and colon (HCT-15) cancer cell lines. Quant. structure activity relation of the 4-substituted series shows that only STERIMOL L values are well correlated. The increment of substituent’s volume enhances the activity against both cell lines. The small substituent at 3-position addnl. increases the activity. However naphthyl group in place of Ph reduces the activity. Therefore the Ph motif with sterically large substituent at 4-position and small substituent at 3-position may be important for their activity. Integration of these substituents’ effects into the structural design led to discover the more potent analog, 4-phenyl-1-(N-acetylindoline-5-sulfonyl)imidazolidinone.

Benzofuroquinolinediones (7c and 7d) were synthesized by base-catalyzed condensation of dichloroquinolinediones with phenolic derivatives Their dialkylaminoalkoxy derivatives (8i-8p) were prepared by reaction with various dialkylaminoalkyl chlorides. The cytotoxicity of the synthesized compounds was evaluated against eight types of human cancer cell lines, and their topoisomerase II inhibition was assessed. In general, the cytotoxicity of benzofuroquinolinediones (8i-8p) was similar or superior to that of doxorubicin and showed more potent inhibitory activity than naphthofurandiones (8a-8h). Also, most of the compounds exhibited excellent topoisomerase II inhibitory activity at a concentration of 5 μM and two compounds, 8d and 8i, showed IC50 values of 1.19 and 0.68 μM, resp., and were much more potent than etoposide (IC50 = 78.4 μM), but similar to doxorubicin (IC50 = 2.67 μM). However their inhibitory activity on topoisomerase I was lower, and 8d and 8i showed IC50 values of 42.0 and 64.3 μM, resp.

The growing interest in regenerative medicine has created a need for superior polymer matrixes that suit multiple phys., mech., and biol. requirements. While the phospholipid bilayer of a cell membrane is considered optimal for interacting with biologics, polymeric materials composed of 2-methacryloyloxyethyl phosphorylcholine (MPC) offer a cell membrane-like synthetic alternative. In this work, thiol-containing phosphorylcholine polymers were synthesized by radical copolymerization of a lipoic acid-functionalized methacrylate with MPC. The canonical cell adhesion oligopeptide (GRGDS) was incorporated into the polymers by copolymerization of a GRGDS-containing methacrylamide prepared by solid phase peptide synthesis. The relative amounts of phosphorylcholine, lipoic acid and oligopeptide were controlled by the monomer feed ratios, and the polymers were characterized by NMR spectroscopy and aqueous gel permeation chromatog. (GPC). These multifunctional polymers formed hydrogels rapidly (<10 min) by Michael addition when poly(ethylene glycol)diacrylate (PEGDA) was added at pH 9 – an initiator-free gelation performed in a completely aqueous environment. Two cell lines, live mouse skeletal muscle myoblasts (C2C12) and human ovarian cancer (SKOV3) cells, were observed to specifically attach, spread and proliferate only on hydrogels containing the GRGDS peptide sequence, with a notable dependence on peptide concentration The remarkable hydrophilicity and biocompatibility attributed to polyMPC combined with the facile gelation conditions of these polymers affords a platform of new bio-cooperative materials suitable for cell studies.

A fast and efficient microwave-assisted solid phase peptide synthesis (MW-SPPS) of a 51mer peptide, the main heparin-binding site (60-110) of human pleiotrophin (hPTN), using 2-chlorotrityl chloride resin (CLTR-Cl) following the 9-fluorenylmethyloxycarbonyl/tert-Bu (Fmoc/tBu) methodol. and with the standard N,N’-diisopropylcarbodiimide/1-hydroxybenzotriazole (DIC/HOBt) coupling reagents, is described. An MW-SPPS protocol was for the first time successfully applied to the acid labile CLTR-Cl for the faster synthesis of long peptides (51mer peptide) and with an enhanced purity in comparison to conventional SPPS protocols. The synthesis of such long peptides is not trivial and it is generally achieved by recombinant techniques. The desired linear peptide was obtained in only 30 h of total processing time and in 51% crude yield, in which 60% was the purified product obtained with 99.4% purity. The synthesized peptide was purified by reversed phase high performance liquid chromatog. (RP-HPLC) and identified by electrospray ionization mass spectrometry (ESI-MS). Then, the regioselective formation of the two disulfide bridges of hPTN 60-110 was successfully achieved by a two-step procedure, involving an oxidative folding step in dimethylsulfoxide (DMSO) to form the Cys77-Cys109 bond, followed by iodine oxidation to form the Cys67-Cys99 bond.

The biopharmaceutical properties of supramol. vesicular aggregates (SVAs) were characterized with regard to their physicochem. features and compared with cationic liposomes (CLs). Neutral and cationic SVAs were synthesized using two different copolymers of poly(aspartyl hydrazide) by thin-layer evaporation and extrusion techniques. Both copolymers were self-assembled in pre-formulated liposomes and formed neutral and cationic SVAs. Gemcitabine hydrochloride (GEM) was used as an anticancer drug and loaded by a pH gradient remote loading procedure, which significantly increased drug loading inside the SVAs. The resulting average size of the SVAs was 100 nm. The anticancer activity of GEM-loaded neutral and cationic SVAs was tested in human alveolar basal epithelial (A549) and colorectal cancer (CaCo-2) cells. GEM-loaded cationic SVAs increased the anticancer activity in A549 and CaCo-2 cells relative to free drug, neutral SVAs, and CLs. In vivo biodistribution in Wistar rats showed that cationic SVAs accumulate at higher concentrations in lung tissue than neutral SVAs and CLs. Cationic SVAs may therefore serve as an innovative future therapy for pulmonary carcinoma.

The fish embryo acute toxicity (FET) test with the zebrafish (Danio rerio) embryo according to OECD TG 236 was originally developed as an alternative test method for acute fish toxicity testing according to, e.g., OECD TG 203. Given the versatility of the protocol, however, the FET test has found application beyond acute toxicity testing as a common tool in environmental hazard and risk assessment. Whereas the standard OECD guideline is restricted to four core endpoints (coagulation as well as lack of somite formation, heartbeat, and tail detachment) for simple, rapid assessment of acute toxicity, further endpoints can easily be integrated into the FET test protocol. This has led to the hypothesis that an extended FET test might allow for the identification of different classes of toxicants via a “fingerprint” of morphol. observations. To test this hypothesis, the present study investigated a set of 18 compounds with highly diverse modes of action with respect to acute and sublethal endpoints. Especially at higher concentrations, most observations proved toxicant-unspecific. With decreasing concentrations, however, observations declined in number, but gained in specificity. Specific observations may at best be made at test concentrations ≤ EC10. The existence of a “fingerprint” based on morphol. observations in the FET is, therefore, highly unlikely in the range of acute toxicity, but cannot be excluded for experiments at sublethal concentrations

Cinnamon is an important spice crop that is widely cultivated in tropical regions. In this study, the antibacterial activities of four accessions of cinnamon essential oil (CEO) belonging to three different species (CEO1 and CEO4, Cinnamomum cassia Presl. (Lauraceae) bark; CEO2, Cinnamomum zeylanicum Blume (Lauraceae) bark; CEO3, Cinnamomum burmannii Blume (Lauraceae) bark) toward Salmonella enterica subsp. enterica serovar pullorum (S. pullorum) were evaluated by microdilution assay and kinetic anal. The min. inhibitory concentration (MIC) of the CEOs were all 0.31 mg/mL, and kinetic anal. suggested that the lag phase and maximum specific growth rate of bacteria were concentration dependent. Furthermore, to explore the synergistic antibacterial effects between main components and minor components, the volatile constituents of CEOs were determined by gas chromatog.-mass spectrometry (GC-MS). Cinnamaldehyde (CM) (57.73 %-91.79 %) was the principal constituent in CEO1-CEO4 (p < 0.05), with CEO3 having the highest CM contents. A synergistic effect against S. pullorum was observed when CA was combined with CM. To explore the potential synergistic mechanism, the membrane glycerophospholipid (GPL) composition of S. pullorum was characterized by ultra-performance liquid chromatog.-tandem mass spectrometry (UPLC-MS/MS). CM, CA, and their combination regulated the levels of most phosphatidylethanolamines (PEs), phosphatidylglycerols (PGs), phosphatidic acids (PAs), and some cardiolipins (CLs).
