Glioblastoma is the most common primary brain tumor in adults and still remains incurable, due to the limited accumulation of drugs in the tumor area. Herein, iRGD-modified nanoparticles, DOX@MSN-SS-iRGD&1MT, are developed for simultaneous delivery of chemotherapeutic agents (doxorubicin, DOX) and immune checkpoint inhibitor (1-methyltryptophan, 1MT) into orthotopic glioma. The nanoparticles are comprised of mesoporous silica nanoparticles loaded with DOX, combined with Asp-Glu-Val-Asp (DEVD) connected 1MT, and finally modified by iRGD. These nanoparticles show the capability of penetrating through blood brain barrier into the tumor area, and significantly improve accumulation of drugs in orthotopic brain tumors with minimal side effects. The nanoparticles also activate cytotoxic CD8+ T lymphocytes and inhibit CD4+ T cells in both GL261 cells cocultured with splenocytes in vitro and GL261-luc orthotopic tumors in vivo. Moreover, the expression of antitumor cytokines IFNα/β, IFN-γ, TNF, IL-17, STING, and GrzB is upregulated while protumor proteins p-STAT3 and IL-10 are downregulated in the brain tumor area. This study demonstrates the advantages of chemo-immunotherapeutic nanoparticles accumulated in the brain tumor area and their effectively inhibiting tumor proliferation, which establishes a delivery platform to promote antitumor immunity against glioblastoma.

The photodissociation of 2,3-dibromopropionyl chloride (CH2BrCHBrC(O)Cl, 2,3-DBPC) at 248 nm was carried out to study Br2 as the primary mol. product in the B3Π+0u â†?X1Σ+g transition using cavity ring-down absorption spectroscopy. The rotational spectra (v” = 0-2) were acquired and assigned with the aid of spectral simulation. It is verified that the obtained Br2 fragment is attributed to the one-photon dissociation of 2,3-DBPC and is free from contributions of secondary reactions. The vibrational ratio of the Br2 population of v(0):v(1):v(2) is equal to 1:(0.58 ± 0.12):(0.23 ± 0.09), corresponding to the Boltzmann vibrational temperature of 623 ± 38 K. The quantum yield of Br2 eliminated from 2,3-DBPC is estimated to be 0.09 ± 0.04. The dissociation pathways of 2,3-DBPC and its potential energy surfaces were calculated using d. functional theory. By employing the CCSD(T)//M062X/6-31+g(d,p) level of theory, transition state barriers and corresponding reaction energies were calculated for the Br, Cl, Br2, BrCl, HBr and HCl elimination channels. The unimol. rate constant for Br2 elimination was determined to be 2.09 x 105 s-1 using Rice-Ramsperger-Kassel-Marcus (RRKM) theory, thus explaining the small quantum yield of the Br2 channel.

NEDDylation pathway regulates multiple physiol. process, unlike inhibitors, NEDDylation activators are rarely studied. Novel amide derivatives were synthesized and evaluated for antiproliferative activity against MGC803, MCF-7 and PC-3 cells. Among them, VII-31 displayed the most potent activity with an IC50 value of 94 nmol/L against MGC803 cells. Cellular mechanisms elucidated that VII-31 inhibited the cell viability, arrested cell cycle at G2/M phase and induced apoptosis via intrinsic and extrinsic pathways against MGC803 cells. In addition, VII-31 activated NAE1-Ubc12-Cullin1 NEDDylation via interacting with NAE1 directly. Furthermore, the activation of NEDDylation resulted in the degradation of inhibitor of apoptosis proteins (IAPs). Importantly, VII-31 inhibited tumor growth in xenograft models in vivo without the apparent toxicity. In summary, it is the first time to reveal that VII-31 deserves consideration for cancer therapy as a NEDDylation activator.

Recycling of valuable metals from spent catalysts in a green way is gaining extensive interest for economic and environment reasons. In this study, we developed novel hydrophobic deep eutectic solvents to extract Mo from spent catalysts. The hydrophobic DESs have been designed and synthesized by mixing one molar of the quaternary ammonium salt and two molars of various saturated fatty acids with different carbon chain lengths. The extraction ability and extraction mechanism of these DESs were studied, some factors influencing the extraction efficiency, including the structure of hydrogen bond acceptors and hydrogen bond donors, initial aqueous pH, reaction time and temperature, phase ratios were investigated. It is found that the synthesized hydrophobic DESs exhibit excellent extraction performance toward Mo, where the Mo distribution ratio is more than 2200 in the presence of other metals, corresponding to an extraction efficiency of 99% at optimal reaction conditions. This work reveals a distinct class of materials, guiding an effective and green way for spent catalyst treatment. Implications: Novel hydrophobic deep eutectic solvents have been developed to extract Mo from spent catalysts, the synthesized hydrophobic DESs possess several advantages, such as green, low price, low toxicity, and biodegradability. It exhibits excellent extraction performance under an optimized extraction condition. This work reveals a distinct class of materials, guiding a promising way for green and economical utilization of spent catalysts.

The present method is complementary to the previously reported catalytic enantioselective methods in terms of ylides used, and trans products, e.g., I, were exclusively obtained in good yield (96-73%) and high enantioselectivity (99-84% ee). Use of biphenyldiol and NaI additive was essential to achieve high enantioselectivity.

A series of thiadiazol-5-one derivatives I (Ar = 3,4-(Cl)2C6H3, 4-FC6H4, 4-MeC6H4, etc.) were synthesized by a simple method and their structures were established by phys. and spectroscopic methods. Log P value and in vitro hydrolysis, in simulated gastric fluid and simulated intestinal fluid, of all the compounds were determined by standard methods. Synthesized 1,2,4-thiadiazolines were screened for their anticonvulsant activity against maximal electroshock method and isoniazid induced seizures and all the compounds showed good anticonvulsant activity. The compound I (Ar = 3,4-(Cl)2C6H3) was found to be the most potent member of the series. Mol. docking studies of the synthesized compounds depicted their stable ligand-receptor complex conformation with the typical binding pocket of γ-aminobutyric acid A receptor protein. Compound I (Ar = 3,4-(Cl)2C6H3) confined its effect in the mol. docking studies also by non-covalent interactions with Tyr 157, Phe 200 and Tyr 205, the key interacting residues of γ-aminobutyric acid A receptor protein 4COF. In silico absorption, distribution, metabolism and elimination performance of all the compounds also appear to favor anticonvulsant effect. “LAZAR” and “OSIRIS” property explorer predicted nontoxic, nonmutagenic, noncarcinogenic, etc. nature for all the compounds In conclusion, some γ-aminobutyric acid A receptor agonists have been synthesized in this study with promising drug-like properties, which merit further development.
