Adsorption of a diverse set of chemicals onto goethite was evaluated by column chromatography. The pH of the effluents was 4.7-5.2. Van der Waals forces dominate the exothermic adsorption of 8 nonpolar compounds (e.g., PAHs and chlorobenzenes). H-bonding is responsible for the adsorption of 32 mono-carboxylic acids (i.e., benzoic acids, naphthoic acids and acidic pharmaceuticals) and their adsorption tends to be endothermic. Steric effects significantly decreased the bonding of monocarboxylic acids with ortho-substitutions. Exothermic adsorption of 10 monophenols is controlled by weak H-bonding. Bonding of these 50 solutes onto goethite is totally reversible. In contrast, inner-sphere complexation of phthalic acid and chlortetracycline with goethite occurred according to their low desorption ratio (1.1%-54.4%). Polyparameter linear free energy relationship (PP-LFER) models were established to provide acceptable fitting results of the goethite-solute distribution coefficients (RMSE = 0.32 and 0.30 at 25 degrees C and 5 degrees C, respectively). In this paper, a comprehensive method for the identification of the unknown halogenated DBPs (X-DBPs, X = CI, Br, and I) in the tap water of Wuhan, China via liquid chromatography-high resolution mass spectrometry (LC-HRMS) was developed. 123 X-DBPs were identified through the stepwise procedure, 94 of them were newly identified, and 3 of them were confirmed by standards. Most X-DBPs were aliphatic compounds and highly unsaturated and phenolic compounds, some X-DBPs contained multiple halogen atoms and rich in carboxyl groups, such as C2H2O2BrCl, C2H2O2Br2, and C2H2O2ClI. It was worth noting that the concentration of some X-DBPs had the same trend with time. Most CI-DBPs remained stable and I-DBPs were detected occasionally by monitoring the change of concentration of these X-DPBs with the time during three consecutive months. The results demonstrate that the proposed method could provide valuable molecular formula and structure information on unknown multiple halogenated DBPs, or be used for the identification of other multiple halogenated organic compounds in different media. A series of new Pd(II) complexes with simple structures were designed and synthesized for Suzuki-Miyaura coupling reactions of aryl chlorides. The new Pd(II) complexes contain bidentate amine ligands, and their structures were characterized by single-crystal X-ray diffraction. They are highly efficient for Suzuki-Miyaura coupling reactions of aryl chlorides with low catalyst loadings (0.01 mol %) in aqueous media at room temperature. Two possible reaction pathways involving a Pd-II(/0/)II and a Pd-II(/IV/II) catalytic cycle are proposed, and the mechanism was further investigated using density functional theory (DFT) calculations.

A series of novel N-phenylbenzamide-4-methylamine acridine derivatives were designed and synthesized based initially on the structure of amsacrine (m-AMSA). Molecular docking suggested that the representative compound 9a had affinity for binding DNA topoisomerase (Topo) II, which was comparable with that of m-AMSA, and furthermore that 9a could have preferential interactions with Topo I. After synthesis of 9a and analogues 9b-9f, these were all tested in vitro and the synthesized compounds displayed potent antiproliferative activity against three different cancer cell lines (K562, CCRF-CEM and U937). Among them, compounds 9b, 9c and 9d exhibiting the highest activity with IC50 value ranging from 0.82 to 0.91 mu M against CCRF-CEM cells. In addition, 9b and 9d also showed high antiproliferative activity against U937 cells, with IC50 values of 0.33 and 0.23 mu M, respectively. The pharmacological mechanistic studies of these compounds were evaluated by Topo I/II inhibition, western blot assay and cell apoptosis detection. In summary, 9b effectively inhibited the activity of Topo I/II and induced DNA damage in CCRF-CEM cells and, moreover, significantly induced cell apoptosis in a concentration-dependent manner. These observations provide new information and guidance for the structural optimization of more novel acridine derivatives.

The second ligand 4,4′-bipyridine (4,4′-bpy) was introduced into the above reaction systems to provide four new lanthanide-organic CPs [Ln(2)(2,4-DCB)(6)(4,4′-bpy)(H2O)(2)] [Ln = Eu (5), Gd (6), Tb (7), Dy (8)]. Single-crystal and powder X-ray diffraction analyses reveal that CPs 1 and 2 possess similar one-dimensional (1D) structures, and CPs 2-4 and 5-8 are isostructural, respectively. In 1-4, the 2,4-DCB anions bridge the Ln(III) ions to form a 1D chain. In 5-8, six 2,4-DCB anions link two Ln(III) ions into a binuclear Ln(2)(2,4-DCB)(6) unit, and the Ln(2)(2,4-DCB)(6) units are further connected by the 4,4′-bpy co-ligands to generate a 1D chain. The photoluminescent and magnetic properties of 1-8 were investigated.

Mycobacterium tuberculosis (M.tb), the etiologic agent of tuberculosis, remains the leading cause of death from a single infectious agent worldwide. The emergence of drug-resistant M.tb strains stresses the need for drugs acting on new targets. Mycolic acids are very long chain fatty acids playing an essential role in the architecture and permeability of the mycobacterial cell wall. Their biosynthesis involves two fatty acid synthase (FAS) systems. Among the four enzymes (MabA, HadAB/BC, InhA and KasA/B) of the FAS-II cycle, MabA (FabG1) remains the only one for which specific inhibitors have not been reported yet. The development of a new LC-MS/MS based enzymatic assay allowed the screening of a 1280 fragment-library and led to the discovery of the first small molecules that inhibit MabA activity. Densities (rho), ultrasonic speeds (u) and dynamic viscosity (eta) of the binary 2-chlorotoluene or 4-chlorotoluene or 1,3-dichlorobenzene (1) + n-hexane (2) mixtures were measured at (T = 298.15-318.15) K and atmospheric pressure. Using these measurements, excess volume (V-m(E)), excess ultrasonic speed (u(E)) and deviation in dynamic viscosity (Delta eta) were calculated. Excess isentropic compressibility (kappa(E)(s)) was calculated using rho and u values. The VmE values have been interpreted by Prigogine-Flory-Patterson (PFP) theory. The VmEobtained using PFP theory agree well with experimental values. The u values were also calculated using various empirical relations. The available volume (V-a) and intermolecular free length (L-f) were calculated using experimental u values employing Jacobson free length theory. The Delta eta values were coupled with V-m(E) values to understand intermolecular interaction. Further, Bloomfield and Dewan model was applied to calculate eta and Delta eta values of the studied binary mixture and calculated values agree well with experimental values. (C) 2020 Elsevier B.V. Malaria remains one of the deadliest diseases in the world today. Novel chemoprophylactic and chemotherapeutic antimalarials are needed to support the renewed eradication agenda. We have discovered a novel antimalarial acridone chemotype with dual-stage activity against both liver stage and blood-stage malaria. Several lead compounds generated from structural optimization of a large library of novel acridones exhibit efficacy in the following systems: (1) picomolar inhibition of in vitro Plasmodium falciparum blood-stage growth against multidrug-resistant parasites; (2) curative efficacy after oral administration in an erythrocytic Plasmodium yoelii murine malaria model; (3) prevention of in vitro Plasmodium berghei sporozoite-induced development in human hepatocytes; and (4) protection of in vivo P. berghei sporozoite-induced infection in mice. This study offers the first account of liver-stage antimalarial activity in an acridone chemotype. Details of the design, chemistry, structure-activity relationships, safety, metabolic/pharmacokinetic studies, and mechanistic investigation are presented herein.

BACKGROUND X-linked agammaglobulinemia is a primary immunodeficiency disease caused by gene mutations of Bruton’s tyrosine kinase (BTK). We found a new mutation point and summarized the correlation analysis and performed a literature review. CASE SUMMARY The proband was a 5-year-old boy. He was admitted to our hospital due to a recurrent cough and a fever that had persisted for a month. He had a history of multiple respiratory infections and sinusitis. There was no immunodeficiency or recurrent infection history among his family members. Agammaglobulinemia was characterized as follows: Immunoglobulin (Ig) A, 90.0 mg/dL (90-450 mg/dL); IgG, 20.0 mg/dL (800-1800 mg/dL); and IgM, 18.0 mg/dL (60-280 mg/dL). Notably, the assessment of IgG subtypes revealed the following very low levels: Subtype 1, 0.26 g/L (3.62-12.28 g/L); subtype 2, 0.10 g/L (0.57-2.9 g/L); subtype 3, 0.009 g/L (0.129-0.789 g/L); and subtype 4, 0.003 g/L (0.013-1.446 g/L). Cellular immunological test results were as follows: CD3, 74.6% (50%-84.0%); CD4, 47.3% (27.0%-51.0%); and CD8, 24.9% (15.0%-44.0%). Ade novohemizygous deletion inBTKwas detected: c.902_c.904delAAG/p.E301del. Transcript levels of the mutantBTKwere similar to those of the wild-type gene, though overexpression resulted in markedly reduced levels of mutantBTK(9.49% +/- 1.58%), relative to the wild-typeBTK(75.8% +/- 2.98%, P < 0.01). CONCLUSION This case of X-linked agammaglobulinemia was attributed to ade novohemizygous deletion mutation inBTK(c.902_c.904delAAG/p.E301del).
