The energy released when tetradecyltrimethylammonium bromide (C(14)TAB) is combined with different derivatives of chlorobenzoates in an isothermal titration calorimeter (ITC) allows a direct evaluation of the spontaneity of the threading of the aromatic anions into the micellar palisade. The comparison between the enthalpimetric curves with the ones for viscosity of the solutions, hydrodynamic radii of the aggregates (dynamic light scattering experiments), and the fraction of aromatic anions incorporated (diffusion-ordered spectroscopy experiments) allows the establishment of the variations of enthalpy with formation, growth, and decrease of the wormlike micelles (WLMs). The formation of WLMs with C(14)TAB is very favored (very exothermic) for titrations of chlorobenzoate derivatives which present the chlorine atom in positions 3 or 4 of the aromatic ring. However, the aggregation is highly unfavorable if chlorine is at position 2 of chlorobenzoate. According to the results, the high potential of the ITC to determine critical concentrations and the energies associated with the aggregation of a cationic surfactant and aromatic anions for the formation of WLMs was demonstrated.

Our condition provides a convenient protocol for the synthesis of a diverse collection of 2-mercaptobenzothiazoles and 6-(4-substituted-1H-1,2,3-triazol-1-yl)-2-mercaptobenzothiazoles with a very simple purification process. This report provides an alternative protocol for fast access to the wide range of compounds for sequence synthesis and biological studies.

Herein, we report novel catalytic methodologies for protodecarboxylations and decarboxylative deuterations of carboxylic acids utilizing a silver-containing hybrid material as a heterogeneous noble metal catalyst. After an initial batch method development, a chemically intensified continuous flow process was established in a simple packed-bed system which enabled gram-scale protodecarboxlyations without detectable structural degradation of the catalyst. The scope and applicability of the batch and flow processes were demonstrated through decarboxylations of a diverse set of aromatic carboxylic acids. Catalytic decarboxylative deuterations were achieved on the basis of the reaction conditions developed for the protodecarboxylations using D2O as a readily available deuterium source.

Triple-negative breast cancer (TNBC), a subset of breast cancers, have poorer survival than other breast cancer types. Recent studies have demonstrated that the abnormal Hedgehog (Hh) pathway is activated in TNBC and that these treatment-resistant cancers are sensitive to inhibition of the Hh pathway. Smoothened (Smo) protein is a vital constituent in Hh signaling and an attractive drug target. Vismodegib (VIS) is one of the most widely studied Smo inhibitors. But the clinical application of Smo inhibitors is limited to adult patients with BCC and AML, with many side effects. Therefore, it’s necessary to develop novel Smo inhibitor with better profiles. Twenty [1,2,4]triazolo[4,3-a]pyridines were designed, synthesized and screened as Smo inhibitors. Four of these novel compounds showed directly bound to Smo protein with stronger binding affinity than VIS. The new compounds showed broad anti-proliferative activity against cancer cell lines in vitro, especially triple-negative breast cancer cells. Mechanistic studies demonstrated that TPB15 markedly induced cell cycle arrest and apoptosis in MDA-MB-468 cells. TPB15 blocked Smo translocation into the cilia and reduced Smo protein and mRNA expression. Furthermore, the expression of the downstream regulatory factor glioma-associated oncogene 1 (Glil) was significantly inhibited. Finally, TPB15 demonstrated greater anti-tumor activity in our animal models than VIS with lower toxicity. Hence, these results support further optimization of this novel scaffold to develop improved Smo antagonists.

We report herein P(III)-directed C-H bond alkenylation of (dialkyl)- and (diaryl)biarylphosphines using internal alkynes. Chloride-free [Rh(OAc)(COD)](2) acts as a better catalyst than commercially available [RhCl(COD)](2). Conditions were developed to control the mono- and difunctionalization depending on the alkyne stoichiometry. One of these novel bisalkenylated (dialkyl)biarylphosphines was employed for the preparation of a palladium(II) complex, and some of these functionalized ligands outperformed their corresponding unfunctionalized phosphines in Pd-catalyzed amidation with sterically hindered aryl chlorides.

The current report represents a transition-metal-free synthesis of oxazoline and isoxazoline derivatives by a tandem electro-oxidative chalcogenation-cyclization process. Both C-Se and C-S bond-forming protocols were developed without using any external oxidant and the reaction was performed at room temperature, open to the air. Using this methodology, 29 substituted oxazoline and 16 substituted isoxazoline derivatives were synthesized with up to 91% isolated yield.

An unusual phenomenon occurred in new display cases at the Rijksmuseum four months after their installation in April 2013. White deposits were visible on glass windows, silicone door gaskets, black structural adhesive seals, and on works of art. The works of art most affected by these deposits were bronze sculptures, wooden and waxed objects, tempera, and oil paintings. In this study the deposits were chemically characterised using (pyrolysis-)gas chromatography-mass spectrometry ((Py-)GC-MS), mu-Raman spectroscopy (mu-Raman), ion chromatography (IC), and scanning electron microscopy combined with energy dispersive X-ray spectroscopy (SEM-EDX). Five chemically different crystalline deposits were found. They were identified as organic salts of the base 2,2,6,6-tetramethyl-4-piperidinol (TMP-ol), a secondary amine, and five different carboxylic acids. It was found that TMP-ol, which is part of the UV-light stabiliser Tinuvin-770, emitted from the structural adhesive Terostat-9220. Terostat-9220 was used in large quantities in the display cases to adhere glass windows to metal parts. The carboxylic acids derived from both construction materials used to build the cases and from conservation materials present on the exhibited works of art. The carboxylic acids involved were 2,4-dichlorobenzoic acid, formic acid, methacrylic acid, palmitic acid, and an unknown carboxylic acid, respectively emitted from peroxide-cured silicone gaskets, panels of medium-density fibreboard (MDF), UV-adhesive, beeswax containing products, and from an unknown acidic conservation product or binding medium. The identification of the crystalline deposits was supported by their syntheses in the laboratory. Since 2013, similar deposits have been observed in a number of museum collections worldwide. A treatment for preventing further growth of the deposits was developed and applied in the Rijksmuseum showcases.

Much of the worlds groundwater supply has been contaminated by aromatic hydrocarbons originating from anthropogenic sources. To study the occurrence and distribution characteristics of aromatic hydrocarbons in groundwater, 24 aromatic hydrocarbon compounds were selected: Five BTEX compounds (benzene, toluene, ethylbenzene, o-xylene, m-xylene, and p-xylene), 10 alkyl-substituted benzene, and 9 halogenated aromatics. These aromatic hydrocarbons were then analyzed from 355 samples collected from across China. Results indicated that aromatic hydrocarbons were detected in 59 out of 355 samples. Of the selected aromatic hydrocarbons, BTEX compounds were detected with high frequency and at low concentrations; comparatively, halogenated aromatics were detected with low frequency and at high concentrations. The aromatic hydrocarbon characteristics found in both karst and pore groundwater samples were then determined using their respective hydrogeological conditions and corresponding human activities. In karst groundwater, BTEX compounds were the most frequently detected aromatic hydrocarbon. The high detection frequencies of aromatic hydrocarbons were caused by their rapid migration, owing to the developed conduit system in the sampled karst area. The low concentrations of aromatic hydrocarbons in karst groundwater samples were caused by low-intensity human activity along with special hydrogeological conditions with higher redox potential and the unique compositions of aromatic hydrocarbons. Alkyl-substituted aromatics and halogenated aromatics were detected at higher concentrations in pore groundwater, owing to high-intensity human activity. Aromatic hydrocarbon pollution was gradually decreased along piedmont-alluvial plain-coast line, owing to a decrease in aquifer vulnerability. These were positively correlated with the size of the aquifer’s particles. Samples with a high accumulative concentration of these aromatic hydrocarbons tended to occur in pore groundwater with a high concentration of either SO42- or Cl-.

Background:Licensed therapies for nonalcoholic fatty liver disease (NAFLD) do not yet exist, but clinical trials are testing treatment options. Inclusion criteria often require liver biopsy showing fibrosis (F2/3) or cirrhosis (F4) and nonalcoholic steatohepatitis (NASH). However, histological criteria pose a serious obstacle for recruitment. Aims:Characterize the relevance of liver biopsies in the selection of patients with NAFLD. Methods:Patients between 2013 and 2018 with the ICD-10 code K76.0 were analyzed. Fibrosis was defined by the NASH clinical research network (CRN) fibrosis staging system, NASH by a NAFLD activity score (NAS) >= 4. Predictive factors were determined by logistic regression. Results:Liver biopsy was performed in 87/638 (13.6%) patients (49% female, age 52.5 +/- 14.0, BMI 30.4 +/- 5.9 kg/m(2)). Fibrosis stage F0/F1/F2/F3/F4 was observed inN = 7/47/7/17/9, an NAS >= 4 inN = 27. Fibrosis stage F2/F3 and F4 along with NAS >= 4 was found in 1.7% and 0.5% of cases. Liver stiffness measurement, LSM (OR 2.3 per doubling of value; CI 1.3-4.4,p = .005) and FIB-4 (OR 2.3 per doubling of value; CI 1.2-4.4,p = .012) were significant predictors for fibrosis >= F2. Predictive factors for NASH were not identified. Conclusion:The biopsy rate in NAFLD patients is low and fibrosis >= F2 along with NAS >= 4 only present in a few cases. Transient elastography and FIB-4 are useful to select patients at risk for fibrosis for liver biopsy.
