Colitis-associated colorectal cancer (CAC) develops from chronic intestinal inflammation. Dihydroartemisinin (DHA) is an antimalarial drug exhibiting anti-inflammatory and anti-tumor effects. DHA inhibited inflammatory responses in the early stage of the AOM/DSS model and subsequent tumor formation. Herein, authors present a straightforward synthetic strategy mediated by triethylamine to prepare the target salicylaldehyde functional group’s bearing bis(sulfonate) derivatives The novel bis(sulfonate) derivatives were designed, synthesized, and characterized for the first time. Enzyme inhibition effects and enzyme interactions of compounds were evaluated on tyrosinase and pancreatic lipase enzymes by using in silico and in vitro methods. According to the enzyme assays, compound I had more effective inhibition against the pancreatic lipase enzyme with a lower IC50 value (53.3 ± 2.7μM) than the other compounds On the other hand, two of the newly synthesized compounds (II and I) had effective inhibitions against the tyrosinase enzyme with having (49.5 ± 2.5μM) IC50 values. In addition, mol. docking studies were performed to determine the interactions and binding energy levels of the bis(sulfonate) derivatives with tyrosinase and pancreatic lipase. Also, addnl. ADME studies of the bis(sulfonate) compounds were evaluated.

New series of 2-amino-1,4,5,6,7,8-hexahydroquinoline-3-carbonitriles I (R = 2,3-dichlorophenyl, 2,4-difluorophenyl) and 2-amino-5,6,7,8-tetrahydronaphthalene-1,3-dicarbonitriles II (R1 = 2,6-difluorophenyl, [1,3]benzodioxol-4-yl, anthracen-9-yl, etc.) were synthesized and evaluated for their antitumor activity. In vitro antitumor screening of the new members against HepG2, HCT-116 and MCF-7 cancer cells showed that the tetrahydronaphthalene-1,3-dicarbonitrile II (R1 = 2-chloro-6-fluorophenyl) (A) has the highest potency against the three tested cancer cells (IC50 = 6.02, 8.45 and 6.28μM, resp.). In addition, compound A displayed low cytotoxicity against WI38 and WISH normal cells (IC50 = 51.78 and 42.36μM, resp.), and it might be utilized as a potent and selective antitumor agent. Compound A was further studied for its effect on tubulin polymerization, different phases of cell cycle, apoptosis and caspases 3/9 levels. Results revealed that analog compound A has good tubulin polymerization inhibitory activity (IC50 = 3.64μM). Addnl., it induced significant accumulation of the tested cancer cells in G2/M phase, and induced cell death primarily via apoptosis. Besides, it showed evident increase in caspase-3 level in HepG2 and HCT-116 cells, and caspase-9 level in MCF-7 cells.
