Src homol. 2 (SH2) domain-containing phosphatase 2 (SHP2) plays a role in receptor tyrosine kinase (RTK), neurofibromin-1 (NF-1), and Kirsten rat sarcoma virus (KRAS) mutant-driven cancers, as well as in RTK-mediated resistance, making the identification of small-mol. therapeutics that interfere with its function of high interest. Our quest to identify potent, orally bioavailable, and safe SHP2 inhibitors led to the discovery of a promising series of pyrazolopyrimidinones that displayed excellent potency but had a suboptimal in vivo pharmacokinetic (PK) profile. Hypothesis-driven scaffold optimization led us to a series of pyrazolopyrazines with excellent PK properties across species but a narrow human Ether-á-go-go-Related Gene (hERG) window. Subsequent optimization of properties led to the discovery of the pyrimidinone series, in which multiple members possessed excellent potency, optimal in vivo PK across species, and no off-target activities including no hERG liability up to 100μM. Importantly, compound 30 (IACS-15414) potently suppressed the mitogen-activated protein kinase (MAPK) pathway signaling and tumor growth in RTK-activated and KRASmut xenograft models in vivo.

Dai, Linlin;Wang, Luyao;Tan, Cheng;Cai, Jun;Shen, Hongsheng;Zhang, Ting;Zhi, Shuang;Yang, Zibo;Hu, Yunhui;Zhao, Xiumei;Li, Dongdong research published 《 Sophoridine Derivatives Induce Apoptosis and Autophagy to Suppress the Growth of Triple-Negative Breast Cancer through Inhibition of mTOR Signaling》, the research content is summarized as follows. In order to improve the antitumor potency and therapeutic margins of natural product sophoridine, its novel nitrogen mustard carbamate derivatives were designed and synthesized. In screening their in vitro activity, we found all the tested compounds were more potent against the highly aggressive triple-neg. breast cancer cell line MDA-MB-231. Cellular functional assays showed that representative compounds could induce G1-phase arrest and trigger apoptosis, evidenced by the alteration of Bax, Bcl-2, caspase-3 and PARP levels. Furthermore, these compounds significantly enhanced the autophagic flux with increased expression of LC3-II and Beclin-1, as well as decreased level of p62, which may attribute to simultaneously inhibition of the phosphorylation of p70S6K, 4E-BP1 and AKT, the key substrates of the mTOR signaling pathway. In vivo, two compounds revealed potent antitumor activity in mice bearing MDA-MB-231. Altogether, our work describes novel leads to yield more potent chemotherapeutics against triple-neg. breast cancers, possibly mesenchymal stem-like subtype.

Dai, Yating;Liang, Shuangshuang;Zeng, Guangkuo;Huang, Hongchun;Zhao, Xiaowei;Cao, Shanshan;Jiang, Zhiyong research published 《 Asymmetric [3 + 2] photocycloadditions of cyclopropylamines with electron-rich and electron-neutral olefins》, the research content is summarized as follows. Radical addition to olefins is a common and useful chem. transformation. In the context of offering enantioenriched three-dimensional mols. via such a highly reactive process, chiral hydrogen-bonding (H-bonding) catalysis has been widely used to provide enantiocontrol. The current strategies for operating H-bonding induction are confined to following that are prevalent in ionic-type manifolds. Here, authors report a novel protocol towards electron-rich olefins based on converting these species from acting as H-bonding donors to acceptors. It facilitates the first development of asym. [3 + 2] photocycloadditions with cyclopropylamines. The method is also effective for electron-neutral olefins, in which the successful construction of all-carbon quaternary stereocentres from 1,1-diaryl ethylenes that feature two structurally similar aryl substituents demonstrates the versatility of this new chiral H-bonding catalytic strategy. Furthermore, the importance of the obtained six kinds of products in pharmaceuticals and asym. catalysis underscores the practicability of this work.

A highly atom profitable nucleophilic addition reaction of aromatic aldehydes and heterocyclic amines for the generation of amides I [R = Ph, 2-furanyl, benzylidene, etc.] having an excellent yield and high reproducibility were reported. The main force responsible for the forward reaction was the presence of the nitroso group at the C-5 position on uracil scaffold, which resulted in the formation of formidable range products. The conclusive mechanism was based on the dehydrogenation reaction of the carbinolamine product formed during the nucleophilic addition reaction of the aromatic aldehyde with the free heterocyclic amino group to yield the corresponding amides. The synthesized compounds I were predicated for biol. properties and proved it to be an ideal G protein-coupled receptor kinase2 (GRK2) inhibitor. Out of all, the compound I [R = 2-methoxyphenyl] showed the most preeminent properties when compared to Paroxetine, studied using mol. docking. Daubit, Isabelle Marie;Wortmann, Svenja;Siegmund, Daniel;Hahn, Stephan;Nuernberger, Patrick;Metzler-Nolte, Nils research published 《 Unveiling Luminescent Ir(I) and Rh(I) N-Heterocyclic Carbene Complexes: Structure, Photophysical Specifics, and Cellular Localization in the Endoplasmic Reticulum》, the research content is summarized as follows. Complexes of Rh(I) and Ir(I) of the [M(COD)(NHC)X] type (M = Rh or Ir, COD = 1,5-cyclooctadiene, NHC = N-heterocyclic carbene, and X = halide) have recently shown promising cytotoxic activities against several cancer cell lines. Initial mechanism of action studies provided some knowledge about their interaction with DNA and proteins. However, information about their cellular localization remains scarce owing to luminescence quenching within this complex type. Herein, the synthesis of two rare examples of luminescent Rh(I) and Ir(I) [M(COD)(NHC)I] complexes with 1,8-naphthalimide-based emitting ligands is reported. All new complexes are comprehensively characterized, including with single-crystal x-ray structures. Steric crowding in one derivative leads to two distinct rotamers in solution, which apparently can be distinguished both by pronounced NMR shifts and by their resp. spectral and temporal emission signatures. When the photophys. properties of these new complexes are exploited for cellular imaging in HT-29 and PT-45 cancer cell lines, the complexes accumulate predominantly in the endoplasmic reticulum, which is an entirely new finding and provides the 1st insight into the cellular localization of such Ir(I)(NHC) complexes.

A series of coumarin derivatives and isosteres were synthesized from the reaction of triflic intermediates with phenylboronic acids, terminal alkynes, and organozinc compounds through palladium-catalyzed cross-coupling reactions. The in vitro cytotoxic effect of the compounds was evaluated against two non-small cell lung carcinoma (NSCLC) cell lines (A-549 and H2170) and a normal cell line (NIH-3T3) using cisplatin as a reference drug. Addnl., the effects of the most promising coumarin derivative (9f) in reversing the epithelial-to-mesenchymal transition (EMT) in IL-1β-stimulated A549 cells and in inhibiting the EMT-associated migratory ability in A549 cells were also evaluated. 9f had the greatest cytotoxic effect (CC50 = 7.1 ± 0.8 and 3.3 ± 0.5 μM, resp. against A549 and H2170 cells) and CC50 value of 25.8 μM for NIH-3T3 cells. 9f inhibited the IL-1β-induced EMT in epithelial cells by inhibiting the F-actin reorganization, attenuating changes in the actin cytoskeleton reorganization, and downregulating vimentin in A549 cells stimulated by IL-1β. Treatment of A549 cells with 9f at 7 μM for 24 h significantly reduced the migration of IL-1β-stimulated cells, which is a phenomenon confirmed by qual. assessment of the wound closure. Taken together, our findings suggest that coumarin derivatives, especially compound 9f, may become a promising candidate for lung cancer therapy, especially in lung cancer promoted by NSCLC cell lines.

Demonti, Luca;Saffon-Merceron, Nathalie;Mezailles, Nicolas;Nebra, Noel research published 《 Cross-Coupling through Ag(I)/Ag(III) Redox Manifold》, the research content is summarized as follows. Trifluoromethyl argentates(III) undergo reductive elimination with arylboronic acids, yielding trifluoromethylarenes. In ample variety of transformations, the presence of silver as an additive or co-catalyst is believed to be innocuous for the efficiency of the operating metal catalyst. Even though Ag additives are required often as coupling partners, oxidants or halide scavengers, its role as a catalytically competent species is widely neglected in cross-coupling reactions. Most likely, this is due to the erroneously assumed incapacity of Ag to undergo 2e redox steps. Definite proof is herein provided for the required elementary steps to accomplish the oxidative trifluoromethylation of arenes through AgI/AgIII redox catalysis (i. e. CEL coupling), namely: (i) easy AgI/AgIII 2e oxidation mediated by air; (ii) bpy/phen ligation to AgIII; (iii) boron-to-AgIII aryl transfer; and (iv) ulterior reductive elimination of benzotrifluorides from an [aryl-AgIII-CF3] fragment. More precisely, an ultimate entry and full characterization of organosilver(III) compounds [K]+[AgIII(CF3)4] (K-1), [(bpy)AgIII(CF3)3] (2) and [(phen)AgIII(CF3)3] (3), is described. The utility of 3 in cross-coupling has been showcased unambiguously, and a large variety of arylboron compounds was trifluoromethylated via [AgIII(aryl)(CF3)3] intermediates. This work breaks with old stereotypes and misconceptions regarding the inability of Ag to undergo cross-coupling by itself.
