Do, Thomas H.;Brown, Seth N. research published 《 Mono- and Bis(iminoxolene)iridium Complexes: Synthesis and Covalency in π Bonding》, the research content is summarized as follows. N-(2,6-Diisopropylphenyl)-4,6-di-tert-butyl-o-iminobenzoquinone (Diso) reacts with the (cyclooctadiene)iridium chloride dimer to form a monoiminoxolene complex, (Diso)Ir(cod)Cl. Reaction of 2 equivalent of the iminoquinone with chlorobis(cyclooctene)iridium dimer affords the bis-iminoxolene (Diso)2IrCl. This five-coordinate complex adopts a distorted square pyramidal structure with an apical chloride ligand and undergoes halide exchange to form an air-stable iodide complex. (Diso)2IrCl can be reduced by one electron to form neutral, square planar (Diso)2Ir, while oxidation with PhICl2 gives octahedral trans-(Diso)2IrCl2. The cis isomer can be prepared by air oxidation of (Diso)2IrCl; cis/trans isomerization is not observed even on prolonged heating. Structural and spectroscopic features of the complexes are consistent with the presence of strong, covalent π bonding between the metal and the iminoxolene ligands, with structural data suggesting between 45 and 60% iridium character in the π bonding orbitals, depending on the ancillary ligands. The spectroscopic similarity of (Diso)2Ir and (Diso)2IrCl to their cobalt congeners suggests that the first-row metal complexes likewise have appreciably covalent metal-iminoxolene π bonds.

The ligand-regulated regiodivergent hydroarylation of the distal double bond of allenamides with aryl boronic acid was achieved in the presence of Pd(II) catalysts, delivering a variety of functionalized enamide with excellent E selectivity and Markovnikov/anti-Markovnikov selectivity. Two possible coordination intermediates probably are responsible for the regiodivergent hydroarylation: (1) The coordination Intermediate I, which probably is formed through the coordination of MeCN, distal double bond, Ph to Pd, led to the aryl group away from the Intermediate I, inducing excellent E selectivity and anti-Markovnikov selectivity. (2) A switch of regioselectivity to 1,2-Markovnikov hydroarylation was obtained using bidentate phosphine ligand (dppf or Xantphos). The formed coordination Intermediate II led to the N-tether away from the Intermediate II and at the trans position of aryl, resulting in excellent E selectivity and Markovnikov selectivity. Meanwhile, tentative study on the mechanism proved that the hydron source of this hydroarylation is more likely to be boronic acid. The transmetalation between aryl boronic acid and Pd catalyst was the initial step of this transformation.

In order to discover highly potent α-glucosidase inhibitors, 20 xanthone-benzenesulfonamide derivatives I (R = H, Me, Br, etc.) and II were designed and synthesized. As expected, most of these derivatives exhibited good to excellent inhibitory activities, the IC50 values of these xanthone sulfonamides varied from 0.4 ± 0.05 to 52.2 ± 2.1μM. Especially, compound I (R = OMe) showed the most potent activity (IC50 = 0.4μM), with 111-fold more potent than the pos. control 1-deoxynojirimycin (DNJ), which is a leap compared to authors previous xanthone compound It was noteworthy that these derivatives can be competitive, noncompetitive or uncompetitive inhibitors, while these inhibitory types differed by the alteration of the substituents on the benzene ring of benzenesulfonamide. To give insight into this interesting effect, further docking study was performed, indicating the flexible conformations introduced by the sulfonamide structure affect the binding site and binding mode of these compounds with the enzyme. Furthermore the uncompetitive inhibitors showed significantly stronger binding interactions with the enzyme-substrate complex than the enzyme. Addnl., the competitive inhibitor I (R = NH2) (1μM) exhibited a similar glucose-level decreasing and glucose up-take increasing effect compared to DNJ (5μM), the pos. control in HepG2 cells at five-fold lower concentration, revealing that this compound has promising hypoglycemic effect at the cellular level.

Duan, Xuelun;Zheng, Nan;Li, Ming;Sun, Xinhao;Lin, Zhuye;Qiu, Pan;Song, Wangze research published 《 Remote ether groups-directed regioselective and chemoselective cycloaddition of azides and alkynes》, the research content is summarized as follows. Remote ether groups could be used as directing groups to prepare fully substituted 5-ether-1,2,3-triazoles with exclusive 1,5-regioselectivities and excellent chemoselectivities. Ether group could coordinate with iridium catalyst by lone-pair electron at a distance (up to four σ bonds) away from alkyne to control the regioselectivity by weak coordination effect. The cycloaddition reaction chemoselectively occurred at the propargyl ether moiety of diyne to give unique fully substituted 4-alkynyl-triazole.
