A series of novel 3-[5-substituted phenyl-1,3,4-thiadiazol-2-yl]-2-styryl quinazoline-4(3H)-ones were synthesized and evaluated for anticonvulsant, sedative-hypnotic and CNS depressant activities. After i.p. injection to mice at doses of 30, 100, and 300 mg/kg body weight 2-styrylquinazolin-4(3H)-one derivatives were examined in the maximal electroshock induced seizures (MES) and s.c. pentylenetetrazole (scPTZ) induced seizure models in mice. The neurotoxicity was assessed using the rotorod method. Out of eighteen compounds only five, e.g., I (R = H or Cl), showed anticonvulsant activity in one or more test models. All except two compounds exhibited significant sedative-hypnotic activity via actophotometer screen. CNS depressant activity screened with the help of the forced swim pool method resulted into some potent compounds From the exptl. observation it can be concluded that synthesized compounds exhibited relatively better sedative-hypnotic and CNS depressant activities.

The facile modification of the ligands in organometallic Ru(II)-arene complexes offers more opportunities to optimize their pharmacol. profiles. Herein, three Ru(II)-arene complexes containing a glutathione S-transferase (GST) inhibitor (NBDHEX) in chelate ligand have been designed and synthesized in this study. In vitro results indicated that the ligation with NBDHEX significantly increased the activities and selectivities of the organometallic Ru(II)-arene complexes against tumor cells, especially complex 3, which was the most active compound among the tested compounds DFT calculations and hydrolysis results demonstrated that complex 3 with more alkyl groups in the arene ligand has increased electron d. at the Ru(II) center as compared with complexes 1 and 2, thus resulting in the improved hydrolysis rate, which may be responsible for its higher anticancer activity. Further studies showed that complexes 1-3 can cause the loss of the mitochondrial membrane potential and upregulate the expression of Bcl-2 and Bax in A549 cells, suggesting that complexes 1-3 induced cell death may be mediated via the mitochondrial apoptotic pathway. Thus, these findings suggested that simultaneous modification of the chelate ligands and arene rings in the organometallic Ru(II) arene complexes is an effective way to improve their pharmacol. properties.

A series of thiophene-2-carboxylic acids and thiophene-3-carboxylic acids were identified as a new class of DAO inhibitors. Structure-activity relationship (SAR) studies revealed that small substituents are well-tolerated on the thiophene ring of both the 2-carboxylic acid and 3-carboxylic acid scaffolds. Crystal structures of human DAO in complex with potent thiophene carboxylic acids revealed that Tyr224 was tightly stacked with the thiophene ring of the inhibitors, resulting in the disappearance of the secondary pocket observed with other DAO inhibitors. Mol. dynamics simulations of the complex revealed that Tyr224 preferred the stacked conformation irresp. of whether Tyr224 was stacked or not in the initial state of the simulations. MM/GBSA indicated a substantial hydrophobic interaction between Tyr244 and the thiophene-based inhibitor. In addition, the active site was tightly closed with an extensive network of hydrogen bonds including those from Tyr224 in the stacked conformation. The introduction of a large branched side chain to the thiophene ring markedly decreased potency. These results are in marked contrast to other DAO inhibitors that can gain potency with a branched side chain extending to the secondary pocket due to Tyr224 repositioning. These insights should be of particular importance in future efforts to optimize DAO inhibitors with novel scaffolds.

The discovery of an aromatic rearrangement reaction of O-(arenesulfonyl)hydroxylamines which lead directly to ortho-aminoarylsulfonic acids and ortho-aminobenzenemethanesulfonic acids through formation of a new C-N bond with excellent levels of regiocontrol for the ortho position(s) over all others was reported. The rearrangement was proceeding through an intermol. mechanism and proposed that the regiocontrol observed was the result of attractive non-covalent interactions occurring during the C-N bond-forming step. Highly dispersed metallic Ag nanoparticles (AgNPs) are promising heterogeneous catalysts for carboxylative coupling of terminal alkynes with CO2 under mild conditions. Yet their size-controlled synthesis is very challenging due to the high surface energy. Here the authors prepared amino-functionalized ordered mesoporous polymers as hosts for anchoring AgNPs. Control experiments and computations showed that electron-rich amines confined in mesochannels with varying electron d. and steric hindrance, creating localized active zones (LAZ) to control the growth of AgNPs. The particle size of AgNPs grows along with the increased volume of LAZ around nitrogen species. The authors also revealed that the catalytic activity of Ag-based catalysts is size-dependent and increases with decreasing particle size. Building on these findings, the authors report a facile 1-pot synthesis strategy for preparing amine-incorporated ordered mesoporous polymer (NOMP) with a high sp. surface area, small LAZ volume, and uniform amine sites with controllable loading. These features gave ultrasmall and monodispersed Ag nanoparticles. Remarkably, Ag@NOMP gave a quant. target yield under the conditions of 1 atm CO2 pressure and 50°, showing superior catalytic activity in the CO2 carboxylation compared to other mesoporous analogs.

Development of small-sized nanoformulations for effective tumor penetration, particularly for those tumors with dense stroma is a major challenge in cancer nanomedicine. It is even more challenging to achieve effective co-loading of both hydrophobic and hydrophilic anticancer agents through a small-sized nanocarrier. In this work, we designed a novel redox-responsive gemcitabine (GEM)-conjugated polymer POEG-co-PVDGEM (PGEM) as a small-sized nanocarrier to co-deliver hydrophilic GEM and hydrophobic paclitaxel (PTX). The in vitro physicochem. and biol. properties of PTX/PGEM NPs were characterized. The efficiency of the PGEM carrier in selective codelivery of GEM and PTX in two murine tumor models as well as a patient derived xenograft model (PDX) was also evaluated. In addition, we investigated the changes in tumor immune microenvironment after treatment with PTX/PGEM nanoparticles. We discovered that GEM conjugation could significantly decrease the nanoparticle size from 160 nm to 13 nm. Moreover, different from most reported GEM-conjugated polymers, PGEM polymer could serve as a prodrug carrier to load a wide variety of hydrophobic agents with high drug loading capacity and excellent stability. More importantly, our strategy could be extended to various nucleotides-based drugs such as azacytidine, decitabine and cytarabine, suggesting a new platform for co-delivery of various first line hydrophilic and hydrophobic anticancer agents. Imaging showed that our small-sized carrier was much more effective in tumor accumulation and penetration compared to the relatively large-sized drug carrier. The PGEM prodrug-based carrier not only well retained the pharmacol. activity of GEM, but also boosted T-cell immune response. Furthermore, delivery of PTX via PGEM led to significantly improved antitumor activity in several murine cancer models and a PDX model of colon cancer. This work not only provided a small-sized carrier platform that was able to load multiple hydrophilic and hydrophobic drugs with high loading capacity, but also provided an effective regimen for enhanced tumor penetration and improved anti-tumor immunity.

In this study, the authors identified I as a novel opioid receptor agonist by high-throughput screening. Structural modifications made to I to improve potency and blood-brain-barrier (BBB) penetration resulted in compounds II and III. Compound II was a potent MOR/KOR (κ-opioid receptor) agonist, and compound III was a potent MOR and medium KOR agonist. Both II and III demonstrated a significant antinociceptive effect in a tail-flick test performed in wild type (WT) B6 mice. The ED50 value of III was 1.059 mg/kg, and the brain concentrations of II and III were 7424 and 11696 ng/g, resp. Accordingly, compounds II and III are proposed for lead optimization and in vivo disease-related pain studies.
