Yadav, Suman;Pal, Saikat;Pal, Nilay Kumar;Din Reshi, Noor U.;Pal, Sourav;Bera, Jitendra K. research published 《 Switchable activity of a Ru catalyst bearing an annulated mesoionic carbene ligand for oxidation of primary amines》, the research content is summarized as follows. The catalytic activity of a Ru complex I, bearing a fused π-conjugated imidazo[1,2-a][1,8]naphthyridine-based mesoionic carbene (MIC) ligand, is examined for the oxidation of primary amines RCH2NH2 (R = n-Pr, cyclohexyl, Ph, pyridin-2-yl, etc.). Complex I affords nitriles RCN or imines RCH:NCH2R depending on the nature of the terminal oxidants and solvents used in the reactions. Primary amines are converted to nitriles using NaIO4 in EtOAc/H2O mixture, whereas imines are obtained under O2 balloon pressure in toluene. A variety of nitriles and imines is accessed with high yields and selectivity. A set of control experiments, reaction profiles, and kinetic studies is undertaken to disclose the mechanistic details for nitrile and imine formation. The catalytic reactions illustrate a subtle dependency on the choice of oxidants and solvents in the oxidation of primary amines.

PIP4K2A is an insufficiently studied type II lipid kinase that catalyzes the conversion of phosphatidylinositol-5-phosphate (PI5P) into phosphatidylinositol 4,5-bisphosphate (PI4,5P2). The involvement of PIP4K2A/B in cancer has been suggested, particularly in the context of p53 mutant/null tumors. PIP4K2A/B depletion has been shown to induce tumor growth inhibition, possibly due to hyperactivation of AKT and reactive oxygen species-mediated apoptosis. Herein, we report the identification of the novel potent and highly selective inhibitors BAY-091 and BAY-297 of the kinase PIP4K2A by high-throughput screening and subsequent structure-based optimization. Cellular target engagement of BAY-091 and BAY-297 was demonstrated using cellular thermal shift assay technol. However, inhibition of PIP4K2A with BAY-091 or BAY-297 did not translate into the hypothesized mode of action and antiproliferative activity in p53-deficient tumor cells. Therefore, BAY-091 and BAY-297 serve as valuable chem. probes to study PIP4K2A signaling and its involvement in pathophysiol. conditions such as cancer.

Wu, Dunqi;Fan, Wenzheng;Wu, Lianqian;Chen, Pinhong;Liu, Guosheng research published 《 Copper-Catalyzed Enantioselective Radical Chlorination of Alkenes》, the research content is summarized as follows. In this report, authors reviewed a copper-catalyzed asym. atom transfer radical addition (ATRA) reaction, which remains a great challenge since the discovery of racemic ATRA in the 1940s. This enantioselective radical chlorination of acrylamides was developed, where three reagents, including Togni-I and TMSCl, PhICF3Cl, and CX3SO2Cl, were employed as the radical sources and chlorine source, affording a series of chlorinated carbon-centered quaternary compounds in good yields with excellent enantioselectivity. Notably, the well-designed bulky chiral ligand plays a key role in the successful enantioselective radical chlorination process.

Open-shell non-alternant polycyclic hydrocarbons (PHs) are attracting increasing attention due to their promising applications in organic spintronics and quantum computing. Herein authors report the synthesis of three cyclohepta[def]fluorene-based diradicaloids, by fusion of benzo rings on its periphery for the thermodn. stabilization, as evidenced by multiple characterization techniques. Remarkably, all of them display a very narrow optical energy gap (Egopt= 0.52-0.69 eV) and persistent stability under ambient conditions (t1/2= 11.7-33.3 h). More importantly, this new type of diradicaloids possess a low-lying triplet state with an extremely small singlet-triplet energy gap, as low as 0.002 kcal mol-1, with a clear dependence on the mol. size. This family of compounds thus offers a new route to create non-alternant open-shell PHs with high-spin ground states, and opens up novel possibilities and insights into understanding the structure-property relationships.

Wu, Lianqian;Wang, Fei;Chen, Pinhong;Liu, Guosheng research published 《 Enantioselective Construction of Quaternary All-Carbon Centers via Copper-Catalyzed Arylation of Tertiary Carbon-Centered Radicals》, the research content is summarized as follows. An enantioselective copper-catalyzed arylation of tertiary carbon-centered radicals, leading to quaternary all-carbon stereocenters, has been developed herein. The tertiary carbon-centered radicals, including both benzylic and nonbenzylic radicals, were produced by the addition of trifluoromethyl radical to α-substituted acrylamides, and subsequently captured by chiral aryl copper(II) species to give C-Ar bonds with excellent enantioselectivity. Importantly, an acylamidyl (CONHAr) group adjacent to the tertiary carbon radical is essential for the asym. radical coupling. The reaction itself features broad substrate scope, excellent functional group compatibility and mild conditions.

Phenanthroimidazole derivatives containing phenanthroline and imidazole heterocyclic aromatic rings were effective agents to inhibit tumor cell growth. Halogen element-modified imidazo[4,5f][1,10]phenanthroline derivatives 1-6 (1, 4-fluorophenyl; 2, 4-chlorophenyl; 3, 4-bromobenyl; 4, 2,3-dichlorophenyl; 5, 3,4-dichlorophenyl; and 6, 2,4-dichlorophenyl) were synthesized, and their antitumor activities were investigated. All of the compounds, especially 4, exhibited an excellent inhibitory effect against nasopharyngeal carcinoma CNE-1 cells. This effect was better than that of doxorubicin. Compound 4 also markedly blocked the proliferation of the CNE-1 cells in a zebrafish xenograft model. The antitumor mechanisms might be attributed to apoptosis induction, which triggered ROS-mediated DNA damage and generated mitochondrial dysfunction by stabilizing c-myc G-quadruplex DNA structure. Results indicated that phenanthroimidazole derivatives could act as promising anticancer agents.

Wu, Qiuyue;Dong, Ziyang;Xu, Jiaxi;Yang, Zhanhui research published 《 Sulfur-controlled and rhodium-catalyzed formal (3 + 3) transannulation of thioacyl carbenes with alk-2-enals and mechanistic insights》, the research content is summarized as follows. A rhodium-catalyzed denitrogenative formal (3+3) transannulation of 1,2,3-thiadiazoles I (Ar = Ph, 2H-1,3-benzodioxol-5-yl, furan-2-yl, etc.) with alk-2-enals R3C(R2)=C(R1)CHO [R1 = H, Me; R2 = H, Me, 4-methoxyphenyl, cyclohexyl, etc.; R3 = H, Me,4-methylpent-3-en-1-yl; R2R3 = -(CH2)4-, -(CH2)6-, -(CH2)12-, etc.] is achieved, producing 2,3-dihydrothiopyran-4-ones II in moderate to excellent yields. An inverse KIE of 0.49 is obtained, suggesting the reversibility of the oxidative addition of thioacyl Rh(I) carbenes to alk-2-enals. The late-stage structural modifications of steroid compounds III are realized. Moreover, this study shows that thioacyl carbenes have different reactivities to those of α-oxo and α-imino carbenes, and highlight the importance of heteroatoms in deciding the reactivities of heterovinyl carbenes.
