A series of new diarylethene compounds with both photochromism and aggregation-induced emission have been synthesized by connecting tetraphenylethene and triphenylethene to the bisthienylethene. All the four compounds exhibit good photochromism upon irradiation with alternative UV and visible light. And they are all provided with aggregation-induced emission properties in THF solution with different fraction of water, non-emissive in the pure THF solution but yellowish green fluorescence in the aggregate or solid state. Furthermore, the compounds connecting tetraphenylethene to the bisthienylethene were found to exhibit the stronger fluorescence emission in the aggregate or solid state than the ones connecting triphenylethene to the bisthienylethene.

Promising vaccine adjuvants of self-assembling peptide hydrogels for protein ovalbumin (OVA) are introduced in this study. The hydrogels are formed by the enzyme of phosphatase, and the vaccine adjuvant potency of both L– and D-peptide hydrogels is evaluated. The results indicate that, compared with the clin. used alum adjuvant, both L– and D-peptide hydrogels can increase the IgG production of OVA for about 1.3 and 3.8 times, resp. Both gels can enhance antigen uptake and induce dendritic cell maturation, and promote and prolong accumulation of antigen in lymph node, as well as evoke germinal center formation. However, the D-peptide hydrogel with OVA exhibits a slightly more efficient accumulation of OVA in the lymph nodes and seems preventing tumor growth more significantly than its L-counterpart. With the good biocompatibility and degradability of peptide hydrogels, the hydrogels described in this study have big potential for the production of protein vaccines for immunotherapy against different diseases.

A recent report suggests that the reaction of dimethylsulfoxonium methylide with esters does not produce a chain-extended sulfur ylide as previously reported, but rather affords the corresponding carboxylate salt. We have investigated this assertion by using a combination of ab initio MO calculations, spiking studies, and isotopic labeling. The formation of carboxylate is unambiguously demonstrated to arise through hydrolysis involving adventitious water, principally derived from moisture in com. trimethylsulfoxonium chloride. Careful vacuum drying of this reagent diminishes the competing hydrolytic pathway resulting in higher yields for the chain-extension reaction than previously reported.

A set of reactive mordant azo dyes was prepared by diazotization of aromatic amines and their subsequent coupling reaction with secondary components. The reactive dyes thus formed contained 2-sulfatoethylsulfonyl (-SO2CH2CH2OSO3Na), N-methyl-N-carboxymethyl-2-aminoethylsulfonyl (-SO2CH2CH2NMeCH2CO2H) and 2,3-dibromopropionamidyl (-NHCOCHBrCH2Br) reactive groups capable of liberating reactive vinyl groups in the application dyebath. Neg.-ion electrospray ionization mass spectrometry, UV/visible spectroscopy, and elemental anal. confirmed the structure of the prepared dyes. Dyed wool fabric samples (combination of acid and reactive dyeing conditions) were treated (mordanted) with iron salts (FeCl2, FeCl3) and a comparison chromium salt (CrF3). The iron mordant dyeing showed acceptable wash fastness and light fastness comparable with chromium complexed dyes.

Protein tyrosine phosphatases (PTPs) constitute a large family of signaling enzymes that control the cellular levels of protein tyrosine phosphorylation. A detailed understanding of PTP functions in normal physiol. and in pathogenic conditions has been hampered by the absence of PTP-specific, cell-permeable small-mol. agents. We present a stepwise focused library approach that transforms a weak and general non-hydrolyzable pTyr mimetic (F2Pmp, phosphonodifluoromethyl phenylalanine) into a highly potent and selective inhibitor of PTP-MEG2, an antagonist of hepatic insulin signaling. The crystal structures of the PTP-MEG2-inhibitor complexes provide direct evidence that potent and selective PTP inhibitors can be obtained by introducing mol. diversity into the F2Pmp scaffold to engage both the active site and unique nearby peripheral binding pockets. Importantly, the PTP-MEG2 inhibitor possesses highly efficacious cellular activity and is capable of augmenting insulin signaling and improving insulin sensitivity and glucose homeostasis in diet-induced obese mice. The results indicate that F2Pmp can be converted into highly potent and selective PTP inhibitory agents with excellent in vivo efficacy. Given the general nature of the approach, this strategy should be applicable to other members of the PTP superfamily.

Through initiation of rhodium(III)-catalyzed C-H activation, a diverse range of N-heterocyclic directing groups, including pyridine, pyrazole, imidazo[1,2-a] pyridine, benzoxazole, benzothiazole, and purine, was feasible for the cascade transformation, exhibiting high efficiency (up to 92% yield), broad substrate scope, and excellent functional group compatibility. Moreover, the modification of natural products and pharmaceutical compounds was also demonstrated to showcase its synthetic utility. Based on d. functional theory (DFT) calculations, a key three-membered ring intermediate through the insertion relay, rather than the direct E/Z isomerization of alkenyl rhodium species, controlled the stereochem. outcome for this trans-exo-selective cyclization. The subsequent ring-opening protonation of the more favored rotamer led to exclusive trans-exo-selectivity.
