We have constructed transgenic Arabidopsis lines that contain a gene for green fluorescent protein (GFP) under the control of auxin-responsive domains A and B of the promoter from the pea PS-IAA4/5 gene. The chimeric transgene was named BA-mgfp5-ER. GFP was detected after the application of indole-3-acetic acid at concentrations as low as 100 nM in epidermal cells in the root elongation zone. The induction of the reporter gene was highly specific to auxin and was correlated with the auxin-induced change in epidermal cell shape. No GFP accumulation was observed in the lateral root meristem that was formed as a result of exogenous auxin application. These results suggest that auxin signals were transmitted through several distinct pathways depending on the cell type. The intensity of the GFP signal was strong enough to be observed through the plastic lid of the culture dish using a dissecting microscope, thereby enabling GFP expression to be monitored in an aseptic environment. Thus, the BA-mgfp5-ER transgenic plant can be a powerful tool for screening mutants that are defective in auxin signaling and the expression of early auxin-response genes.

In this work, polyhomologation (also known as “C1 polymerization”) of a deuterated ylide was performed, yielding linear polyethylene of high deuterium content. Copolymerization with the protonated analog allowed for the preparation of statistical copolymers, in which linear polyethylene with a uniform and tunable amount of deuterium is obtained. The amount of deuterium present, as well as its uniform distribution, is supported by vibrational spectroscopy, size-exclusion chromatog. with IR detection, and 2H NMR spectroscopy. This technique was extended to prepare a precision triblock copolymer in which the chain-ends are selectively deuterated. All polymers reported are of narrow dispersity, and will find use in myriad measurements sensitive to deuterium labeling, most notably small-angle neutron scattering.

Here, the authors describe a process for identifying a 60 nM inhibitor of the interleukin-2 (IL-2)/IL-2 receptor (IL-2Rα) interaction. By use of fragment-based approaches, a compound with millimolar affinity was evolved to a hit series with low micromolar activity, and these compounds were optimized into a lead series with nanomolar affinity. Fragment assembly was useful not only for hit identification, but also for lead optimization. Pyruvate kinase M2 isoform (PKM2) is a crucial protein responsible for aerobic glycolysis of cancer cells. Activation of PKM2 may alter aberrant metabolism in cancer cells. In this study, we discovered a 4-hydroxy-thiazolidine-2-thione compound 2 as a novel PKM2 activator from a random screening of an inhouse compound library. Then a series of novel 4-hydroxy-thiazolidine-2-thione derivatives were designed and synthesized for screening as potent PKM2 activators. Among these, some compounds showed higher PKM2 activation activity than lead compound 2 and also exhibited significant anti-proliferative activities on human cancer cell lines at nanomolar concentration The compound 5w was identified as the most potent antitumor agent, which showed excellent anti-proliferative effects with IC50 values from 0.46 μM to 0.81 μM against H1299, HCT116, Hela and PC3 cell lines. 5w also showed less cytotoxicity in non-tumor cell line HELF compared with cancer cells. In addition, Preliminary pharmacol. studies revealed that 5w arrests the cell cycle at the G2/M phase in HCT116 cell line. The best PKM2 activation by compound 5t was rationalized through docking studies.
