Iron oxide nanoparticle synthesis using trigonella and tomato extracts and their antibacterial activity

Iron oxide nanoparticles (IONPs) were synthesised by combining the extracts of new plants (trigonella and tomato) with ferric chloride salt (FeCl3) via a simple chemical method at 200 degrees C for 2 hours. The biomolecules in the extracts play an important part in transforming FeCl3 to FeO NPs, which demonstrate antibacterial properties. IONPs were characterised via X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), and Fourier-transform infrared spectroscopy (FT-IR). XRD revealed a crystallite size of 17 nm with a body centre cubic (BCC) structure (wustite) phase for FeO NPs made using trigonella extract. FeO NPs synthesised using tomato extract were 41 nm, and the diffraction peaks revealed a BCC structure phase with excellent crystal quality. FE-SEM images demonstrated that the average grain size of Fe2O3 without using any plant extract was approximately 31.36 to 125.5 nm, while for the FeO NPs using tomato extract, it was approximately 48.18 to 77.54 nm, and for trigonella extract, 27.91 to 40.94 nm. In the FT-IR spectra of IONPs with and without plant extract, a strong peak appeared at 699 cm(-1) for FeO NPs using trigonella extract and 655 cm(-1) using tomato extract. Antibacterial activity of IONPs was determined by growth inhibition zones of the gram-negative bacteria Klebsiellapneumonia and gram-positive bacteria Staphylococcus aureus. The growth inhibition zones for FeO NPs using trigonella extract were 42 and 47 mm, respectively. The zones for FeO NPs using tomato extract were smaller (30 mm and 35 mm, respectively). These NPs may be effective in healing infections caused by these bacteria.

Taxifolin improves disorders of glucose metabolism and water-salt metabolism in kidney via PI3K/AKT signaling pathway in metabolic syndrome rats

Aims: Our study was designed to explore the function and mechanism of taxifolin on glucose metabolism and water-salt metabolism in kidney with metabolic syndrome (MS) rats. Main methods: Spontaneous hypertensive rats were induced by fructose to establish MS model. Systolic blood pressure (SBP) and homeostasis model assessment of insulin resistance (HOMA-IR) were measured after 7 weeks of continuous administration with taxifolin. Kidney injury indices and histopathological evaluation were done. The apoptosis rate of primary kidney cells was detected by flow cytometry. Insulin signaling pathway related proteins and renal glucose transport-related proteins were detected by western blotting. We assessed the effects of taxifolin on sodium water retention and renin-angiotensin-aldosterone system (RAAS) in MS rats. We examined not only changes in urine volume, osmotic pressure, urinary sodium and urinary chloride excretion, but also the effects on NA(+)/K+-ATPase and RAAS indicators. We also detected changes in inflammatory factors by immunohistochemical staining and immunofluorescence. In vitro experiment, high glucose and salt stimulated NRK-52E cells. By adding the PI3K inhibitor (wortmannin) to inhibit the PI3K, the effects of inhibiting the PI3K/ AKT signaling pathway on glucose metabolism, water-sodium retention and inflammatory response were discussed. Key findings: Taxifolin effectively reversed SBP, HOMA-IR, the kidney indices and abnormal histopathological changes induced by MS. Comparison among FLOTAC, Kato-Katz and formalin ether concentration techniques for diagnosis of intestinal parasitic infections in school children in an Egyptian rural setting

The study aimed to compare the diagnostic performance of the Kato-Katz, formalin ether concentration method (FECM) and FLOTAC using Sheather’s sugar solution (FS1), saturated sodium chloride (FS2) and zinc sulfate (FS7) for the diagnosis of intestinal parasites among school children, focusing on Schistosoma mansoni. Ninety fecal samples were examined using the above mentioned techniques. The overall infection rate was 87.7%. Concerning protozoa, FLOTAC (FS1 and FS2) and FECM detected nearly equal infection rates (43.3% and 44.4%, respectively) with very good agreement. Kato-Katz diagnosed the highest helminthic infection rate (57.8%) followed by FLOTAC FS7 (44.4%) and FECM showed the lowest helminthic infection rate (27.7%). As for S. mansoni, Kato-Katz showed an infection rate of 38.8% vs FLOTAC (22.2%) and FECM (11.1%). The three techniques detected the same infection rate (11.1%) with egg counts more than 72 eggs/gram of feces. The FLOTAC sensitivity and accuracy for the diagnosis of protozoa were 97% and 99%, respectively. Regarding helminths diagnosis, FLOTAC technique showed higher sensitivity (77%) and accuracy (87%) compared to FECM (48% sensitivity and 70% accuracy). Therefore, FLOTAC can be used synchronously or in replacement to other diagnostic techniques. This can strategically impact future control programmes of intestinal parasitic infections in limited resources settings.

Charge Regulation of Natural Amino Acids in Aqueous Solutions

The aim of this work is to develop a predictive model to describe the electrostatic behavior of 20 natural amino acids under diverse solution conditions. A coarse-grained model is proposed to account for the key ingredients of thermodynamic nonideality arising from the interaction of amino acids with various solvated ions in an aqueous solution including the molecular volume excluded effects, solvent-mediated electrostatic interactions, van der Waals attraction, and hydrophobic and hydration forces. With a small set of parameters characterizing the intermolecular interactions and dissociation equilibrium, the thermodynamic model is able to correlate extensive experimental data for the activity coefficients and solubility of amino acids in pure water and in aqueous sodium chloride solutions. Moreover, it predicts apparent equilibrium constants for the charge regulation of all amino acids in excellent agreement with experimental data. Importantly, the thermodynamic model allows for the extrapolation of the intrinsic equilibrium constants for amino-acids conversion among different charge states (viz., negative, neutral, and positive charges) thereby enabling the prediction of the charge behavior for all natural amino acids under arbitrary solution conditions.

Pathogenic convergence of CNVs in genes functionally associated to a severe neuromotor developmental delay syndrome

Background Complex developmental encephalopathy syndromes might be the consequence of unknown genetic alterations that are likely to contribute to the full neurological phenotype as a consequence of pathogenic gene combinations. Methods To identify the additional genetic contribution to the neurological phenotype, we studied as a test case a boy, with a KCNQ2 exon-7 partial duplication, by single-nucleotide polymorphism (SNP) microarray to detect copy-number variations (CNVs). Results The proband presented a cerebral palsy like syndrome with a severe motor and developmental encephalopathy. The SNP array analysis detected in the proband several de novo CNVs, nine partial gene losses (LRRC55, PCDH9, NALCN, RYR3, ELAVL2, CDH13, ATP1A2, SLC17A5, ANO3), and two partial gene duplications (PCDH19, EFNA5). The biological functions of these genes are associated with ion channels such as calcium, chloride, sodium, and potassium with several membrane proteins implicated in neural cell-cell interactions, synaptic transmission, and axon guidance. Pathogenically, these functions can be associated to cerebral palsy, seizures, dystonia, epileptic crisis, and motor neuron dysfunction, all present in the patient. Conclusions Severe motor and developmental encephalopathy syndromes of unknown origin can be the result of a phenotypic convergence by combination of several genetic alterations in genes whose physiological function contributes to the neurological pathogenic mechanism.

Insights into Primary Ion Exchange between Ion-Selective Membranes and Solution. From Altering Natural Isotope Ratios to Isotope Dilution Inductively Coupled Plasma Mass Spectrometry Studies

Although ion-selective electrodes have been routinely used for decades now, there are still gaps in experimental evidence regarding how these sensors operate. This especially applies to the exchange of primary ions occurring for systems already containing analyte ions from the pretreatment step. Herein, for the first time, we present an insight into this process looking at the effect of altered ratios of naturally occurring analyte isotopes and achieving isotopic equilibrium. Benefiting from the same chemical properties of all isotopes of analyte ions and spatial resolution offered by laser ablation and inductively coupled plasma mass spectrometry, obtaining insights into primary ion diffusion in the preconditioned membrane is possible. For systems that have reached isotopic equilibrium in the membrane through ion exchange and between the membrane phase and the sample, quantification of primary ions in the membrane is possible using an isotope dilution approach for a heterogeneous system (membrane-liquid sample). Experimental results obtained for silver-selective membrane show that the primary ion diffusion coefficient in the preconditioned membrane is close to (6 +/- 1) x 10(-)(9) cm(2)/s, being somewhat lower compared to the previously reported values for other cations. Diffusion of ions in the membrane is the rate limiting step in achieving isotopic exchange equilibrium between the ion-selective membrane phase and sample solution. On the contrary to previous reports, quantification of silver present in the membrane clearly shows that contact of the membrane with silver nitrate solution of concentration 10(-3) M leads to pronounced accumulation of silver ions in the membrane, reaching almost 150% of ion exchanger amount. The magnitude of this effect increases for higher concentration of the electrolyte in the solution.

Ethnopharmacological relevance: Ephedrae Herba (EH, Ephedra sinica Stapf.) and Armeniacae Semen Amarum (ASA, Prunus armeniaca L. var. ansu Maxim.) have been used to treat asthma, cold, fever, and cough in China for thousands of years. Aim of the study: In this study, we aimed to investigate the optimal ratio of EH and ASA compatibility (EAC) to reduce airway injury in asthmatic rats and its possible mechanism. Methods: Rats were sensitized with a mixture of acetylcholine chloride and histamine bisphosphate 1 h before sensitization by intragastric administration of EAC or dexamethasone or saline for 7 days. Subsequently, the ultrastructure of rat airway epithelial tissue changes, apoptosis of the airway epithelial cells, and the expression of mRNA and protein of EGRF and Bcl-2 were detected. Results: Transmission electron microscope: EAC (groups C and E) had the most prominent effect on repairing airway epithelial cells’ ultrastructural changes in asthmatic rats. TUNEL: dexamethasone and EAC (groups B, C, E and F) inhibited the apoptosis of airway epithelial cells in asthmatic rats (P < 0.05). In situ hybridization: EAC (group E) inhibited the overexpression of EGFR and Bcl-2 mRNA (P < 0.05).Western Blotting: EAC (groups A, B, C, E and F) inhibited the upregulation of airway epithelial EGFR and Bcl-2 protein expression (P < 0.01). Conclusions: Our findings indicate that EAC can inhibit abnormal changes in airway epithelial structure and apoptosis of airway epithelial cells, thereby alleviating airway injury. Adsorptivity of cationic cellulose nanocrystals for phosphate and its application in hyperphosphatemia therapy

Nanocellulose has gained much attention because of its excellent properties. Cationic cellulose nanocrystals (cCNC) shows good adsorptivity toward negative ions and molecules. Phosphate binders are most used to treat hyperphosphatemia and it is significant to develop its alternatives with high specific and low cost in the clinic. Herein, we prepared cCNC and characterized it by FTIR, TEM, dynamic light scattering, and viscosity method. We simulated the binding process of cationic cellulose for phosphate and used it as phosphate binder for hyperphosphatemia therapy to study the phosphate binding effect and evaluate the oral toxicity. Cationic cellulose improved the conditions of mice models and efficiently decreased the level of phosphate in the serum. cCNC had a better binding effect than cationic micmcrystalline cellulose both in vitro and in vivo. cCNC could be used as alternatives to phosphate binder for therapy of chronic renal failure and hyperphosphatemia.

Pt nanoparticle decorated carbon nanotubes nanocomposite based sensing platform for the monitoring of cell-secreted dopamine

Development of reliable sensors for real-time quantification of dopamine is highly essential to understand various biological processes and still remains challenge. Here, we proposed a novel and ultrasensitive electrochemical sensing platform for the monitoring of cell-secreted dopamine based on Pt nanoparticles (PtNPs) decorated multi-wall carbon nanotubes (MWCNTs) nanocomposite, which is synthesized by in-situ reduction of platinum on poly (diallyldimethylammonium chloride) (PDDA) functionalized MWCNTs. The unique structure of the hybrid nanocomposite and the interconnectivity between PtNPs and carbon nanotubes endow the nanocomposite with the enhancing conductivity and electrochemical activity. The Pt-MWCNTs hybrid modified screen-printed electrode exhibited excellent electrocatalytic ability for the detection of dopamine with a wide linear range (5 nM to 1 mu M) and a low detection limit (2 nM). The developed sensor enables the real-time recording of dopamine released from PC12 cells induced by the stimulation of K+ saline. Also, this approach can be used to quantitatively evaluate the effects of antipsychotic drug (aripiprazole) on the dopamine release from cells treated with high K+. Results indicated that the sensor could provide an effective tool for the monitoring of extracellular neurotransmitters related to the exogenous agent and drug on the nervous system.

Experimental mapping of a pH gradient from a positively charged micellar interface to bulk solution

The nanoscale pH gradients from a charged membrane interface to the bulk solution, determine fundamental properties of chemical and biological systems. The methylene of the terminal oxyethylene was connected to a quaternary nitrogen containing a long hydrocarbon chain. We calculated the apparent pK(a) (pK(ap)) of the benzoic acid derivatives with surfactant micelles by UV spectroscopy. The difference between the pK(a) in solution and the pK(ap) in micelles (Delta pK) of all probes was very similar with negatively charged sodium dodecyl sulphate (SDS) or zwitterionic N-hexadecyl-N,N-dimethyl-3-ammonio-1-pmpanesulfonate (HPS) micelles, indicating that the dissociation of the substituted benzoates occurred at the micellar interface in both SDS and HPS. In contrast, the Delta pK of the probes decreased as a function of the benzoate’s distance to the micelle interface in hexadecyltrimethylammonium chloride (CTAC) micelles, The variation of Delta pK with distance, from the CTAC micellar interface to bulk solution, was quantitatively correlated to H+ distribution from the interface. The experimental results coincided with independently calculated Poisson Boltzmann H+ distribution.
