C-60-based chemical labeling strategy for the determination of polyamines in biological samples using matrix-assisted laser desorption/ionization mass spectrometry

Bioactive polyamines play important roles in many biological processes such as gene expression, cell growth, protein synthesis, and signal transduction. Accurate determination of polyamines is helpful for studying their biological functions. Herein, a C-60-based chemical labeling strategy was proposed for the determination of polyamines (putrescine, cadaverine, spermidine, and spermine) in biological samples using matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS). An N-hydroxysuccinimide ester functionalized C-60 (NHS-C-60) was used as a labeling reagent and the m/z of the labeled polyamines reached up to more than 900 Da, which avoided matrix interferences in the low m/z region. In addition, as NHS-C-60 derivatives, mono- and bis-substituted polyamines were produced simultaneously, which benefited the qualitative analysis of polyamines. The analytical method was validated using NHS-C(60)labeled polyamines in cells and mice feces samples. Good linearities were obtained with correlation coefficients ranging from 0.9786 to 0.9982. The limits of quantification were in the range of 0.68-1.48 pmol. Good reproducibility and reliability of our proposed method were confirmed by intra- and inter-day precisions ranged from 2.8 to 16.6%, and the recoveries ranged between 81.8 and 119.9%. Finally, the proposed method was applied to determine polyamines in cells and mice feces. Three polyamines were detected in the cells, and the contents of cadaverine and spermidine in the feces of high-fat diet mice were found to be significantly lower than those in the normal diet mice. The results show that the proposed NHS-C-60 labeling coupled with MALDI MS strategy is suitable for the determination of polyamines in biological samples.

Organometallic Complex Strongly Impairs Chikungunya Virus Entry to the Host Cells

Chikungunya fever is a disease caused by the Chikungunya virus (CHIKV) that is transmitted by the bite of the female of Aedes sp. mosquito. The symptoms include fever, muscle aches, skin rash, and severe joint pains. The disease may develop into a chronic condition and joint pain for months or years. Currently, there is no effective antiviral treatment against CHIKV infection. Treatments based on natural compounds have been widely studied, as many drugs were produced by using natural molecules and their derivatives. Alpha-phellandrene (alpha-Phe) is a naturally occurring organic compound that is a ligand for ruthenium, forming the organometallic complex [Ru2Cl4(p-cymene)(2)] (RcP). Organometallic complexes have shown promising as candidate molecules to a new generation of compounds that presented relevant biological properties, however, there is a lack of knowledge concerning the anti-CHIKV activity of these complexes. The present work evaluated the effects of the RcP and its precursors, the hydrate ruthenium(III) chloride salt (RuCl3.xH(2)O) (Ru) and alpha-Phe, on CHIKV infection in vitro. To this, BHK-21 cells were infected with CHIKV-nanoluciferase (CHIKV-nanoluc), a viral construct harboring the nanoluciferase reporter gene, at the presence or absence of the compounds for 16 h. Cytotoxicity and impact on infectivity were analyzed. The results demonstrated that RcP exhibited a strong therapeutic potential judged by the selective index > 40. Antiviral effects of RcP on different stages of the CHIKV replicative cycle were investigated; the results showed that it affected early stages of virus infection reducing virus replication by 77% at non-cytotoxic concentrations. Further assays demonstrated the virucidal activity of the compound that completely blocked virus infectivity. In silico molecular docking calculations suggested different binding interactions between aromatic rings of RcP and the loop of amino acids of the E2 envelope CHIKV glycoprotein mainly through hydrophobic interactions. Additionally, infrared spectroscopy spectral analysis indicated interactions of RcP with CHIKV glycoproteins. These data suggest that RcP may act on CHIKV particles, disrupting virus entry to the host cells. Therefore, RcP may represent a strong candidate for the development of anti-CHIKV drugs.

Production of manno-oligosaccharide from Gleditsia microphylla galactomannan using acetic acid and ferrous chloride

A novel and efficient method for manno-oligosaccharides (MOS) production has been proposed by utilizing Gleditsia microphylla galactomannan as the starting material. This co-operative hydrolysis using ferrous chloride (Fe2+) and acetic acid (HAc) effectively improved the MOS yield and meanwhile decreased the amount of monosaccharide and the 5-hydroxymethyl-furfural (HMF). The highest yields under the optimum conditions were 46.7% by HAc hydrolysis (5 M HAc at 130 degrees C for 120 min); 37.3% by Fe2+ hydrolysis (0.1 M Fe2+ at 150 degrees C for 120 min); and 51.4% by co-operative hydrolysis (2 M HAc, 0.05 M Fe2+ at 160 degrees C for 10 min). From the changes in the value of M/G (mannose/galactose) ratios, it was deduced that Fe2+ predominantly cleaves the main chain, and HAc assists in the breakage of the side chain, thus resulting in the high-efficient co-operative hydrolysis for the production of MOS.

Synthetic and Mechanistic Implications of Chlorine Photoelimination in Nickel/Photoredox C(sp(3))-H Cross-Coupling

In recent years, the development of light-driven reactions has contributed numerous advances in synthetic organic chemistry. A particularly active research area combines photoredox catalysis with nickel catalysis to accomplish otherwise inaccessible cross-coupling reactions. In these reactions, the photoredox catalyst absorbs light to generate an electronically excited charge-transfer state that can engage in electron or energy transfer with a substrate and the nickel catalyst. Our group questioned whether photo-induced activation of the nickel catalyst itself could also contribute new approaches to cross-coupling. Over the past 5 years, we have sought to advance this hypothesis for the development of a suite of mild and site-selective C(sp(3))-H cross-coupling reactions with chloride-containing coupling partners via photoelimination of a Ni-Cl bond. On the basis of a report from the Nocera laboratory, we reasoned that photolysis of a Ni(III) aryl chloride species, generated by single-electron oxidation of a typical Ni(II) intermediate in cross-coupling, might allow for the catalytic generation of chlorine atoms. Combining this with the ability of Ni(II) to accept alkyl radicals, we hypothesized that photocatalytically generated chlorine atoms could mediate hydrogen atom transfer (HAT) with C(sp(3))-H bonds to generate a substrate-derived alkyl radical that is captured by the Ni center in cross-coupling. A photoredox catalyst was envisioned to promote the necessary single-electron oxidation and reduction of the Ni catalyst to facilitate an overall redox-neutral process. Overall, this strategy would offer a visible-light-driven mechanism for chlorine radical formation enabled by the sequential capture of two photons. As an initial demonstration, we developed a Ni/photoredox-catalyzed alpha-oxy C(sp(3))-H arylation of cyclic and acyclic ethers. This method was extended to a mild fonnylation of abundant and complex aryl chlorides through selective 2-functionalization of 1,3-dioxolane. Seeking to develop a suite of reactions that introduce carbon at all different oxidation states, we explored C(sp(3))-H cross-coupling with trimethyl orthoformate, a common laboratory solvent. We found that trimethyl orthofonnate serves as a source of methyl radical for a methylation reaction via beta-scission from a tertiary radical generated upon chlorine-mediated HAT. Since chlorine radical is capable of abstracting unactivated C(sp(3))-H bonds, our efforts have also been directed at cross-coupling with a range of feedstock chemicals, such as alkanes and toluenes, along with late-stage intermediates, using chloroformates as coupling partners. Overall, this platform enables access to valuable synthetic transformations with (hetero)aryl chlorides, which despite being the most ubiquitous and inexpensive aryl halide coupling partners, are rarely reactive in Ni/photoredox catalysis. Little is known about the photophysics and photochemistry of organometallic Ni complexes relevant to cross-coupling. We have conducted mechanistic investigations, including computational, spectroscopic, emission quenching, and stoichiometric oxidation studies, of Ni(II) aryl halide complexes common to Ni/photoredox reactions. These studies indicate that chlorine radical generation from excited Ni(III) is operative in the described C(sp(3))-H functionalization methods. More generally, the studies illustrate that the photochemistry of cross-coupling catalysts cannot be ignored in metallaphotoredox reactions. We anticipate that further mechanistic understanding should facilitate new catalyst design and lead to the development of new synthetic methods.

The synthesis and coordination chemistry of cis-macrocyclic diphosphine (PhPA)(2) (A = C14H10 or anthracene), which features rigid, sterically encumbering groups in its macrocyclic backbone, is reported. Treatment of [EtOP(2)A(2)]AlCl4, obtained in 63% yield from dibenzo-7-phosphanorbornadiene EtOPA and aluminum(III) chloride, with 2 equiv of phenylmagnesium chloride gives (PhPA)(2) in 59% yield. The purple, pseudo-square-planar nickel dichloride complex [(PhPA)(2)]NiCl2 was obtained in 89% yield upon combination of (PhPA)(2) with (DME)NiCl2 in dichloromethane. By virtue of the ligand backbone, [(PhPA)(2)]-NiCl2 exhibits a large percent buried volume of 61.4%, greater than that of other common chelating diphosphines, and the steric bulk is extended into the axial positions of the square-planar complex, as revealed by a single-crystal X-ray diffraction study. Treatment of this nickel dichloride complex with K[B(C6F3)(4)] provided a red dimeric, cationic nickel chloride complex. This dimer was isolated in 43% yield and characterized in a single-crystal X-ray diffraction study.

Lithium-induced overexpression of beta-catenin delays murine palatal shelf elevation by Cdc-42 mediated F-actin remodeling in mesenchymal cells

Background Lithium chloride (LiCl) is widely used for the treatment of manic and other psychotic disorders, but the administration of lithium can result in several congenital defects in the fetus, including cleft palate (Meng, Wang, Torensma, Jw & Bian, 2015) (Szabo, 1970). However, the mechanism of Lithium’s action as a developmental toxicant in palatogenesis is not well known. Methods In this study, hematoxylin-eosin and immunofluorescence staining were employed to evaluate the phenotypes and the expression of related markers in the LiCl-treated mice model. The palatal mesenchymal cells were cultured in vitro, and stimulated with LiCl or SKL2000, and co-treated with CASIN. beta-catenin protein and other cytoskeleton associated markers were evaluated by Western blotting. Results We found that Lithium disrupted palate elevation by increasing the expression of beta-catenin in C57BL/6J mice with the high incidence of cleft palate (62.5%). LiCl disturbed the F-actin responsible for cytoskeletal remodeling in mesenchymal cells, which proved to be essential in generating the elevating force during palatal elevation. Additionally, our Western blotting analysis revealed that the overexpression of beta-catenin resulted in up-regulation of Cdc42, which mediated the downstream F-actin synthesis. Conclusions We concluded the LiCl-induced beta-catenin overexpression delayed murine palatal shelf elevation by disturbing Cdc42 mediated F-actin cytoskeleton synthesis in the palatal mesenchyme.

Development of a Sustainable Alternative for the Ammoniacal Cyanidation of Copper-Gold Ores Through a Biological Approach

Due to the problems arising from the presence of copper in the gold cyanidation process, including high cyanide consumption and occupation of the activated carbon surface, biological ammoniacal cyanidation was implemented as a resolution. Bacillus megaterium was implemented to produce cyanide and ammonia in nutrient broth and vinasse media. Maximum cyanide concentrations of 29.8 and 47.5 mg/L were produced using 3 g/L glycine in the mentioned media, respectively. Afterward, the Box-Behnken experiment design method was implemented to optimize ammonia biogenesis in vinasse culture, where 1974 mg/L ammonia was synthesized at 13.56 g/L calcium chloride, 7.2 g/L urea, and pH=7.9. Three different strategies, namely: (1) bio-cyanidation, (2) bio-ammoniacal cyanidation, and (3) bio-ammonia pretreatment/bio-cyanidation, were employed to recover gold and copper from a 1% solid pulp. Afterward, using the nutrient broth medium, 49% gold was recovered by the first approach; the second approach extracted 9.2% gold and 33.5% copper, and 77% copper removal and 76% gold recovery were achieved through the third technique. Nutrient broth substitution with vinasse increased the gold recovery to 97% in the third approach. Hence, ammoniacal pretreatment, in combination with bio-cyanidation, is suggested to recover gold from copper-gold ores. In conclusion, the originality of the present work lies first in developing a bacterial culture procedure in which nutrient broth was replaced with vinasse as an industrial byproduct to make the process more eco-friendly. And secondly, in using the biogenic ammonia to leach out copper from the copper-gold ore and prepare it for gold biocyanidation.Graphical Abstract

Synthesis of biocompatible and highly fluorescent N-doped silicon quantum dots from wheat straw and ionic liquids for heavy metal detection and cell imaging

Silane-based precursors for the synthesis of water-dispersible silicon quantum dots (SiQDs) present harmful effects on both researchers and the environment, due to their high toxicity. Though waste wheat straw is an abundant source of natural silicon, its application towards the synthesis of biocompatible SiQDs for metal detection has not yet been explored. In this study, N-doped SiQDs demonstrating uniform spherical morphologies, excellent water dispersity and strong fluorescence emission with a quantum yield of 28.9% were facilely synthesized by using wheat straw (WS) as silicon source and allyl-3-methylimidazolium chloride (AMIMCl) as nitrogen source. The wheat straw based SiQDs (WS-SiQDs) showed linear fluorescence quenching ((F-0-F)/F) with Cr(VI) and Fe(III) concentration in the range of 0-6×10(-4) M. Following immobilization on hydrophilic silica hydrogels, WS-SiQDs@silica hydrogels demonstrated enhanced fluorescence emission which can selectively detect Cr(VI) and Fe (III) to the limits of 142 and 175 nM, respectively. Moreover, cell imaging results reflected thatWS-SiQDs can penetrate the membranes of dental pulp stem cells and react with the nucleuses of the stem cells. The stem cells maintained high viability under the conditions of 24 h incubation and SiQD concentration below 50 mg.L-1, thus indicating low cytotoxicity ofWS-SiQDs. The as-prepared SiQDs demonstrated notable structural and fluorescent properties, therefore representing promising biocompatible fluorescent nanomaterials for metal detection and cell imaging. Biomass-Derived Nitrogen-Doped Porous Carbons Activated by Magnesium Chloride as Ultrahigh-Performance Supercapacitors

Porous carbons with high capacitive properties derived from biomass have been increasingly studied for supercapacitor applications. In this work, we prepared N-doped porous carbon through a one-pot MgCl2 activation of lotus root flour with urea. A series of lotus root carbons with different physical/chemical properties were synthesized by adjusting precursors and pyrolysis temperatures. The MgCl2 activation could produce abundant micropores and mesoporous channels in nanocarbons, which are beneficial for ions adsorption and diffusion. The addition of urea improved the porosity, specific surface area, and N content in the carbons, which increased the double-layer capacitance and pseudo-capacitance. MgLC-142-800 with best performance was prepared from a mixture of lotus root flour, MgCl2, and urea with the proportion of 1:4:2 at 800 degrees C, exhibiting high specific capacitance (331.80 and 267.63 F g(-1) at 1 A g(-1 )in the three- and two-electrode system, respectively), excellent rate capability (331.78 and 295.45 F g(-1) at 0.5 and 10 A g(-1), respectively), and good durability (98% capacitance retention a ft er 10000 galvanostatic charge/discharge cycles at 10 A g(-1) in the two-electrode system) in 6 M KOH. Additionally, corn- and black soybean-derived porous carbons were prepared to undergo the same procedures, showing excellent capacitive performance as well. Therefore, biomass-derived nanocarbons with the MgCl2 activation strategy show a promising future for application as a supercapacitor.

Study of Activated Carbon Sheets Used for Air-Cathodes of Portable Quasi-Solid Aluminum-Air Batteries

The air-cathode is investigated for the development and optimization of low-cost, miniaturized, portable, quasi-solid aluminum-air batteries. The battery used aluminum as the anode and gelation sodium chloride as the electrolyte. The air-cathode was activated carbon sheet made of carbon fiber, cellulose, and activated carbon powder.
