Graphitic carbon nitride materials have attracted significant interest in recent years and found applications in diverse light-to-energy conversions such as artificial photosynthesis, CO2 reduction, or degradation of organic pollutants. However, their utilization in synthetic photocatalysis, especially in the direct functionalization of C(sp(3))-H bonds, remains underexplored. Herein, we report mesoporous graphitic carbon nitride (mpg-CN as a heterogeneous organic semiconductor photocatalyst for direct arylation of C(sp(3))-H bonds in combination with nickel catalysis. Our protocol has a broad synthetic scope (>70 examples including late-stage functionalization of drugs and agrochemicals), is operationally simple, and shows high chemo- and regioselectivities. Facile separation and recycling of the mpg-CN catalyst in combination with its low preparation cost, innate photochemical stability, and low toxicity are beneficial features overcoming typical shortcomings of homogeneous photocatalysis. Detailed mechanistic investigations and kinetic studies indicate that an unprecedented energy-transfer process (EnT) from the organic semiconductor to the nickel complex is operating.

V2O5/TiO2-SiO2-MoO3 (TSM) catalysts exhibit enhanced redox capabilities, but the role of each species in the mixed oxide support is not well understood. Herein, the physicochemical characteristics of molybdenum oxide in TSM were investigated using X-ray diffraction, X-ray photoelectron spectroscopy, and X-ray absorption fine structure spectroscopy, and the performance of V/TSM catalysts for the oxidative decomposition of o-chlorotoluene was examined. The V/TSM catalysts exhibited superior performances in this decomposition reaction at low temperatures. The molybdenum species were highly dispersed as Mo6+, replacing Ti4+ in the anatase TiO2 framework and forming a solid solution. The redox capability of the vanadium species in the catalyst was enhanced by electron transfer from the TSM support to vanadium, likely due to the coexistence of Mo6+ and Ti(4+ )in the TSM solid solution, which has a unique structure. These results are expected to contribute to enhancing the abatement of dioxins at low temperatures.

An efficient and operationally simple protocol has been demonstrated for the synthesis of 1,3,5,7-tetrasubstituted pyrimido[4,5-d]pyrimidines via TBHP-mediated direct oxidative coupling of N-uracil amidines and methylarenes under metal-free conditions. Due to the inherent stability of methylarenes compared to aldehydes, the presented synthetic protocol is adaptable to a broad substrate scope, is operationally simple, has no need for stringent protection in the whole preparation process, and has the potential to prepare valuable products that are currently inaccessible or challenging to prepare using conventional methods. It is a significantly important complement to the conventional synthetic methods. The reaction possesses an efficient tandem oxidation-imination-cyclization process.

Herein we disclosed the utilization of copper loaded zinc oxide (ZnO-Cu) for its stimuli (O-2/light) responsive switchable performance between its reduced (S-1) and oxidized (S-2) state for two antagonistic reactions, namely oxidation of alkyl arenes/heteroarenes to aldehydes/ketones and reduction of nitro arenes/heteroarenes corresponding amines. The two states of the catalyst showed its switchable performance as highly active and poorly active catalyst for oxidation and reduction, and both reactions could be turned off and on by changing the stimuli (light and O-2/N-2). The switching efficiency between the states and their relative reactivity were found to be consistent under variety of reaction conditions and remain unaltered irrespective of oxidation-reduction (or vice versa) sequence and substrates used in the reaction. The photo catalysts (S-1 and S-2) demonstrated good catalytic activity, multiple reusability, broad substrate scope, and reasonable functional group tolerance for both the reactions and probed its quality performance in a large-scale setup. The system was used in an assisted tandem catalysis setup for the synthesis of benzyl amines utilizing both oxidation and reduction reaction by stimuli responsive switching between the states of the catalyst.

Providing new methods for the selective functionalization of small molecules is highly desirable, because installing molecular diversity in a desired position, for example, allows one to modulate bioactive molecules. This work reports a method for the selective functionalization of anilines using hexafluoroisopropanol (HFIP) as a solvent to promote an acid-catalyzed hydroarylation of olefins. Mechanistic experiments revealed that HFIP both protonates the alkene and selectively enables anilines toward the electrophilic aromatic substitution. This powerful strategy has been applied to the functionalization of the anti-inflammatory mefenamic acid with chemocontrol and regiocontrol.

An operationally simple nickel-catalyzed hydroarylation reaction for alkynes is described. This three component coupling reaction utilizes commercially available alkynes and aryl bromides, along with water and Zn. An air-stable and easily synthesized Ni(II) precatalyst is the only entity used in the reaction that is not commercially available. This reductive cross-coupling reaction displays a fairly unusual anti selectivity when aryl bromides with ortho substituents are used. In addition to optimization data and a preliminary substrate scope, complementary experiments including deuterium labeling studies are used to provide a tentative catalytic mechanism. We believe this report should inspire and inform other Ni-catalyzed carbofunctionalization reactions.

A new synthetic method toward direct C-N bond formation through saturated C-H amination of benzylic hydrocarbons and inactive aliphatic alkanes with primary aromatic amines under an inexpensive catalyst/oxidant (Cu/DTBP) system has been developed. Both aminopyridines and anilines could react smoothly with primary and secondary benzylic C-H substrates or cyclohexane to form the corresponding aromatic secondary amines in moderate to good yields. This protocol has the advantages of wide functional group tolerance and use of readily available raw materials.

Subunit-selective proteasome inhibitors are valuable tools to assess the biological and medicinal relevance of individual proteasome active sites. Whereas the inhibitors for the beta 1c, beta 1i, beta 5c, and beta 5i subunits exploit the differences in the substrate-binding channels identified by X-ray crystallography, compounds selectively targeting beta 2c or beta 2i could not yet be rationally designed because of the high structural similarity of these two subunits. Here, we report the development, chemical synthesis, and biological screening of a compound library that led to the identification of the beta 2c- and beta 2i-selective-compounds LU-002c (4; IC50 beta 2c: 8 nM, IC50 beta 2i/beta 2c: 40-fold) and LU-002i (5; IC50 beta 2i: 220 nM, IC50 beta 2c/beta 2i: 45-fold), respectively. Co-crystal structures with beta 2 humanized yeast proteasomes visualize protein-ligand interactions crucial for subunit specificity. Altogether, organic syntheses, activity-based protein profiling, yeast mutagenesis, and structural biology allowed us to decipher significant differences of beta 2 substrate-binding channels and to complete the set of subunit-selective proteasome inhibitors.

Environmental transformation products of pesticides (ETPPs) have a great deal of ecological impact owing to their ability to cause toxicity to the aquatic organisms, which can then be translated to the humans. The limited experimental data on biochemical and toxic effects of ETPPs, the high test costs together with regulatory limitations and the international push to reduce animal testing encourage greater dependence on predictive in silico techniques like quantitative structure-activity relationship (QSAR) models. The aim of the present work was to explore the key structural features, which regulate the toxicity towards fishes, for 85 ETPPs using a partial least squares (PLS) regression based chemometric model developed according to Organisation for Economic Co-operation and Development (OECD) guidelines. The model was extensively validated using both internal and external validation metrics, and the results so obtained justify the reliability and usefulness of the developed model (Q(2) = 0.648, R-pred(2) or Q(F1)(2) = 0.734 and Q(F2)(2) = 0.733). From the developed model, we can conclude that lipophilicity, polarity, presence of branching and the functional form of 0-atom in the transformed structures of pesticides are the important features that are to be considered during ecotoxicity assessment of ETPPs. The information obtained from the descriptors of the developed model could be utilized in the future for assessing ETPPs with the benefit of providing an early warning of their potentially detrimental effect on fishes for regulatory purposes.
