The present study reports on the degradation of chlorinated benzoic acids (CBAs), commonly present in the environment as pollutants, by a hydrodehalogenation reaction utilizing the Raney Al-Ni alloy (50:50 wt% Al:Ni). The hydrodehalogenation reaction using the Raney Al-Ni alloy has already been proven as an efficient tool for fast and efficient degradation of halogenated persistent organic pollutants (POPs). Herein, the nano-structured Raney Al-Ni alloy was prepared by an alternative mechano-thermal approach starting from pure elements in a form of powders. The prepared alloy was characterized by X-ray diffractometry, scanning electron microscopy, particle size distribution, and active surface area analyses. The properties of the material were compared with a commercial sample of the same alloy prepared by the atomization process. The activity of the synthesized alloy was evaluated as removal efficiency and a rate of dehalogenation of three different CBAs – 2-chlorobenzoic acid, 2,6-dichlorobenzoic acid, and 2,3,6-trichlorobenzoic acid (trysben); used in the past as an herbicide. Dehalogenation of all three tested CBAs yielded benzoic acid as the only product and followed the first-order reaction kinetics. Compared to the commercially available alloy, enhanced kinetics of CBAs removal was achieved, owing to the solid-state properties of the mechano-thermally prepared alloy.

SRC is a major regulator of many signaling pathways and contributes to cancer development. However, development of a selective SRC inhibitor has been challenging, and FDA-approved SRC inhibitors, dasatinib and bosutinib, are multitargeted kinase inhibitors. Here, we describe our efforts to develop a selective SRC covalent inhibitor by targeting cysteine 277 on the P-loop of SRC. Using a promiscuous covalent kinase inhibitor (CKI) SM1-71 as a starting point, we developed covalent inhibitor 15a, which discriminates SRC from other covalent targets of SM1-71 including TAK1 and FGFR1. As an irreversible covalent inhibitor, compound 15a exhibited sustained inhibition of SRC signaling both in vitro and in vivo. Moreover, 15a exhibited potent antiproliferative effects in nonsmall cell lung cancer cell lines harboring SRC activation, thus providing evidence that this approach may be promising for further drug development efforts.

Densities (rho), ultrasonic speeds (u) and dynamic viscosity (eta) of the binary 2-chlorotoluene or 4-chlorotoluene or 1,3-dichlorobenzene (1) + n-hexane (2) mixtures were measured at (T = 298.15-318.15) K and atmospheric pressure. Using these measurements, excess volume (V-m(E)), excess ultrasonic speed (u(E)) and deviation in dynamic viscosity (Delta eta) were calculated. Excess isentropic compressibility (kappa(E)(s)) was calculated using rho and u values. The VmE values have been interpreted by Prigogine-Flory-Patterson (PFP) theory. The VmEobtained using PFP theory agree well with experimental values. The u values were also calculated using various empirical relations. The available volume (V-a) and intermolecular free length (L-f) were calculated using experimental u values employing Jacobson free length theory. The Delta eta values were coupled with V-m(E) values to understand intermolecular interaction. Further, Bloomfield and Dewan model was applied to calculate eta and Delta eta values of the studied binary mixture and calculated values agree well with experimental values. (C) 2020 Elsevier B.V. Information on mixture adsorption equilibrium is vital in developing adsorption-based separation processes. Because measuring mixture adsorption is more difficult than measuring single-component adsorption, far more data of the latter kind are available. Previous efforts to compile experimental mixture adsorption data for gases have given data sets with at most a few dozen examples. Here, we report the results of systematic literature meta-analysis that produced a data set of more than 900 gas mixture adsorption experiments. This collection includes data from 125 different binary mixtures including 60 different molecular species and information from 333 different adsorbents. We refer to this data set as the Binary adsorption ISOtherm ExperimeNtal 2020 (BISON-20) Database. Because the BISON-20 data set enormously expands the number and variety of experimental results for binary gas adsorption that are readily available, it will be a useful resource for future efforts in developing new materials or processes for gas separations. As initial applications of the BISON-20 data set, we show how identifying replicate measurements can be used to assess the reliability of binary adsorption data, how the accuracy of Ideal Adsorbed Solution Theory (IAST) can be systematically tested using experimental data, and how trends in selectivity for gas separations across many materials can be examined.

The energy released when tetradecyltrimethylammonium bromide (C(14)TAB) is combined with different derivatives of chlorobenzoates in an isothermal titration calorimeter (ITC) allows a direct evaluation of the spontaneity of the threading of the aromatic anions into the micellar palisade. The comparison between the enthalpimetric curves with the ones for viscosity of the solutions, hydrodynamic radii of the aggregates (dynamic light scattering experiments), and the fraction of aromatic anions incorporated (diffusion-ordered spectroscopy experiments) allows the establishment of the variations of enthalpy with formation, growth, and decrease of the wormlike micelles (WLMs). The formation of WLMs with C(14)TAB is very favored (very exothermic) for titrations of chlorobenzoate derivatives which present the chlorine atom in positions 3 or 4 of the aromatic ring. However, the aggregation is highly unfavorable if chlorine is at position 2 of chlorobenzoate. According to the results, the high potential of the ITC to determine critical concentrations and the energies associated with the aggregation of a cationic surfactant and aromatic anions for the formation of WLMs was demonstrated.

BackgroundA number of region-specific validated triage systems exist; however very little is known about their performance in resource limited settings. We compare the local triage tool and internationally validated tools among under-fives presenting to an urban emergency department in Tanzania.MethodologyProspective descriptive study of consecutive under-fives seen at Muhimbili National Hospital (MNH), ED between November 2017 to April 2018. Patients were triaged according to Local Triage System (LTS), and the information collected were used to assign acuities in the other triage scales: Canadian Triage and Acuity Scale (CTAS), Australasian Triage Scale (ATS), Manchester Triage Scale (MTS) and South African Triage Scale (SATS). Patients were then followed up to determine disposition and 24h outcome. Sensitivity, specificity, positive and negative predictive values for admission and mortality were then calculated.ResultsA total of 384 paediatric patients were enrolled, their median age was 17months (IQR 7-36months). Using LTS, 67(17.4%) patients were triaged in level one, 291(75.8%) level 2 and 26 (6.8%) in level 3 categories. Overall admission rate was 59.6% and at 24h there were five deaths (1.3%). Using Level 1 in LTS, and Levels 1 and 2 in other systems, sensitivity and specificity for admission for all triage scales ranged between 27.1-28.4% and 95.4-98% respectively, (PPV 90.3-95.3%, NPV 47.1-47.4%). Sensitivity for mortality was 80% for LTS, and 100% for the other scales, while specificity was low, yielding a PPV for all scales between 6.9 and 8%.ConclusionAll triage scales showed poor ability to predict need for admission, however all triage scales except LTS predicted mortality. The test characteristics for the other scales were similar. Future studies should focus on determining the reliability and validity of each of these triage tools in our setting.

V2O5/TiO2-SiO2-MoO3 (TSM) catalysts exhibit enhanced redox capabilities, but the role of each species in the mixed oxide support is not well understood. Herein, the physicochemical characteristics of molybdenum oxide in TSM were investigated using X-ray diffraction, X-ray photoelectron spectroscopy, and X-ray absorption fine structure spectroscopy, and the performance of V/TSM catalysts for the oxidative decomposition of o-chlorotoluene was examined. The V/TSM catalysts exhibited superior performances in this decomposition reaction at low temperatures. The molybdenum species were highly dispersed as Mo6+, replacing Ti4+ in the anatase TiO2 framework and forming a solid solution. The redox capability of the vanadium species in the catalyst was enhanced by electron transfer from the TSM support to vanadium, likely due to the coexistence of Mo6+ and Ti(4+ )in the TSM solid solution, which has a unique structure. These results are expected to contribute to enhancing the abatement of dioxins at low temperatures.

To develop novel therapeutic agents with anticancer activities, two series of novel 2,4-bismorpholinyl-thieno [3,2-d]pyrimidine and 2-morpholinothieno[3,2-d]pyrimidinone derivatives were designed, synthesized and evaluated for their biological activities. Among them, compound A(12) showed the most potent antitumor activities against HCT116, PC-3, MCF-7, A549 and MDA-MB-231 cell lines with IC50 values of 3.24 mu M, 14.37 mu M, 7.39 mu M, 7.10 mu M, and 16.85 mu M, respectively. Further explorations in bioactivity were conducted to clarify the anticancer mechanism of compound A(12). The results showed that compound A(12) obviously inhibited the proliferation of A549 cell lines and decreased mitochondrial membrane potential, which led to the apoptosis of cancer cells and suppressed the migration of tumor cells.

The A-type Aurora kinase is upregulated in many human cancers, and it stabilizes MYC-family oncoproteins, which have long been considered an undruggable target. Here, we describe the design and synthesis of a series of pyrimidine-based derivatives able to inhibit Aurora A kinase activity and reduce levels of cMYC and MYCN. Through structure-based drug design of a small molecule that induces the DFG-out conformation of Aurora A kinase, lead compound 13 was identified, which potently (IC50 < 200 nM) inhibited the proliferation of high-MYC expressing small-cell lung cancer (SCLC) cell lines. Pharmacokinetic optimization of 13 by prodrug strategies resulted in orally bioavailable 25, which demonstrated an 8-fold higher oral AUC (F = 62.3%). Pharmacodynamic studies of 25 showed it to effectively reduce cMYC protein levels, leading to >80% tumor regression of NCI-H446 SCLC xenograft tumors in mice. These results support the potential of 25 for the treatment of MYC-amplified cancers including SCLC.
