Synthesis of 50 analogues of the natural insecticide synergists, dillapiol and sesamol, is reported. These were evaluated as potential insecticide synergists based on their inhibition of human CYP3A4. The most potent inhibitors have a relatively large hydrophobic substituent at either position 5 or 6 of these molecules. For example, 5-(benzyloxy)-6-(3-phenylsulfonyl)propyl)benzo[d] [1,3]dioxole (18) and the diphenyl acetate of (6,7-dimethoxybenzo[d] [1,3]dioxol-5-yl)propan-1-ol (5n) show inhibitory concentrations for 50% activity IC50 values of 0.086 and 0.2 mu M, respectively. These compounds are 106 and 46 times more potent than dillapiol whose IC50 for the inhibition of CYP3A4 is 9.2 mu M. The ortho-chloro analogue (8f), whose activity is 86 times the activity of dillapiol, is the most potent of the fourteen 5-(benzyloxy-6-(2-propenyl)benzo[d][1,3]dioxoles prepared for this study.

Objective A growing body of evidence suggests that testing for influenza virus alone is more appropriate than multiplex respiratory viral panel (RVP) testing for general populations of patients with respiratory tract infections. We aimed to decrease the proportion of RVPs out of total respiratory viral testing ordered during influenza season. Methods We implemented two consecutive interventions: reflex testing for RVPs only after a negative influenza test, and noninterruptive clinical decision support (CDS) including modifications of the computerized physician order entry search behavior and cost display. We conducted an interrupted time series of RVPs and influenza polymerase chain reaction tests pre- and postintervention, and performed a mixed-effects logistic regression analysis with a primary outcome of proportion of RVPs out of total respiratory viral tests. The primary predictor was the intervention period, and covariates included the provider, clinical setting, associated diagnoses, and influenza incidence. Results From March 2013 to April 2019, there were 24,294 RVPs and 26,012 influenza tests ( n = 50,306). Odds of ordering an RVP decreased during the reflex testing period (odds ratio: 0.432, 95% confidence interval: 0.397-0.469), and decreased more dramatically during the noninterruptive CDS period (odds ratio: 0.291, 95% confidence interval: 0.259-0.327). Discussion The odds of ordering an RVP were 71% less with the noninterruptive CDS intervention, which projected 4,773 fewer RVPs compared with baseline. Assuming a cost equal to Medicare reimbursement rates for RVPs and influenza tests, this would generate an estimated averted cost of $1,259,474 per year. Conclusion Noninterruptive CDS interventions are effective in reducing unnecessary and expensive testing, and avoid typical pitfalls such as alert fatigue.

A series of new triazole ligands has been synthesized via copper-catalyzed cycloaddition reaction of readily available azides and alkynes. The synthesized compounds were characterized by FTIR, H-1 and C-13 NMR, and high-resolution mass spectra. The ligands provided excellent yields (up to 92%) in the palladium-catalyzed Suzuki-Miyaura cross coupling of unactivated aryl chlorides with phenylboronic acid. 1-Benzyl-4-(2,6-dimethoxyphenyl)-lH-1,2,3-triazole was found to be the most effective ligand due to the presence of electron-donating 2,6-dimethoxyphenyl substituent, which made it possible to develop a highly active ligand-catalyst system for the Suzuki reaction of aryl chlorides.

Structural modifications of nicotinamide, a form of vitamin B3, gave rise to a series of compounds (8aa-8ce) that exhibit activities as type I positive allosteric modulators (PAMs) of human alpha 7 nAChR expressed in Xenopus oocytes in two-electrode voltage clamp assay. The compound 8ai was a potent and efficacious PAM with an EC50 = 3.34 +/- 1.13 mu M and the maximum activation effect of alpha 7 current over 1474 +/- 246% in the presence of acetylcholine (100 mu M). It is highly specific to alpha 7 nAChR over other subtypes of nAChR and 5-HT3A receptors. The structure-activity relationship analysis identified a key skeleton of nicotinamide nucleus critical for biological activity. Taken together, the 8ai as a type I PAM of alpha 7 nAChR may be beneficial for improvement of cognitive deficit.

Tracing the herbicide bixlozone and its metabolites in food is necessary to assess their risks to human health. In the study, a rapid and effective analytical method using the quick, easy, cheap, effective, rugged, and safe method for the simultaneous determination of bixlozone and its metabolites (2,4-dichlorobenzoic acid, 3-hydroxy-propanamide-bixlozone, and 5 ‘-hydroxy-bixlozone) in plant and animal samples (tomato, cucumber, apple, wheat flour, meat, milk, and egg) was developed based on ultra high performance liquid chromatography-tandem mass spectrometry. The method was validated based on the linearity (R-2 > 0.99), sensitivity (limit of quantification = 0.01 mg/kg), recovery (70.2-115.1%), and precision (intraday 1.2-17.6%, interday 0.3-16.0%). Detection was achieved within 6.0 min. The method is reliable for the determination of four target compounds in all seven matrices. The satisfactory validation criteria and successful application show that the proposed methodology is suitable for the detection of four target compounds in real matrices.

Three kinds of novel hydroxyapatite@terbium complex core-shell composites were synthesized with 2-chlorobenzoic acid, 4-chlorobenzoic acid, and 2,4-dichlorobenzoic acid as the ligand, respectively. The XRD, UV/vis absorption and FT-IR results show that terbium ions are coordinated with ligands and the corresponding complexes are successfully included into composites. SEM and EDS results reveal that Tb complexes are fully and homogenously coated on the surface of hydroxyapatite (HAP) microspheres consisting of a large number of nanosized crystals. The strongest luminescence properties are obtained when 2,4-dichlorobenzoic acid is used as ligand. The study on fluorescence lifetime of composites shows that the lifetime is inversely related to emission intensity. TGA-DSC results indicate that the core-shell composites have an excellent thermal stability. Therefore, the current core-shell structure provides a cost-effective route to manufacture of biocompatible and heat-resistant highly luminescent materials. (C) 2020 Chinese Society of Rare Earths. An efficient and operationally simple protocol has been demonstrated for the synthesis of 1,3,5,7-tetrasubstituted pyrimido[4,5-d]pyrimidines via TBHP-mediated direct oxidative coupling of N-uracil amidines and methylarenes under metal-free conditions. A series of core-shell Co-MOF-74@Mn-MOF-74 samples with different shell thicknesses were prepared by the seed growth method, which were characterized by powder X-ray diffraction (PXRD), thermogravimetric analysis (TGA), inductively coupled plasma mass spectrometry (ICP-MS), X-ray photoelectron spectroscopy, nitrogen adsorption-desorption and scanning electron microscopy (SEM). The shell thickness of these MOF samples were successfully controlled by adjusting the mass ratio between MnCl2 center dot 4H(2)O and Co-MOF-74. On this basis, these MOFs were applied to the catalytic oxidation of toluene. The results showed that with the growth of Mn-MOF-74 in the outer layer of Co-MOF-74, the oxidative selectivity of the substrate to benzaldehyde was greatly improved for the synergy between the core layer and shell layer. The conversion of toluene was 22.4%, and the selectivity of benzaldehyde was 98.1%. In addition, the catalyst can transform various substituted toluene into the corresponding aldehydes in highly selectivity and still keep good stability after four catalytic cycles. The selectivity of the corresponding aldehyde is generally above 80%.

We report the development of a facile protocol for the deoxygenative hydroboration of aliphatic and aryl carboxylic acids to afford corresponding primary alcohols under solvent-free and catalyst-free conditions. The reaction proceeds under ambient temperature exhibits good tolerance towards various functional groups and generates quantitative yields. The plausible mechanism involves the formation of Lewis acid-base adducts as well as the liberation of hydrogen gas.
