Copper (II) immobilized on L-arginine-beta-cyclodextrin-functionalized magnetite nanoparticles (nano-Fe3O4@L-arginine-CD-Cu(II)) were successfully synthesized and fully characterized using FT-IR, XRD, SEM, EDX, ICP, TGA and VSM techniques. The catalytic activity of these magnetically retrievable nanoparticles was evaluated in the direct oxidation of primary alcohols and benzyl halides to acids in neat conditions that was observed to proceed well and products were obtained in good yields. 20(R)-25-hydroxyprotopanaxadiol (25-OH-PPD) is a natural compound showing a variety of anti-tumor effects. In an attempt to search for a new anti-cancer compound with higher antitumor activities, we designed and synthesized a series of 25-OH-PPD derivatives. Cytotoxicity assay of these derivatives towards MCF-7, A549, U87, HO-8901, Hela cancer cell lines and normal IOSE144 cell lines were tested by MTF assay. Results showed that compared with compound 25-OH-PPD, Compounds 4, 5, 6, 10, 11 showed strong anticancer activity, and all compounds showed low toxicity or no toxicity for normal cells. In particular, compound 6 exhibited the best anti-tumor activity in all cancer cell lines (MCF-7, A549, U87, HO-8901, and Hela) with IC50 values of 5.04 mu M, 1.36 mu M, 3.24 mu M, 3.47 mu M, 4.57 mu M, respectively. Among the five cell lines, all the compounds showed strong inhibition on A549 cells. Further studies showed that Compound 6 significantly inhibited the proliferation of A549 cells by inducing apoptosis. Our results indicate that Compound 6 is a potential anticancer inhibitor and provides a theoretical basis for further research.

2-Pyridyl ketones widely appear in bioactive molecules, natural products, and are employed as precursors of chiral 2-pyridine alky/aryl alcohols or 2-aminoalkyl pyridine ligands for asymmetric catalysis. Herein, a practical method for the rapid synthesis of 2-pyridyl ketone library in continuous flow is reported, in which the 2-lithiopyridine formed by Br/Li exchange reacts with commercially available esters to obtain 2-pyridyl ketones in a good yield at short reaction time. This protocol functions broadly on a variety of esters and has been applied to the synthesis of TGF-beta type 1 receptor inhibitor LY580276 intermediate in an environmentally friendly method. It is rapid, reliable, and cost-efficient to afford diverse kinds of 2-pyridyl ketones in the compound library.

The 1,3-benzodioxol moiety present in safrole, apiole, and myristicin essential oils and benzodioxol derivatives have shown a wide range of biological activities including antiepileptic, analgesic, antituberculosis, and antimicrobial potentials. Here, we have tested the antibacterial and antioxidant activities of a series of benzodioxol derivatives. Twelve compounds of aryl acetate and acetic acid benzodioxol were evaluated against different types of bacterial strains, including Staphylococcus aureus, Escherichia coli, Enterococcus faecalis, and Pseudomonas aeruginosa using the broth dilution method, and the most potent compound was 3e, which exhibited the bacterial growth of with MICs of 125 (S. aureus), 250 (E. coli), 220 (E. faecalis), and 100 mu g/mL (P. aeruginosa). Our positive control, cinoxacin, had MICs of 250 (S. aureus), 250 (E. coli), 250 (E. faecalis), and 500 mu g/mL (P. aeruginosa). Antioxidant activity was evaluated for the synthesized compounds utilizing the DPPH assay. The 3a compound was the most active with an IC50 value of 21.44 mu g/mL, while the IC50 values of compounds 3b, 3e, and 3f were 96.07, 58.45, and 72.17 mu g/mL, respectively. In contrast, all compounds with the acetic acid functional group had weaker activity, with an IC50 range of 193.52-289.78 mu g/mL compared with the potent antioxidant agent Trolox (IC50 = 1.93 mu g/mL). In the present paper, new benzodioxol-based aryl acetate and acetic acid derivatives were evaluated for their antibacterial and antioxidant activities. The outcomes revealed that the antibacterial and antioxidant properties of some of the synthesized benzodioxol aryl acetate and acetic acid derivatives can be considered as valuable materials for the pharmaceutical industry. Thus, these molecules should be further evaluated in vivo as lead compounds for the discovery of new drug candidates.

The insect enzyme GH20 beta-N-acetyl-D-hexosaminidase OfHex1 represents an important chitinolytic enzyme found in the agricultural pest Ostrinia furnacalis (Guenee) and inhibition of this enzyme has been considered a promising strategy for the development of eco-friendly pesticides. In this article, based on the structure of the catalytic domains of OfHex1, a series of novel glycosyl triazoles were designed and synthesized via Cu-catalyzed azide-alkyne [3+ 2] cycloaddition reaction. To investigate the potency and selectivity of these glycosyl triazoles, the inhibition activities towards OfHex1 and HsHexB (human beta-N-acetylhexosaminidase B) were studied. Particularly compound 17c (OfHex1, K-i= 28.68 mu M; HsHexB, K-i > 100 mu M) exhibited a suitable activity and selectivity against OfHex1. Furthermore, the possible inhibitory mechanisms of 17c with OfHex1 were studied using molecular docking and MD simulations. The structure-activity relationship results as well as the formed binding patterns may provide promising insights into the further development of novel OfHex1 inhibitors.

Cocrystallization of the widely available 4-dimethylaminopyridine, with a series of carboxylic acids gave nine molecular adducts 4-dimethylaminopyridinium trifluoroacetate (1) , 4-dimethylaminopyridinium glycolate glycolic acid (2) , 4-dimethylaminopyridinium 2-furoate dihydrate (3) , tri(4-dimethylaminopyridinium) tri(o-chlorobenzoate) tetrahydrate (4) , 4-dimethylaminopyridinium 4-bromobenzoate 4-bromobenzoic acid (5) , 4-dimethylaminopyridinium 4-formylbenzoate 4-formylbenzoic acid (6) , 4-dimethylaminopyridinium 2,4-dichlorobenzoate (7) , 4-dimethylaminopyridinium 2-aminobenzoate 2-aminobenzoic acid (8) and bis(4-dimethylaminopyridinium) 5-nitroisophthalate 5-nitroisophthalic acid methanol solvate (9) . The nine adducts have been characterised by X-ray diffraction, IR and elemental analysis, the melting points were also reported. Their structural and supramolecular aspects are fully analyzed. The result reveals that among the nine investigated crystals only the ring Ns in the 4-dimethylaminopyridine moieties are protonated when the carboxylic acids are deprotonated. The crystal packing is interpreted by the strong charge-assisted N-H center dot center dot center dot O hydrogen bond between the NH+ and the deprotonated acidic groups. Except the N-H center dot center dot center dot O hydrogen bonds, the charge assisted or neutral O-H center dot center dot center dot O hydrogen bonds were also found at 2-6, 8 and 9 . The COOH center dot center dot center dot COO- synthon has been at 2, 5-6 and 8-9. Further analysis of the crystal packing indicated that a different family of additional CH-O/CH2-O/CH3-O, F-F, Br-Br, CH-Cl, CH3-Cl, CH-pi/CH3-pi, C-H center dot center dot center dot C-pi, C-pi, Cl-O, O-C, O-pi and pi-pi contacts contribute to the stabilization and expansion of the total high-dimensional (2D-3D) framework structures.
