Type I Interferon (IFN) signaling plays an important role in the immune defense system against virus infection and in the innate immune response, thus IFNs are widely used as anti-viral agents and treatment for immune disorder or cancer. However, there is a growing demand for novel small-molecule IFN inducer due to tolerance, toxicity, or short duration of action following direct administration of IFNs. In this study, we assessed arylpiperazine (ARP) as a new core skeleton of IFN inducer. To investigate structure-activity relationship, we designed and synthesized a series of ARP analogues and evaluated the ability to stimulate IFN response in THP-1 human monocyte cells. Compound 5i was identified as a potent type I IFN inducer as it significantly increased cytokine secretion and increased expression of various IFN-stimulating genes which are representative biomarkers of type I IFN pathway. Our results suggested a beneficial therapeutic potential of 5i as an anti-viral agent.

Isocyanide multicomponent reactions play a prominent role in drug discovery. This chemistry has hardly been investigated for compatibility with DNA-encoded combinatorial synthesis. The Ugi, Ugi-azide, and Groebke-Blackburn-Bienayme reactions are well-tolerated by DNA on the solid phase and show a broad scope. However, an oxadiazole-forming variant of the Ugi reaction caused DNA depurination, requiring a more stable hexathymidine DNA for encoded library synthesis. Cheminformatic analysis revealed that isocyanide multicomponent-reaction-based encoded libraries cover a diverse chemical space.

A visible-light photoredox catalyzed radical cascade cyclization of simple ethers with cyanamides is developed at room temperature. This strategy involves sequential inert C-sp3-H/C-sp2-H functionalizations through intermolecular addition reaction of oxyalkyl radicals to N-cyano groups followed by radical cyclization of iminyl radicals in situ generated with C-2 aryl rings. This method allows for efficient synthesis of tetracyclic benzo[4,5]imidazo[1,2-c]quinazolines. Importantly, this is the first example of an intermolecular-intramolecular radical cascade cyclization reaction of cyanamides.

In the presence of [p-cymene)RuCl2](2), (E)-configured alkenyl bromides couple with aromatic carboxylates to form orthovinylbenzoic acids. This C-H vinylation proceeds in high yields without any activating phosphine ligands and has an excellent functional group tolerance. Starting from commonly available (E/Z)-mixtures of alkenyl bromides, (E)-configured vinyl arenes or dienes are formed exclusively. Mechanistic studies show that this selectivity is achieved because the (E)configured alkenyl bromides undergo a smooth coupling, whereas the (Z)-isomers are rapidly eliminated with the formation of alkynes.

Predictive models are useful tools for aqueous adsorption research; existing models such as multilinear regression (MLR), however, can only predict adsorption under specific equilibrium concentrations or for certain adsorption isotherm models. Also, few studies have discussed data processing beyond applying different modeling algorithms to improve the prediction accuracy. In this research, we employed a cosine similarity approach that focused on mining the available data before developing models; this approach can mine the most relevant data concerning the prediction target to build models and was found to considerably improve the prediction accuracy. We then built a machine-learning modeling process based on neural networks (NN), a group-selection data-splitting strategy for grouped adsorption data for adsorbent-adsorbate pairs under different equilibrium concentrations, and polyparameter linear free energy relationships (pp-LFERs) for aqueous adsorption of 165 organic compounds onto 50 biochars, 34 carbon nanotubes, 35 GACs, and 30 polymeric resins. The final NN-LFER models were successfully applied to various equilibrium concentrations regardless of the adsorption isotherm models and showed less prediction deviations than the published models with the root-mean-square errors 0.23-0.31 versus 0.23-0.97 log unit, and the predictions were improved by adding two key descriptors (BET surface area and pore volume) for the adsorbents. Finally, interpreting the NN-LFER models based on the Shapley values suggested that not considering equilibrium concentration and properties of the adsorbents in the existing MLR models is a possible reason for their higher prediction deviations.

A series of novel indole derivatives containing double 1,3,4-oxadiazole moiety was designed, synthesized and evaluated for their antibacterial activities in vitro. These compounds were fully characterized by H-1 NMR, C-13 NMR, and HRMS. Bioassay results indicated that most of title compounds exhibited excellent antibacterial activities against rice bacterial pathogen Xanthomonas oryzae (Xoo). For example, compounds 7d, 7h, 7i, 7j, 7k, 7l and 7m had the half-maximal effective concentration (EC50) values of 52.31, 54.12, 40.65, 38.80, 51.13, 52.75 and 50.66 mu g/mL, respectively, which was better than that of commercial product bismerthiazol (BMT) (85.18 mu g/mL). The experimental results proved that indole derivatives bearing double 1,3,4-oxadiazole unit are promising candidates for the development of new agricultural bactericides against pathogenic bacterium Xoo. Transition-metal-catalyzed activation of inert C-H bonds and subsequent C-C bond formation have emerged as powerful synthetic tools for the synthesis of elaborate cyclic molecules. In this report, we introduce an efficient synthetic method of 3,4-unsubstituted isocoumarins adopting an electron-deficient (CpRh)-Rh-E complex as the catalyst. The use of vinylene carbonate as a vinylene transfer reagent enables the direct construction of isocoumarins from readily available benzoic acids, without any external oxidants as well as bases. The reaction mechanism is evaluated by computational analysis to find an unprecedented rhodium shift event within the catalytic cycle.

Green, simple, and highly efficient various strategies for the straightforward reduction of carboxylic acids to corresponding alcohols have been developed by employing four different types of cost-effective hydrogen donors, namely sodium borohydride (NaBH4), ammonium formate (HCO2NH4), glycerol, and isopropanol (i-PrOH) in the presence of Fe3O4@APTMS@ZrCp2Clx ((x = 0, 1, 2)) as a robust and durable nanocatalyst under neat conditions. Also, the mentioned magnetically separable nanocatalyst was recovered and reused at least five times without significant loss in catalytic activity. In comparison with the known protocols for the direct reduction of carboxylic acids to corresponding alcohols, our current methods offer several benefits such as being straightforward (without isolation of aldehyde intermediates), mild reaction conditions, eco-friendly, short reaction times, good to excellent yields of the products, use of inexpensive hydrogen donors, recoverability, and reusability of the catalyst and so on.

A palladium-catalyzed cross-coupling reaction of aryl halides with 2-chlorobenzoic acids has been developed. The reaction forms C(sp(3)), C(sp(2))-palladacycles through C(sp(3))-H activation. The palladacycles react with 2-chlorobenzoic acids through two successive C-C cross-couplings, and two C-C bonds are formed with high chemoselectivity. The reaction provides an innovative method for the synthesis of 9,10-dihydrophenanthrenes.

Three kinds of novel hydroxyapatite@terbium complex core-shell composites were synthesized with 2-chlorobenzoic acid, 4-chlorobenzoic acid, and 2,4-dichlorobenzoic acid as the ligand, respectively. The XRD, UV/vis absorption and FT-IR results show that terbium ions are coordinated with ligands and the corresponding complexes are successfully included into composites. SEM and EDS results reveal that Tb complexes are fully and homogenously coated on the surface of hydroxyapatite (HAP) microspheres consisting of a large number of nanosized crystals. The strongest luminescence properties are obtained when 2,4-dichlorobenzoic acid is used as ligand. The study on fluorescence lifetime of composites shows that the lifetime is inversely related to emission intensity. TGA-DSC results indicate that the core-shell composites have an excellent thermal stability. Therefore, the current core-shell structure provides a cost-effective route to manufacture of biocompatible and heat-resistant highly luminescent materials. (C) 2020 Chinese Society of Rare Earths.
